Rational design, molecular characterization and dissociation mechanisms of PROTACs targeting hFAAH.

Fatty acid amide hydrolase (FAAH) is a key enzyme in the endocannabinoid system and a promising therapeutic target for pain and depression. However, traditional FAAH inhibitors have faced clinical setbacks due to their limited efficacy and off-target effects. Here, we present the rational design and molecular characterization of C13p, a novel PROTAC molecule targeting hFAAH, composed of a 13-carbon linker connecting the FAAH ligand Carb4d and the CRBN ligand pomalidomide. Our results demonstrate that C13p significantly enhances the protein-protein interactions between hFAAH and CRBN, with a bi

Fibroblastic aspartoacylase suppresses TGFβ-mediated responses and cancer progression.

Metabolic reprogramming is a hallmark of cancer, and the field has predominantly focused on investigating metabolic alterations in tumour cells. However, the relevance, mechanism and consequences of metabolic adaptations in stromal cells remain understudied. Here, we identify aspartoacylase (ASPA) as a metabolic enzyme consistently repressed in tumour stroma and cancer-associated fibroblasts (CAFs). Importantly, we report a reciprocal crosstalk between ASPA and Transforming Growth Factor Beta (TGFβ) signalling that influences fibroblast behaviour. TGFβ suppresses ASPA expression in fibroblasts

Thermostability Engineering of a Chitin-Utilizing GlcNAc Deacetylase from Acinetobacter baumannii via Distal Mutations.

The deacetylation of N-acetylglucosamine (GlcNAc) to glucosamine (GlcN) is a key step in enzymatic chitin valorization. We identified a novel GlcNAc deacetylase (AbNGD) from Acinetobacter baumannii with high substrate specificity and catalytic activity but poor thermal stability (half-life of 5.5 h at 37 °C). Using the FireProt platform, we generated a combinatorial mutant, AbNGD-M3 (T93P-C153T-A244M). Its optimal temperature increased to 37 °C, and its half-life reached 28.1 h (5.1-fold improvement), while retaining 97% of wild-type catalytic efficiency (kcat/Km = 3.4 mM-1·s-1). Site-directed

Targeting microbial bile salt hydrolase reprograms bile acid metabolism and ameliorates metabolic dysfunction-associated steatohepatitis in mice.

Microbial bile salt hydrolase (BSH) plays a central role in shaping bile acid composition and gut-liver metabolic signaling, yet its therapeutic potential in metabolic dysfunction-associated steatohepatitis (MASH) remains incompletely defined. Here, we evaluated the efficacy of the non-absorbable BSH inhibitor GR-7 in a diet-induced mouse model of steatohepatitis using early and late intervention strategies with different dosing regimens. GR-7 reduced food intake and exerted stage- and dose-dependent therapeutic effects, with early intervention robustly suppressing hepatic fibrosis even at a l

Effects of mutations conferring pyrazinamide resistance among multidrug-resistant tuberculosis isolates from China.

UNLABELLED: To investigate the effects of mutations on pyrazinamide (PZA)-associated regions, including pncA, rpsA, panD, and clpC1, among multidrug-resistant tuberculosis (MDR-TB) isolates from China. A total of 139 MDR-TB isolates from China underwent PZA susceptibility testing and genotyping, and the sequences of the four PZA resistance-associated regions were analyzed. The association between lineage and PZA resistance was analyzed, as well as that between lineage and mutation. Among 139 MDR-TB isolates, 62 were resistant to PZA. Of these 62 PZA-resistant isolates, 60 harbored at least one

KLF3 aggravates renal fibrosis in chronic kidney disease through transcriptional activation of DDAH2.

Renal fibrosis is recognized as a critical pathway leading to end-stage renal disease in nearly all cases of chronic kidney disease (CKD). Analysis of the gene expression omnibus (GEO) database revealed that the transcription factor Krüppel-like factor 3 (KLF3) was elevated in kidney tissues of CKD patients, however, its specific role in CKD-associated renal fibrosis is yet to be deciphered. This study aimed to investigate the function of KLF3 on unilateral ureteral obstruction (UUO) mice and transforming growth factor-β1 (TGF-β1)-stimulated HK2 cells. Our data demonstrated that KLF3 was remar

HNF4A Regulated APEH Deficiency Promotes UPR Activation in Diabetic Kidney Disease.

Diabetic kidney disease (DKD) constitutes a severe microvascular complication of diabetes and ranks among the leading causes of end-stage renal disease (ESRD). Endoplasmic reticulum stress (ERS) significantly contributes to the initiation and progression of DKD, as persistent ERS leads to excessive activation of the unfolded protein response (UPR), triggering cellular apoptosis. Protein carbonylation, a prominent oxidative modification caused by reactive oxygen species (ROS), is recognized as a critical initiator of ERS. Acyl peptide enzyme hydrolase (APEH) demonstrates cytoprotective properti

Fatty acid amide hydrolase gene inactivation induces hetero-cellular potentiation of microglial function in the 5xFAD mouse model of Alzheimer's disease.

