Dual effects of 2-deoxyglucose on synthesis of the glycoprotein hormone common alpha-subunit in butyrate-treated HeLa cells.

Sodium butyrate (Btr) (3 mM) causes a 10-fold increase in production of the glycoprotein hormone alpha-subunit in HeLa cells. The following report demonstrates that this response could be inhibited about 95% by 5 mM 2-deoxy-D-glucose (dGlc), whereas alpha-subunit production in uninduced cells was affected little or not at all. Addition of D-mannose restored the Btr induction of Hela-alpha in cultures that had been treated with dGlc. When the alpha-subunits secreted by cells cultured in Btr plus dGlc or in Btr alone were compared by gel filtration (Sephadex G-75) and lectin affinity (concanaval

Toxofactor associated with Toxoplasma gondii infection is toxic and teratogenic to mice.

A toxin associated with Toxoplasma gondii infection was obtained from the trophozoites and culture medium used to propagate the parasite in cell cultures. The toxin, named Toxofactor (TF), administered parenterally or nonparenterally in adult mice, produces transient symptoms of lethargy, ruffled fur, and body weight loss. Organ changes which accompanied the outward symptoms included hepatosplenomegaly and involuted thymus. TF activity was detected in extracts of the blood, peritoneal fluid, liver, and spleen of infected mice. Severe damage to embryonal and fetal development was induced when T

Collectins and collectin receptors in innate immunity.

This thesis is based on nine papers and a review on the collectins and collectin receptors in innate immunity. The collectins are a family of proteins in which the individual chains consist of a C-type lectin domain attached to a collagen domain via an alpha-coiled neck region. The chains are organized into a triple collagen helix and oligomerized through N-terminally located cysteines. The collectins have a dual function: one is to bind specifically to carbohydrate structures on the surface of a pathogen; the other is subsequently to recruit other cells and molecules to destroy the pathogen.

Optimized Expression and Purification of Outer Membrane Protein 25 (Omp25) of Brucella melitensis.

Our previous immunoproteomic studies on patients with acute brucellosis identified outer membrane protein 25 (Omp25) of Brucella melitensis as one of the immunogenic antigens, indicating its potential to be used as a diagnostic marker and a tool to differentiate acute and chronic human brucellosis. Expression and purification of Omp25 are technically challenging using conventional approaches owing to its membrane association. Existing expression and extraction protocols frequently lead to host cell toxicity, low protein yields, and poor reproducibility. To address these limitations, we develop

Design, Characterization, and Efficacy of ASEnd: A Novel Chimeric Endolysin for Targeting Methicillin-Resistant Staphylococcus aureus.

The emergence of antibiotic-resistant pathogens, particularly methicillin-resistant Staphylococcus aureus (MRSA), represents a major public health threat, with limited treatment options. Bacteriophage-derived endolysins and their engineered chimeric derivatives have attracted attention as alternative antibacterial agents due to their rapid bacteriolytic activity, target specificity, and low resistance propensity. ASEnd was constructed by fusing the enzymatically active LYZ2 domain from S. aureus phage φMR11 with the SH3b-type cell wall-binding domain from phage K via a flexible (G4S)3 linker.

A review of exosomes as nanomedicine for antimicrobial delivery and combating antimicrobial resistance.

Exosomes are nanoscale vesicles derived from cell membranes with crucial roles in physiological and disease-specific processes. EVs may offer biological compatibility and cell-interactive surfaces, but these properties are source-, process-, cargo-, and route-dependent and should not be assumed to be superior to liposomes or other synthetic carriers. These unique properties make exosomes highly attractive platforms for drug delivery, diagnostics, and therapeutic applications, as they combine natural stability with functional versatility in cellular communication and disease modulation. This re

Xiao Qing Long Tang ameliorates neutrophil extracellular trap-dendritic cells-T helper 17 cell axis in Neutrophilic Asthma.

BACKGROUND: Neutrophilic asthma (NA) is an allergic airway inflammation disease featuring heterogeneous neutrophil infiltration, which is driven by the interactions between dendritic cells (DCs) and T helper (Th) 17 cells. Neutrophils release neutrophil extracellular traps (Nets), which promote disease progression and glucocorticoid resistance. Therefore, targeting the interaction among Nets, DC and Th17 is a promising pathway for preventing organ damage. Traditional Chinese Medicine (TCM), especially Xiao-qing-long-tang (XQLT), has shown potential in managing eosinophilic asthma by modulating

Optimized production of full-length PCV2d virus-like particles in Escherichia coli: A cost-effective and high-yield approach for potential vaccine antigen development.

Porcine circovirus type 2 (PCV2) is a globally prevalent infectious pathogen affecting swine, with its capsid protein (Cap) being the sole structural protein critical for vaccine development. Prior research has demonstrated that PCV2 Cap proteins produced in Escherichia coli (E. coli) can form virus-like particles (VLPs) in vitro, and nuclear localization signal peptides (NLS) play a pivotal role in stabilizing PCV2 VLPs. Recently, PCV2d has emerged as an important strain within the PCV2 epidemic. In this study, we systematically optimized the PCV2d Cap protein and successfully produced intact
