Enhancing the translational potential of the ovine pulmonary adenocarcinoma model.

Ovine pulmonary adenocarcinoma (OPA) is a transmissible lung cancer of sheep caused by Jaagsiekte sheep retrovirus (JSRV), resulting in substantial animal welfare concerns and economic losses worldwide. We previously developed a bronchoscopic JSRV catheter instillation technique targeting the cardiac lung lobe that reliably reproduced the spectrum of OPA lesions observed in naturally occurring disease. Beyond its value for investigating JSRV pathogenesis, this experimental system provides a large animal platform for studying human lung cancer. Here, we sought to enhance its translational utili

From center to edge: Prognostic implications of spatial metabolic heterogeneity, normalized hotspot-to-centroid and perimeter distance, on PET imaging. A systematic review and meta-analysis.

OBJECTIVE: Tumor heterogeneity remains a major challenge in oncology, influencing prognosis and treatment response. Novel PET-derived spatial radiomic features, including the normalized hotspot-to-centroid (NHOC) and normalized hotspot-to-perimeter (NHOP) distances, aim to quantify intratumoral metabolic heterogeneity and spatial distribution of metabolic activity. These biomarkers may provide biologically interpretable indicators of tumor aggressiveness and patient outcomes. The aim of this study was to systematically evaluate the prognostic and predictive performance of NHOC and NHOP derived

NeuroTrustNet: a cost-effective multimodal ensemble framework for brain tumor classification under cross-dataset variability.

Accurate brain tumor classification from Magnetic Resonance Imaging (MRI) remains challenging due to dataset heterogeneity, class imbalance, and limited interpretability, while practical deployment further requires models that balance predictive performance with computational efficiency. In this work, we propose NeuroTrustNet, an integrated multimodal framework that combines complementary convolutional neural network (CNN), Vision Transformer (ViT), and handcrafted radiomic representations through the proposed Adaptive Attention Stacking (AAS) mechanism, enabling sample-specific feature fusion

Feasibility and repeatability of DWI-derived diffusion heterogeneity for discriminating clinically significant prostate cancer.

PURPOSE: To evaluate feasibility and repeatability of entropy derived directly from diffusion weighted imaging (DWI) of prostate cancer (PCa), and compare its performance to entropy derived from the conventional monoexponential model calculated apparent diffusion coefficient (ADC). METHODS: 112 men with PCa underwent two repeated 3 T MRIs before prostatectomy. DWI was obtained using 12 b-values (0-2000 s/mm2). DWI entropy was derived directly from source DWI image using b-values of 100 s/mm2, 500 s/mm2, and 900 s/mm2. ADC-based radiomic features were calculated for comparison. Repeatability w

Graph-based multi-modality network for axillary lymph node metastasis prediction in early-stage breast cancer.

BACKGROUND AND OBJECTIVE: Accurate evaluation of the axillary lymph node (ALN) status in early-stage breast cancer is crucial for prognosis and guiding treatment decisions, while also preventing unnecessary surgeries and postoperative complications. Numerous methods have been proposed for ALN status classification using radiomic images which face the challenge of low diagnostic accuracy. This study aims to explore the potential of multi-modality that integrates clinical parameters and dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) to provide complementary information for image

Validated machine learning tools to distinguish immune checkpoint inhibitor, radiotherapy, COVID-19 and other infective pneumonitis.

BACKGROUND: Pneumonitis is a well-described, potentially disabling, or fatal adverse effect associated with both immune checkpoint inhibitors (ICI) and thoracic radiotherapy. Accurate differentiation between checkpoint inhibitor pneumonitis (CIP) radiation pneumonitis (RP), and infective pneumonitis (IP) is crucial for swift, appropriate, and tailored management to achieve optimal patient outcomes. However, correct diagnosis is often challenging, owing to overlapping clinical presentations and radiological patterns. METHODS: In this multi-centre study of 455 patients, we used machine learning

Texture Analysis of the Apparent Diffusion Coefficient Focused on Contrast-Enhancing Lesions in Predicting Survival for Bevacizumab-Treated Patients with Recurrent Glioblastoma.

PURPOSE: Glioblastoma often recurs after treatment. Bevacizumab increases progression-free survival in some patients with recurrent glioblastoma. Identifying pretreatment predictors of survival can help clinical decision making. Magnetic resonance texture analysis (MRTA) quantifies macroscopic tissue heterogeneity indirectly linked to microscopic tissue properties. We investigated the usefulness of MRTA in predicting survival in patients with recurrent glioblastoma treated with bevacizumab. METHODS: We evaluated retrospective longitudinal data from 33 patients (20 men; mean age 56 ± 13 years)

Radiophysiomics: Brain Tumors Classification by Machine Learning and Physiological MRI Data.

