Antral acidification and the gastric secretory response to pentagastrin and acetylcholine in dogs.

In 6 dogs with innervated antral pouches (3 with vagally innervated fundic pouches and 3 with vagally denervated fundic pouches) gastric sec … the acid response of the fundic pouch to perfusion of the innervated antral pouch with 0–5 per cent acetylcholine was significantly reduced when the pH of the perfusate was lowered from 7.0 to 1.0. The results provide no support for the existence of either an antral inhibitory hormone or inhibitory neural reflex active against pentagastrin-stimulated gastric se …

Loss of α7 nicotinic acetylcholine receptor exacerbates adrenergic-induced cardiac damage.

Previous studies have implicated the α7 nicotinic acetylcholine receptor (α7nAChR) in cardioprotection via its anti-inflammatory effects, yet the underlying mechanisms remain poorly understood. Here, we investigated the impact of α7nAChR deficiency on cardiac injury induced by a 7-day isoproterenol (ISO) treatment in littermate wild-type (WT) and α7nAChR-knockout (α7-KO) mice. ISO administration in WT mice led to a marked upregulation of α7nAChR expression in cardiac tissue and isolated cardiomyocytes, suggesting a compensatory response to adrenergic stress. To investigate this hypothesis, we …

α6 nicotinic acetylcholine receptors: A pharmacological challenge with untapped therapeutic promise.

α6-containing nicotinic acetylcholine receptors (α6∗ nAChRs), particularly the α6β2∗ and α6β4∗ subtypes, are promising therapeutic targets for neurological disorders such as nicotine addiction, Parkinson's disease, and pain. However, progress is significantly hampered by two interrelated challenges: difficulties in achieving robust functional expression in vitro and the limited availability of subtype-selective ligands. This review synthesizes the current advances in overcoming these obstacles. We detail the evolution of in vitro models, highlighting the success of engineered constructs such a…

Nicotinic acetylcholine receptors regulate growth hormone in pituitary somatotrophs of tigers.

The Felidae exhibits remarkable diversity in body size, with lengths ranging from 50 to 370 cm and weights from 1.1 to 423 kg. However, the underlying mechanisms driving this variation remain poorly understood. Here, we focused on the Siberian tiger (Panthera tigris altaica), the largest of the six extant tiger subspecies, and revealed the surprising expression of nicotinic acetylcholine receptors (nAChRs) in pituitary somatotrophs, which are crucial for regulating growth hormone (GH) secretion. Single-nucleus RNA sequencing of Siberian tiger pituitary cells exhibited the coexpression of CHRNA…

Responsiveness to Tocilizumab in Anti-Acetylcholine Receptor-Positive Generalized Myasthenia Gravis.

Tocilizumab, a humanized IL-6R monoclonal antibody, has been used in autoimmune diseases closely related to humoral immunity. This report aims to evaluate the efficacy and safety in patients with anti-acetylcholine receptor-positive (AChR+) generalized myasthenia gravis (gMG). We performed a prospective, open-label, single-arm study in patients with gMG in a 48-week follow-up. All patients were AChR+ and were given tocilizumab by intravenous infusion at a dose of 8 mg/kg at intervals of 4 weeks. The primary endpoint was mean change from baseline in quantitative MG (QMG) score at week 12. The s…

baz-2 enhances systemic proteostasis in vivo by regulating acetylcholine metabolism.

Neurodegenerative disorders, such as Alzheimer's disease (AD) or Huntington's disease (HD), are linked to protein aggregate neurotoxicity. According to the "cholinergic hypothesis," loss of acetylcholine (ACh) signaling contributes to the AD pathology, and therapeutic restoration of ACh signaling is a common treatment strategy. How disease causation and the effect of ACh are linked to protein aggregation and neurotoxicity remains incompletely understood, thus limiting the development of more effective therapies. Here, we show that BAZ-2, the Caenorhabditis elegans ortholog of human BAZ2B, limi…

Functional Characterization of the Oxantel-Sensitive Acetylcholine Receptor from Trichuris muris.

