Hepatoprotection with a chloroform extract of Launaea procumbens against CCl4-induced injuries in rats.

BACKGROUND: Launaea procumbens (Asteraceae) is used as a folk medicine to treat hepatic disorders in Pakistan. The effect of a chloroform extract of Launaea procumbens (LPCE) was evaluated against carbon-tetrachloride (CCl4)-induced liver damage in rats. METHODS: To evaluate the hepatoprotective effects of LPCE, 36 male Sprague-Dawley rats were equally divided into six groups. Animals of group 1 (control) had free access to food and water. Group II received 3 ml/kg of CCl4 (30% in olive oil v/v) via the intraperitoneal route twice a week for 4 weeks. Group III received 1 ml of silymarin via g

Increased vulnerability of neuronal cell lines to sodium nitroprusside-mediated toxicity is caused by the decreased level of nitric oxide metabolites.

Nitric oxide (NO) is an unstable radical produced during the oxidative deamination catalyzed by NO synthase (NOS) that converts L-arginine to L-citrulline. NO is also generated nonenzymatically from a group of compounds, called NO donors, such as sodium nitroprusside (SNP). NO directly or through its metabolites has been implicated in several disorders, including Alzheimer's disease (AD). Since NO is a highly labile unstable free gas, we measured the stable end products, nitrite and nitrate (NOx). Here, we investigated the effect of SNP-mediated NO release in different cell types and its effec

Six Dehydrogenase Gatekeepers of Carbohydrate Metabolism: Metabolic Integration in Health and Disease.

Dehydrogenases function as metabolic gatekeepers, regulating carbon flux, redox balance, and biosynthetic capacity at critical branch points in cellular metabolism. This narrative review examines six key dehydrogenases, namely glyceraldehyde-3-phosphate dehydrogenase (GAPDH), lactate dehydrogenase (LDH), pyruvate dehydrogenase complex (PDHC), malate dehydrogenase (MDH1/2), isocitrate dehydrogenase (IDH1/2/3), and glucose-6-phosphate dehydrogenase (G6PDH), that collectively orchestrate the partitioning of nutrients among energy production, biosynthesis, and redox homeostasis. These enzymes shar

Renal tissue microbiota and metabolite profiling reveal dysregulated signatures in diabetic kidney disease.

INTRODUCTION: Diabetic kidney disease (DKD) has become the main cause of end-stage renal disease in China. Mounting evidence links microecological disorders to DKD progression. However, the composition and the functional characteristics of the renal microecology in DKD patients remain poorly defined. This study characterized renal tissue microbiota and metabolite profiles in DKD patients, aiming to provide insights for novel diagnostic strategies for DKD. METHODS: Renal tissue microbiome was analyzed in 42 DKD patients and 10 controls via 16S ribosomal DNA sequencing. Renal metabolomics was p

Acute intestinal intussusception revealing Peutz-Jeghers Syndrome in a 16-year-old male: a case report.

INTRODUCTION AND IMPORTANCE: Peutz-Jeghers Syndrome (PJS) is a rare autosomal dominant disorder characterized by hamartomatous polyps throughout the gastrointestinal tract and mucocutaneous pigmentation. Patients are at high risk for complications such as bleeding, obstruction, and especially small-bowel intussusception, which often occurs in childhood or adolescence. Early recognition is essential, as intussusception can be the first life-threatening presentation of PJS. This case highlights the importance of considering PJS in young patients presenting with acute intestinal obstruction. PRE

Preprocedural Biomarkers of Inflammation and Fibrosis and Echocardiographic Markers of Myocardial Remodeling in New-Onset Conduction Disorders After Transcatheter Aortic Valve Implantation.

Background and Objectives: Conduction disorders (CDs) and permanent pacemaker implantation (PPI) remain among the most common complications after transcatheter aortic valve implantation (TAVI). Given their potential impact on long-term outcomes, improved preprocedural identification of patients at high risk for new-onset CDs is increasingly important. We investigated whether selected preprocedural inflammatory and fibrotic biomarkers, along with echocardiographic indices of regional myocardial remodeling, were associated with new-onset CDs after TAVI. Materials and Methods: This single-center

Diamine oxidase as a biomarker for depression and disease activity in inflammatory bowel disease: A cross-sectional observational study.

BACKGROUND: Diamine oxidase (DAO) is secreted by epithelial cells in the intestinal villi, and its serum levels are elevated after intestinal mucosal damage. d-lactate (D-LA) is a gut microbial metabolite that can enter the systemic circulation if intestinal barrier function is impaired. Both DAO and D-LA are serum markers of small bowel mucosal integrity, and can be valuable biomarkers of intestinal barrier damage in inflammatory bowel disease (IBD). Intestinal barrier dysfunction was recently found to contribute to psychological symptoms in IBD patients. However, the correlations among DAO,

WNT-inhibitory factor 1-mediated glycolysis protects photoreceptor cells in diabetic retinopathy.

BACKGROUND: In diabetic retinopathy (DR), hypoxia-inducible factor (HIF-1α) induces oxidative stress by upregulating glycolysis. This process leads to neurodegeneration, particularly photoreceptor cell damage, which further contributes to retinal microvascular deterioration. Further, the regulation of Wnt-inhibitory factor 1 (WIF1), a secreted Wnt signaling antagonist, has not been fully characterized in neurodegenerative eye diseases. We aimed to explore the impact of WIF1 on photoreceptor function within the context of DR. METHOD: Twelve-week-old C57BL/KsJ-db/db mice were intravitreally inj
