Experience in autologous and allogeneic cord blood banking.

Transplantation and gene therapy offer the hope of treating thousands of individuals with various malignancies and genetic disorders. However, if this approach is to become a reality, there must be readily available sources of suitable HLA-A typed stem cells. Cord blood has been demonstrated to be a suitable source of hematopoietic stem cells that can be used in place of bone marrow stem cells for transplant. However, to implement the widespread clinical use of cord blood, it is necessary to establish both autologous and allogeneic cord blood banks from which stem cell specimens can be obtaine

Porphyrin-Peptide Bioconjugates: Photophysical Properties and Targeted Singlet Oxygen-Mediated Photooxidation of Amyloid-β.

The aberrant self-assembly of amyloid β (Aβ) peptides into toxic oligomers is a central event in Alzheimer's disease, motivating the development of molecular systems capable of modulating or degrading these species. Porphyrins and porphyrin-peptide bioconjugates have emerged as promising photoactive platforms due to their known ability to generate the highly oxidant singlet oxygen (1O2) upon light excitation. Still, the mechanisms underlying their oxidative effects on Aβ remain unclear. Here, we investigate the photophysical properties and the 1O2-mediated oxidation capability of two porphyrin

Hierarchically Molecular Programming of Antiparallel H‑Aggregates for NIR Type I Photochemistry.

Developing near-infrared (NIR) photosensitizers with effective type I reactive oxygen species (ROS) generation remains challenging. H‑aggregates promote triplet formation and charge transfer, but often suffer from substantial absorption blueshifts and excessive thermal deactivation that compromise photosensitization efficacy. Herein, a hierarchically molecular programming strategy is proposed to construct minimal-blueshift H-aggregates as robust NIR type I photosensitizers. Primary engineering via π-bridge extension and acceptor strengthening within a twisted-planar donor-π-acceptor scaffold e

A Tumor-Targeting Perovskite Quantum Dot with High Luminescence and Stability for Radiotherapy Sensitization through X-ray-Activated Photoelectrocatalysis.

Radiotherapy (RT) remains a cornerstone in lung cancer management, yet its efficacy is often compromised by radiation resistance and off-target toxicity. Metal halide perovskites exhibit high potential for biomedical applications owing to their exceptional fluorescence imaging and photoelectric conversion capabilities, offering distinct radiosensitization advantages via high-Z elements and radiation-excited electron‒hole pairs. However, their poor physiological stability and limited biocompatibility have hindered biomedical translation. Herein, we construct an optimized multifunctional perovsk

Redundant mispair excision mechanisms in eukaryotic DNA mismatch repair.

DNA mismatch repair (MMR) corrects DNA replication errors, some forms of chemical damage to DNA bases, and acts in processes such as heteroduplex rejection, triplet repeat expansion, antibody maturation and cytotoxicity of alkylating agents. During MMR, the newly synthesized DNA strand containing the error must be removed and resynthesized to prevent mutations. In bacteria, this strand is displaced by the UvrD DNA helicase. In eukaryotes, this strand is excised by redundant pathways. In the first excision pathway identified, Saccharomyces cerevisiae Msh2-Msh6 (or Msh2-Msh3) and/or Mlh1-Pms1 (h

Learning ordinal representation across MRI sequences for liver fibrosis staging.

BACKGROUND: Accurate liver fibrosis staging (LFS) is important for the diagnosis and treatment planning of patients with liver diseases. Noncontrast MRI is suitable for early screening and long-term monitoring in clinical practice. However, due to the lack of contrast-enhanced details, and given the continuous progression of liver fibrosis and the subtle differences between stages, the use of noncontrast MRI for LFS requires further exploration. PURPOSE: Here, we develop and evaluate a fine-grained deep learning pipeline using noncontrast MRI for precise and improved diagnosis. METHODS: To r

Rational Design of an AIE-Active BODIPY-Quinolone-Imidazole Conjugate for Effective Photodynamic Therapy of Breast Cancer Cells.

Photodynamic therapy (PDT) has found itself as one of the leading cancer therapeutic modalities, owing to its minimally invasive nature. The outcomes of PDT can be improved with the development of efficient photosensitizers (PSs). The present work describes the development of three photosensitizers (C1, C2, and C3) by reigning in the photophysical attributes of heavy atom-free boron dipyrromethenes (BODIPYs) based on an imidazole-quinolone-BODIPY conjugate connected through 1,2,3-triazole linkers. The intricate molecular construct of these PSs has endowed two PSs (C2, C3) with an aggregation-i

Ligand-field symmetry and magneto-optical correlations in a luminescent Dy(III) single-molecule magnet.

The establishment of ligand field (LF) symmetry around lanthanoid (Ln(III)) centres is of paramount importance to understand factors that govern magnetization relaxation in single-molecule magnets (SMMs). We investigated a luminescent mononuclear Dy(III) compound [Dy(BA)4] (pip) (1), where pip is a piperidinium cation and BA is a benzoylacetonate ligand. The stoichiometric compound exhibits zero-field SMM characteristics; quantum tunnelling of magnetization (QTM) dominates magnetization relaxation. In the diluted version of the compound (1@Y), the QTM is mitigated, and Raman and Orbach process

Achieving Blue-Shifted and Optimal Room Temperature Phosphorescence Emission via Hydrostatic Pressure Effect: A Theoretical Perspective.