The precise initial characterization of contrast-enhancing brain tumors has significant consequences for clinical outcomes. Various novel neuroimaging methods have been developed to increase the specificity of conventional magnetic resonance imaging (cMRI) but also the increased complexity of data analysis. Artificial intelligence offers new options to manage this challenge in clinical settings. Here, we investigated whether multiclass machine learning (ML) algorithms applied to a high-dimensional panel of radiomic features from advanced MRI (advMRI) and physiological MRI (phyMRI; thus, radiop

A Novel Mouse Model of Radiation-Induced Cardiac Injury Reveals Biological and Radiological Biomarkers of Cardiac Dysfunction with Potential Clinical Relevance.

PURPOSE: Radiation-induced cardiotoxicity is a significant concern in thoracic oncology patients. However, the basis for this disease pathology is not well characterized. We developed a novel mouse model of radiation-induced cardiotoxicity to investigate pathophysiologic mechanisms and identify clinically targetable biomarkers of cardiac injury. EXPERIMENTAL DESIGN: Single radiation doses of 20, 40, or 60 Gy were delivered to the cardiac apex of female C57BL/6 mice ages 9-11 weeks, with or without adjacent lung tissue, using conformal radiotherapy. Cardiac tissue was harvested up to 24 weeks

A contrast‑enhanced CT histogram‑driven nomogram for predicting post‑radiotherapy liver regeneration in hepatocellular carcinoma.

BACKGROUND: Postoperative radiotherapy (RT) is increasingly used in hepatocellular carcinoma (HCC) patients with high-risk pathological features to reduce recurrence. However, RT may compromise liver regeneration (LR), particularly in patients with underlying hepatic dysfunction. Accurate evaluation of LR potential is therefore critical for individualized treatment planning. METHODS: In this retrospective, multicenter study, 315 HCC patients were enrolled—139 in the training cohort and 85 and 91 in two external validation cohorts. All patients underwent postoperative intensity-modulated radiot

Intraoperative Ex Vivo Shear-Wave Elastography of Sentinel Lymph Nodes in Endometrial Cancer and Other Gynaecological Malignancies.

BACKGROUND: Accurate intraoperative assessment of sentinel lymph node (SLN) status is critical for staging and guiding surgical management in gynaecological malignancies. Frozen-section histopathology remains the gold standard, but it is time-consuming and resource-intensive. Shear-wave elastography (SWE) quantifies tissue stiffness in real time and may offer a rapid alternative. METHODS: In this prospective single-centre study, 63 women (median age 62 years) undergoing primary surgery with sentinel lymph node biopsy (SLNB) for endometrial, cervical, vulvar, or early ovarian carcinoma were en

Predicting cardiovascular events in hemodialysis patients based on the fusion of physicochemical indicators and tongue images: a prospective and multicenter study.

BACKGROUND: Cardiovascular events (CVEs) are the leading cause of mortality in hemodialysis patients. Current prediction models rely on clinical and biochemical data, but non-invasive alternatives are needed. Inspired by the Traditional Chinese Medicine (TCM) principle that "the heart opens into the tongue," this study investigated whether quantitative features from tongue images could enhance CVE prediction. OBJECTIVE: To develop and validate a machine learning framework that integrates tongue image features with conventional clinical variables to predict CVEs in hemodialysis patients. METH

Beyond plaque and stenosis: Coronary inflammation imaging with the fat attenuation index.

Pericoronary adipose tissue (PCAT) imaging has emerged as a promising noninvasive marker of coronary inflammation and an adjunctive risk-stratification tool beyond conventional coronary computed tomography angiography (CCTA) findings such as luminal stenosis and plaque morphology. The fat attenuation index (FAI), derived from CCTA, quantifies phenotypic changes in pericoronary fat driven by vascular inflammation. The CRISP-CT study demonstrated that elevated FAI around the right coronary artery independently predicted cardiac mortality. The ORFAN study further established that the FAI-Score-an

AI-Driven Tumor Characterization and Histological Subtype Classification in Lung Cancer Using CT Imaging.

Background/Objectives: Lung cancer is still one of the top cancer mortality causes around the world, and there is a need for an accurate and clinically reliable diagnostic tool. While Computed Tomography (CT) imaging is very useful for evaluation of pulmonary nodules and tumor morphology, its interpretation is complicated by inter-patient variability, imaging artifacts, low tissue contrast, and tumor heterogeneity. Although Computer-Aided Diagnosis (CAD) systems have enhanced the diagnostic process, handcrafted feature-based approaches often fail to capture complex tumor characteristics, and n
