Antibody to transforming growth factor-beta ameliorates tubular apoptosis in unilateral ureteral obstruction.

BACKGROUND: Unilateral ureteral obstruction (UUO) is characterized by progressive renal atrophy, renal interstitial fibrosis, an increase in renal transforming growth factor-beta (TGF-beta), and renal tubular apoptosis. The present study was undertaken to determine the effect of a monoclonal antibody to TGF-beta (1D11) in UUO. METHODS: Mechanical stretch was applied to tubular epithelial cells (NRK-52E) by a computer-assisted system. Three doses of 1D11 (either 0.5, 2, or 4 mg/rat) were administered to rats one day prior to UUO and every two days thereafter, and kidneys were harvested at day

Arginine therapy of transgenic-knockout sickle mice improves microvascular function by reducing non-nitric oxide vasodilators, hemolysis, and oxidative stress.

In sickle cell disease, nitric oxide (NO) depletion by cell-free plasma hemoglobin and/or oxygen radicals is associated with arginine deficiency, impaired NO bioavailability, and chronic oxidative stress. In transgenic-knockout sickle (BERK) mice that express exclusively human alpha- and beta(S)-globins, reduced NO bioavailability is associated with induction of non-NO vasodilator enzyme, cyclooxygenase (COX)-2, and impaired NO-mediated vascular reactivity. We hypothesized that enhanced NO bioavailability in sickle mice will abate activity of non-NO vasodilators, improve vascular reactivity, d

Towards a stratified targeted approach with biologic treatments in rheumatoid arthritis: role of synovial pathobiology.

Rheumatoid Arthritis (RA) is a chronic, inflammatory, autoimmune disease affecting diarthrodial joints and extra-articular tissues; in the absence of an effective treatment, it is characterized by persistent symmetrical and erosive synovitis which leads to structural joint damage and lifelong disability. Several autoantibodies have been associated with RA such as rheumatoid factor (RF) and anti-citrullinated protein antibodies (ACPA). B cells have been shown to play a crucial role in the pathogenesis of RA by producing autoantibodies and promoting synovial inflammation through antigen presenta

Remnant cholesterol inflammation index (RCII) as a potential biomarker for ACPA-negative rheumatoid arthritis: evidence from dual-cohort analysis and serum lipidomics.

INTRODUCTION: Compared with anti-citrullinated protein autoantibody (ACPA)-positive RA, the etiology of ACPA-negative RA remains unclear, posing significant challenges to diagnosis. Recently, evidence has emerged that ACPA-negative RA is characterized by metabolic reprogramming, as well as dysregulated lipid metabolism. The remnant cholesterol inflammation index (RCII) is a comprehensive index of lipid burden and inflammation; this study evaluated the predictive value of RCII as a novel biomarker for ACPA-negative RA. METHODS: In the Beijing Hospital (BJH) cohort, we first compared RCII acros

Fecal metabonomics combined with 16S rRNA gene sequencing to analyze the changes of gut microbiota in rats with kidney-yang deficiency syndrome and the intervention effect of You-gui pill.

ETHNOPHARMACOLOGICAL RELEVANCE: A myriad of evidence have shown that kidney-yang deficiency syndrome (KYDS) is associated with metabolic disorders of the intestinal microbiota, while TCMs can treat KYDS by regulating gut microbiota metabolism. However, the specific interplay between KYDS and intestinal microbiota, and the intrinsic regulation mechanism of You-gui pill (YGP) on KYDS' gut microbiota remains largely unknown so far. MATERIALS AND METHODS: In the present study, fecal metabonomics combined with 16S rRNA gene sequencing analysis were used to explore the mutual effect between KYDS an

Heat shock protein 90 as an endogenous protein enhancer of inducible nitric-oxide synthase.

Nitric oxide (NO) generated by inducible NO synthase (iNOS) plays crucial roles in inflammation and host defense. With an intrinsically bound calmodulin, iNOS is fully active once expressed in cells. Thus, regulation of NO production from iNOS was thought to primarily occur at the enzyme transcriptional level. Here we show that NO synthesis from iNOS can be profoundly modulated by heat shock protein 90 (hsp90) through protein-protein interaction. To study whether hsp90 affects iNOS function, recombinant murine iNOS was purified from an Escherichia coli expression system by affinity chromatogra

Danshen-Chuanxiong suppresses ROS-MAPK-mediated neutrophil extracellular traps to alleviate deep vein thrombosis without increasing the risk of bleeding.

ETHNOPHARMACOLOGICAL RELEVANCE: Danshen-Chuanxiong (DS-CX), a classical herb pair composed of Salviae Miltiorrhizae Radix et Rhizoma (the dried roots and rhizomes of Salvia miltiorrhiza Bunge) and Chuanxiong Rhizoma (the dried rhizomes of Ligusticum chuanxiong Hort.), has been widely used in traditional Chinese medicine (TCM) for promoting blood circulation and resolving blood stasis. DS-CX has been developed into Guanxinning preparations, which are widely used to treat cardiovascular disorders. However, the potential role of DS-CX in deep vein thrombosis (DVT) and its underlying mechanisms re

[The immunologic homunculus in rheumatoid arthritis. A new viewpoint of immunopathogenesis in rheumatoid arthritis and therapeutic consequences].

Autoreactivity plays a major role in the pathogenesis of RA. The rheumatoid factor has been and still is for now more than 50 years the only autoreactivity that is clinically applied in the diagnosis of RA. This well reflects the current way of thinking that a single antigen or a single cause drives an individual into disease. Although by now many other autoantigens and autoreactivities have been described, their discovery was always on the search for the one and only autoreactivity that causes RA. This includes also immune reactivities directed against xenogenic antigens. But, none of the kno

Circulating Nucleosomes are Elevated in Trauma Patients with Venous Thromboembolism: A Prospective Case-Cohort Study.

OBJECTIVE: The identification of reliable biomarkers in trauma patients (pts) remains a clinical challenge. Nucleosomes, which are DNA wrapped around histone protein cores, have been used as reliable markers for quantification in sepsis, acute respiratory distress syndrome, and lymphoma. We hypothesized that: 1) levels of circulating nucleosomes quantified after traumatic injury, specifically histone H3.1 and its citrullinated R8 posttranslation modification (H3R8 Cit), would be increased after trauma and 2) that levels of circulating nucleosomes would be further, increased in pts who went on

Unveiling the Clinical and Biochemical Impact of Vitamin D Deficiency in Indian Rheumatoid Arthritis Patients: A Cross-Sectional Analysis.

Background Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent synovial inflammation, joint destruction, and systemic complications. Despite therapeutic advances, disease activity remains variable, and modifiable factors influencing severity are under investigation. Vitamin D, beyond its role in bone metabolism, exerts immunomodulatory effects and may influence RA pathogenesis. However, evidence regarding its association with disease activity remains inconsistent. This study aimed to evaluate the clinical and biochemical correlates of serum 25-hydroxyvitamin D