Hydrostatic pressure provides a powerful external stimulus to modulate the excited-state properties of organic room-temperature phosphorescent (RTP) materials, yet the microscopic origin of pressure-induced spectral evolution remains insufficiently understood. Herein, we present a comprehensive theoretical investigation into the origin of blue-shifted RTP emission in IH-MPT under hydrostatic pressure. The calculations reveal that increasing pressure induces a pronounced blue shift in the phosphorescence spectrum, which originates from a continuous upward shift of the lowest triplet excited sta

A Planarity-Hindrance Co-Balance Strategy to Develop Antiparallel H-Aggregates With Minimal Absorbance Blueshift for Type I Photodynamic Therapy.

H-aggregates offer intrinsic features for type I photodynamic therapy (PDT) by concurrently promoting triplet state formation and strengthening charge transfer ability. However, their exploitation remains limited by the inherently large absorption blueshift (usually >100 nm) arising from strong H-type excitonic coupling in conventional parallel-packed H-aggregates, forcing short-wavelength laser excitation with poor tissue penetration. Herein, this study reports a planarity-hindrance co-balance strategy to develop donor-π-acceptor-based antiparallel-packed H-aggregates with minimal absorption

An In Situ-Generated Near-Infrared Chemiluminescent Nanoprobe for High-Contrast Lymph Node Mapping and Tumor-Guided Surgery.

Chemiluminescent nanoprobes have attracted increasing attention in bioimaging owing to their excitation-free emission and high signal-to-background ratio. However, most reported self-sustaining chemiluminescent nanosystems rely on strong photosensitizers to generate large amounts of reactive oxygen species to initiate the emission process. Although this strategy may result in deep tissue penetration, the excessive ROS accumulation could raise potential phototoxicity concerns. In this work, we designed a bioimaging probe in which the ROS generation, ROS trapping, and near-infrared (760 nm) chem

De novo design of NIR-II thioxanthene dye and phosphate-driven charge transfer-coupled J-aggregates for high resolution tumor angiography and type I phototherapy against hypoxic tumors.

Regulating the photophysical properties of NIR-II dyes via atomic engineering to inhibit type II reactive oxygen species (ROS, 1O2) generation and only generate type I ROS (O2˙- and ˙OH), and modulating dye aggregation for fabricating charge transfer (CT)-coupled J-aggregates (J CT-aggregates) to amplify O2˙- and ˙OH yield remain major challenges with rare reports. Herein, we de novo designed a NIR-II thioxanthene dye SOH. The heavy atom effect of sulfur and the twisted conjugated skeleton strengthened spin-orbit coupling (SOC) by 9.3-fold and intersystem crossing (ISC) efficiency by 86.5-fold

MediTEDNet: Visual State Space Model for Thyroid Eye Disease Classification.

Thyroid eye disease (TED) is a prevalent autoimmune orbital disorder that can severely impair visual function and significantly diminish patients' quality of life. In recent years, several studies have attempted to automate TED diagnosis using optical coherence tomography (OCT) images. However, existing approaches primarily rely on convolutional neural networks (CNNs) combined with attention mechanisms and are mostly trained using traditional cross-entropy loss. Although Transformers excel at modeling long-range dependencies, their quadratic computational complexity when processing high-resolu

Supramolecular Predisposition Promotes Intramolecular Heavy-Atom Effects for Self-Sensitized Oxidation.

Supramolecular confinement is widely used to control molecular architecture, but its use to direct excited-state reaction pathways remains underexplored. This limitation is particularly evident for spin-forbidden processes such as intersystem crossing (ISC), which are difficult to regulate through supramolecular design. The heavy-atom effect, although central to promoting ISC, is typically regarded as an intrinsic substituent property rather than a geometry-dependent supramolecular parameter. Here we show that macrocycle-directed supramolecular predisposition can deliberately enforce intramole

Engineering Fast Reverse Intersystem Crossing and Radiative Decay in [1,2,4]Triazolo[1,5-a]pyridine-Based TADF Emitters via Donor Strength and Positional Control of Dihedral Angle and Reorganization Energy: A Density Functional Theory Study.

Achieving a balance between the radiative decay rate (kr) and the reverse intersystem-crossing (rISC) rate (krISC) persists as a fundamental hurdle in thermally activated delayed-fluorescence (TADF) emitters because the processes depend oppositely on the overlap of frontier molecular orbitals. We computationally designed and explored 30 [1,2,4]triazolo[1,5-a]pyridine (TP)-based donor-π-acceptor emitters divided into two molecular series, 2,6TP and 2,7TP, according to the site for substituting the donor-appended phenyl linker on the TP acceptor core. The relationships among the donor-acceptor d
