Chronobiological rhythms contribute to our understanding of the effect of alcohol and of several features of alcoholism. The observation of infradian periodicities suggests preventive seasonal strategies aimed at reducing the consumption of alcohol and its consequences. Circadian variations in blood alcohol levels and behavioral impairment indicate a complex temporal rhythmicity with both physiological and psychosocial determinants. Circadian differences are noted among possible mediators of susceptibility towards alcoholism. Ultradian patterns may be of prognostic value in assessing risks suc

The relative importance of malnutrition and alcohol toxicity in the pathogenesis of cirrhosis has been controversial. In epidemiological studies the incidence of cirrhosis can be correlated with the duration and amount of alcohol imbibed. The importance of nutrition has been discounted. In these studies few analyses of dietary intake were included. Diets of patients with alcoholic cirrhosis characteristically are poor. Furthermore, alcohol toxicity impairs nutrition by interfering with absorption, transport, and utilization of essential nutrients. Patients with cirrhosis respond favorably to n

Acute alcoholic hepatitis is the first alcoholic lesion of the liver in the process of progression to cirrhosis. It is due to the toxic action of alcohol on hepatocytes, in particular in the centrolobular region. It may affect a liver which is the site of fatty infiltration, fibrosis or cirrhosis, i.e. during all the stages of alcoholic liver disease. Its severity depends upon the degree of alcoholic intoxication. It may be fatal by malignant hepatic failure in a quarter of cases or, at the extreme, be totally asymptomatic. The aims of treatment are: 1) in the immediate, to prevent death; 2) s

Alcohol intoxication is probably the most common intoxication worldwide, and may be lethal. The exact mechanism by which ethanol intoxication contributes to death is unknown, although ventricular tachyarrhythmias degenerating into fibrillation is a possible cause. Alcoholics have increased risk of sudden death and, possibly, higher risk than occasional drinkers. In 32 consecutive patients with alcohol intoxication Delta-voltage was the differences in voltage between values at admission (when patients had high blood alcohol levels) and at discharge (when the patients were assumed sober), and wa

Chronic alcohol consumption results in considerable damage to many of the body's organs, and particularly to the brain. Beyond the confusional state occurring with acute intoxication or withdrawal, alcohol abuse is responsible of a constellation of neuropsychiatric syndromes including cognitive dysfunction, Wernicke-Korsakoff Syndrome, alcoholic cerebellar degeneration, Marchiafava-Bignami disease and alcohol-related dementia, ARD. ARD would account for nearly 20% of all admissions to state mental hospitals in the United-States. According to the DSM-IV, ARD is defined by a dementia associated

The main focus of this work is to clarify how protein macromolecules become solvated in monohydric alcohols with low water content. Alcohols with varying hydrophobicity (1-propanol, ethanol, and methanol) were used as model organic liquids. Two independent experimental techniques (isothermal immersion calorimetry and water/alcohol adsorption) were employed to provide complementary thermodynamic functions of the preferential solvation of hen egg-white lysozyme in alcohols with low and high water contents at 298.15 K. The advantage of this approach is that the enthalpies and Gibbs energies of wa…

Homoallylic alcohols are valuable motifs, but three-component syntheses from butadiene, carbonyl compounds, and organic electrophiles often require stoichiometric metal reductants. We report a nickel/photoredox-catalyzed coupling of 1,3-butadiene, aldehydes, and aryl bromides using Hantzsch ester as an organic reductant. The reaction proceeds under mild conditions with high regio- and stereoselectivity and broad functional-group tolerance. Control experiments and DFT calculations support a pathway involving nickel photoreduction, butadiene insertion, and carbonyl addition of a nucleophilic π-a…

The enantioselective synthesis of chiral diaryl alcohols, crucial pharmaceutical intermediates, remains challenging due to the poor stereochemical differentiation between similar aryl groups. We address this by reporting a highly enantioselective Rh-catalyzed transfer hydrogenation of cubane-aryl ketones. Leveraging the three-dimensional cubane as a sterically distinct surrogate for a flat aryl group, this method provides chiral cubane-aryl alcohol bioisosteres in excellent yield (up to 99%) and enantioselectivity (up to >99% ee). DFT studies implicate key noncovalent C-H···π interactions in t…

Catalytic carbon-carbon bond-forming reactions provide the basis for synthetic organic chemistry. To move towards a circular carbon economy, it is imperative to efficiently convert sustainable and inexpensive starting materials, e.g. broadly available renewable feedstocks, in an economically viable and practical manner. In this work , we present a method for the atom-efficient and "waste-free" construction of C-C bonds in ketones with alcohols, including lignin-derived compounds. The broad applicability of this protocol is based on the development of a novel heterogeneous cobalt-based single a…

A divergent synthetic strategy to access γ-arylated ketones and α-arylated tetrahydrofurans from readily available homopropargylic alcohols and arene nucleophiles is reported. This method expands the accessible chemical space of γ-arylated ketones via a unique triflic acid-catalyzed hydroalkoxylation/ring-opening arylation sequence proceeding through a 2,3-dihydrofuran intermediate, enabled by the properties of 1,1,1,3,3,3-hexafluoroisopropan-2-ol (HFIP) as solvent. This protocol provides a solution for preparing γ-arylated ketones incorporating sterically hindered arenes, while displaying com…

The catalytic conversion of lignin into N-containing aromatic compounds expands lignin's potential for high-value agrochemicals. However, achieving selective C-C bond cleavage and subsequent C-N bond formation with compatible catalysts remains challenging. This work reports an inexpensive CuI-catalyzed oxidative amidation of lignin β-O-4 moieties, primary alcohols, and secondary alcohols with aromatic amines under an oxygen atmosphere, yielding diverse functionalized aromatic amides in moderate to high yields, up to 86%. Mechanistic investigations suggest a reaction pathway involving the selec…

A copper-catalyzed one-pot (2 + 4) cycloaddition of propargyl alcohols with o-hydroxyphenyl-substituted secondary phosphine oxides was established, affording a series of six-membered P(V)-containing heterocycles in high yields (up to 98%). More importantly, biological activity screening of these six-membered P(V)-containing heterocycles demonstrated their in vitro inhibitory effect on the proliferation of MCF-7 human breast cancer cells. This reaction not only represents the first cycloaddition involving propargyl alcohols with phosphorus-containing dinucleophiles but also provides access to a…

Alcohol coupling reactions that are induced by alcohol dehydrogenation, and generate hydrogen or water as the only byproducts, have become a well-recognized way to carry out important synthetic transformations, such as esterification and alkylation, in a green and atom-economical fashion. Herein, we report a new type of alcohol-alcohol coupling reaction that involves the dehydrogenative annulation of ethylene glycol with secondary alcohols to give 1,2-cyclopentanedione derivatives in a single synthetic step. This process, which is catalyzed by a pincer complex of earth-abundant manganese, repr…

Sugar alcohols such as mannitol and sorbitol are frequently used as pharmaceutical excipients in oral dosage forms. Their versatility stems from extensive evidence of use in humans. However, investigations over many years have clearly indicated their osmolarity-derived biopharmaceutics effects on oral drug absorption. Despite this, these accumulated insights have not been fully integrated into pharmaceutical formulation development or clinical use, leading to suboptimal industrial and clinical practices. This review provides a comprehensive summary of the biopharmaceutics of sugar alcohols, de…
