Genetic approach to study the relationship between penicillin-binding protein 3 mutations and Haemophilus influenzae beta-lactam resistance by using site-directed mutagenesis and gene recombinants.

To clarify the relationship between mutations commonly found for penicillin-binding protein 3 (PBP 3) of beta-lactamase-nonproducing ampicillin-resistant (BLNAR) Haemophilus influenzae isolates and beta-lactam resistance, single and multiple amino acid mutations at positions 377, 385, 389, 517, and 526 were introduced into PBP 3 of a beta-lactam-susceptible Rd strain by site-directed mutagenesis. Twelve isogenic recombinant strains were challenged with nine beta-lactam antibiotics. Replacement of the asparagine at position 526 with lysine (N526K) increased the resistance to imipenem eightfold

Phenotypic and genetic diversity of invasive pneumococcal isolates recovered from French children.

We investigated the phenotypic and genetic diversity (by ribotyping) of Streptococcus pneumoniae isolates recovered from French children, by blood culture from meningitis-free patients (n = 244) and by cerebrospinal fluid culture from patients with meningitis (n = 154). Isolates belonging to serotypes associated with carriage and penicillin-resistant isolates were significantly more frequent in children under 2 years of age than in older children. The seven-valent pneumococcal conjugate vaccine covered 68% of strains associated with bacteremia and 61% of strains associated with meningitis in c

Complementation of a speA negative Streptococcus pyogenes with speA: effects on virulence and production of streptococcal pyrogenic exotoxin A.

We have shown previously that an isogenic SPEA-negative Streptococcus pyogenes strain did not attenuate virulence in a murine model of necrotizing fasciitis. The aim of this study was to confirm that streptococcal pyrogenic exotoxin A (SPEA) is not crucial for streptococcal invasiveness in murine invasive infection. The SPEA-negative S. pyogenes (H326) was complemented with speA extra-chromosomally to create strain H361 which produced 2.2-fold more SPEA compared with the parental speA(+)wild-type (H305). The growth phase-regulated expression of SPEA in vitro was unaffected in this strain. Comp

Overexpression of L-glutamine:D-fructose-6-phosphate amidotransferase provides resistance to methylmercury in Saccharomyces cerevisiae.

To identify novel genes that confer resistance to methylmercury (MeHg), a yeast genomic DNA library was transfected into Saccharomyces cerevisiae. Two functional plasmids were isolated from transfected yeast clones D1 and H5 that exhibited resistance to MeHg. The yeast transfected with plasmid isolated from clone H5 was several-fold more resistant than yeast transfected with plasmid from clone D1. Functional characterization of the genomic DNA fragment obtained from clone H5 determined that the GFA1 gene conferred resistance to MeHg. GFA1 was reported to encode L-glutamine:D-fructose-6-phospha

Studies on multiple antibiotic resistance in obligate anaerobes.

Forty-eight Gram-negative obligate anaerobic bacterial strains were isolated from faeces on selective media and their antibiotic resistance spectra were determined. R factor transfer was not detected in a series of experiments involving matings among the anaerobes, nor between the anaerobes and known donor and recipient facultative E. coli strains. Antibiotic resistance plasmids could not be demonstrated by treatment with acridine dyes and ethidium bromide in four multiply-resistant strains.

Characterization of Staphylococcus aureus strains isolated from bovine milk in Hungary.

Staphylococcus aureus is a major foodborne pathogen due to its capability to produce a wide range of heat-stable enterotoxins. The primary purpose of this research was to characterize S. aureus isolates recovered from mammary quarter milk of mastitic cows and from bulk tank milk produced on Hungarian dairy farms of different sizes. Macrorestriction analysis of chromosomal DNA from S. aureus isolates was performed using the restriction enzyme SmaI followed by pulsed-field gel electrophoresis (PFGE). The prevalence rates of nine S. aureus enterotoxin genes (sea, seb, sec, sed, see, seg, seh, sei

A Review of Inflammatory Bowel Disease: A Model of Microbial, Immune and Neuropsychological Integration.

Objective: Inflammatory bowel diseases (IBDs) are complex chronic inflammatory disorders of the gastro-intestinal (GI) tract with uncertain etiology. IBDs comprise two idiopathic disorders: Crohn's disease (CD) and ulcerative colitis (UC). The aetiology, severity and progression of such disorders are still poorly understood but thought to be influenced by multiple factors (including genetic, environmental, immunological, physiological, psychological factors and gut microbiome) and their interactions. The overarching aim of this review is to evaluate the extent and nature of the interrelationsh

Not so innocent: Environmental Klebsiella pneumoniae disseminating resistance and virulence via conjugation.

The acquisition of carbapenem resistance and hypervirulence traits in Klebsiella pneumoniae results in the emergence of certain high-risk clones posing significant public health threat. This convergence is largely driven by conjugative plasmids, however, there are limited studies on environmental reservoirs and transmission potential of such isolates. In this study, we investigated the phenotypic trait for hypervirulence in Carbapenem-resistant hypervirulent Klebsiella pneumoniae (CR-hvKp) isolated from a river system in Odisha, India, assessed their ability to co-transfer carbapenemase and vi

Novel Two-Component System of Streptococcus sanguinis Affecting Functions Associated with Viability in Saliva and Biofilm Formation.

Streptococcus sanguinis is a pioneer species of teeth and a common opportunistic pathogen of infective endocarditis. In this study, we identified a two-component system, S. sanguinis SptRS (SptRS Ss ), affecting S. sanguinis survival in saliva and biofilm formation. Isogenic mutants of sptRSs (SKsptR) and sptSSs (SKsptS) showed reduced cell counts in ex vivo assays of viability in saliva compared to those of parent strain SK36 and complemented mutants. Reduced counts of the mutants in saliva were associated with reduced growth rates in nutrient-poor medium (RPMI) and increased susceptibility t