The human whipworm, Trichuris trichiura, is estimated to infect 289.6 million people globally. Control of human trichuriasis is a particular challenge, as most anthelmintics have a limited single-dose efficacy, with the striking exception of the narrow-spectrum anthelmintic, oxantel. We recently identified a novel ACR-16-like subunit from the pig whipworm, T. suis which gave rise to a functional acetylcholine receptor (nAChR) preferentially activated by oxantel. However, there is no ion channel described in the mouse model parasite T. muris so far. Here, we have identified the ACR-16-like and …

Acetylcholine-induced whealing in cholinergic urticaria - What does it tell us?

BACKGROUND: Cholinergic urticaria (CholU) is characterized by the occurrence of itchy wheals induced by sweating. Intradermal injections of acetylcholine (ACh) have been proposed to help with diagnosing CholU and subgrouping of patients, but controlled studies are largely missing. OBJECTIVE: To compare the rates of positive ACh test results in well characterized CholU patients and controls and to identify clinical features of CholU linked to ACh reactivity. METHODS: Acetylcholine was injected intradermally into 38 CholU patients and 73 matched healthy controls. Wheal and flare skin responses…

Ganglionic Acetylcholine Receptor Antibodies and Autonomic Dysfunction in Autoimmune Rheumatic Diseases.

Autonomic neuropathy has been reported in autoimmune rheumatic diseases (ARD) including Sjögren's syndrome, systemic sclerosis, rheumatoid arthritis, and systemic lupus erythematosus. However, the pathophysiological mechanism underlying autonomic dysfunction remains unknown to researchers. On the other hand, autoimmune autonomic ganglionopathy (AAG) is an acquired immune-mediated disorder, which causes dysautonomia that is mediated by autoantibodies against ganglionic acetylcholine receptors (gAChRs). The purpose of this review was to describe the characteristics of autonomic disturbance throu…

M3 muscarinic acetylcholine receptor-reactive Th17 cells in primary Sjögren's syndrome.

M3 muscarinic acetylcholine receptor (M3R) is one of the autoantigens associated with Sjögren's syndrome (SS) and is localized in exocrine glands where disease-specific inflammation occurs. The inflammatory lesion is characterized by infiltration of CD4+ T cells, including clonally expanded Th17 cells. We undertook this study to identify circulating M3R-specific Th17 cells and to determine functional properties of those cells. Using the enzyme-linked immunospot assay (ELISpot) method, we identified M3R-reactive Th17 cells in the peripheral blood of patients with primary SS (pSS). Among 10 exam…

False-positive acetylcholine receptor antibody results in patients without myasthenia gravis.

Acetylcholine receptor antibodies are very specific for myasthenia. During a large prospective cohort study of myasthenia, we encountered five patients, positive for acetylcholine receptor (AChR) antibodies by radioimmunoprecipitation assay (RIA), whose clinical course revealed diagnoses other than myasthenia. Two patients had transiently raised AChR antibodies associated with Guillain-Barré syndrome. Antibodies to clustered AChRs, in a live cell-based assay, were negative in all five patients, suggesting that results from the RIAs were false-positives. It is possible that the AChR antibodies …

Regulation of nicotinic acetylcholine receptor α3 subtype in adipose tissue dysfunction.

Nicotinic acetylcholine receptor α3 subtype (α3-nAChR) plays a pivotal role in regulating inflammatory responses. Inflammation leads to the adipose tissue dysfunction and further increases the risk of metabolic and cardiovascular diseases. Therefore, we hypothesize that α3-nAChR could regulate the disorder of adipose functions. Adipocytokines and inflammatory cytokines were evaluated in apolipoprotein E knockout (ApoE-/-) mice after α3-nAChR was antagonized and in adipocytes after α3-nAChR gene was silenced. Results showed that in high fat diet-fed ApoE-/- mice with α3-nAChR blocked and in IL-…