Neuroinflammation has recently emerged as a crucial factor in Alzheimer's disease (AD) etiopathogenesis. Microglial cells play an important function in the inflammatory response; specifically, the emergence of disease-associated microglia (DAM) has offered new insights into the conflicting perspectives on the detrimental or beneficial roles of microglia. We previously showed that modulating the endocannabinoid tone by fatty acid amide hydrolase (FAAH) inactivation renders beneficial effects in an amyloidosis context, paradoxically accompanied by an exacerbated neuroinflammatory response and th

Cytosolic N-terminal formyl-methionine deformylation derives cancer stem cell features and tumor progression.

Eukaryotic cells can synthesize formyl-methionine (fMet)-containing proteins not only in mitochondria but also in the cytosol to some extent. Our previous study revealed substantial upregulation of N-terminal (Nt)-fMet-containing proteins in the cytosol of SW480 colorectal cancer cells. However, the functional and pathophysiological implications remain unclear. Here, we demonstrated that removal of the Nt-formyl moiety of Nt-fMet-containing proteins (via expressing Escherichia coli PDF peptide deformylase) resulted in a dramatic increase in the proliferation of SW480 colorectal cancer cells. Cholesterol-lowering effect of bile salt hydrolase from a Lactobacillus johnsonii strain mediated by FXR pathway regulation.

Hypercholesterolemia is a major risk factor for cardiovascular diseases worldwide. Healthy intestinal microbiota can contribute to reducing the high cholesterol symptoms by producing bile salt hydrolase (BSH). In this study, recombinant BSH from the strain L. johnsonii 334 with high cholesterol reduction ability was selected to study the cholesterol-lowering mechanism mediated by farnesoid X receptor (FXR) regulation in mice. In the presence of recombinant BSH, mice had a larger bile acid pool. Analysis of individual bile acids revealed that bile acid composition was affected not only by recom

A Recombinant Dimethylarginine Dimethylaminohydrolase-1-Based Biotherapeutics to Pharmacologically Lower Asymmetric Dimethyl Arginine, thus Improving Postischemic Cardiac Function and Cardiomyocyte Mitochondrial Activity.

High serum levels of asymmetric dimethyl arginine (ADMA) are associated with cardiovascular disease and mortality. Pharmacological agents to specifically lower ADMA and their potential impact on cardiovascular complications are not known. In this study, we aimed to investigate the effect of specific lowering of ADMA on myocardial response to ischemia-reperfusion injury (I/R) and direct effects on cardiomyocyte function. Effects of recombinant dimethylarginine dimethylaminohydrolase (rDDAH)-1 on I/R injury were determined using isolated mouse heart preparation. Respiration capacity and mitochon

Chronic Inhibition of FAAH Reduces Depressive-Like Behavior and Improves Dentate Gyrus Proliferation after Chronic Unpredictable Stress Exposure.

Symptoms of depressive disorders such as anhedonia and despair can be a product of an aberrant adaptation to stress conditions. Chronic unpredictable stress model (CUS) can generate an increase in the activity of the hypothalamic-pituitary-adrenal axis (HPA) and induce a reduction of neurotrophin signaling and the proliferation of neural progenitors in the adult dentate gyrus, together with increased oxidative stress. Levels of the endocannabinoid anandamide (AEA) seem to affect these depression-by-stress-related features and could be modulated by fatty acid amide hydrolase (FAAH). We aimed to

Ursodeoxycholic acid enriches intestinal bile salt hydrolase-expressing Bacteroidetes in cholestatic pregnancy.

Ursodeoxycholic acid (UDCA) treatment can reduce itch and lower endogenous serum bile acids in intrahepatic cholestasis of pregnancy (ICP). We sought to determine how it could influence the gut environment in ICP to alter enterohepatic signalling. The gut microbiota and bile acid content were determined in faeces from 35 pregnant women (14 with uncomplicated pregnancies and 21 with ICP, 17 receiving UDCA). Faecal bile salt hydrolase activity was measured using a precipitation assay. Serum fibroblast growth factor 19 (FGF19) and 7α-hydroxy-4-cholesten-3-one (C4) concentrations were measured fol

Fatty Acid Amide Hydrolase Binding in Brain of Cannabis Users: Imaging With the Novel Radiotracer [11C]CURB.

BACKGROUND: One of the major mechanisms for terminating the actions of the endocannabinoid anandamide is hydrolysis by fatty acid amide hydrolase (FAAH), and inhibitors of the enzyme were suggested as potential treatment for human cannabis dependence. However, the status of brain FAAH in cannabis use disorder is unknown. METHODS: Brain FAAH binding was measured with positron emission tomography and [11C]CURB in 22 healthy control subjects and ten chronic cannabis users during early abstinence. The FAAH genetic polymorphism (rs324420) and blood, urine, and hair levels of cannabinoids and metab

Reexamination of aspartoacylase: is this human enzyme really a glycoprotein?

Aspartoacylase catalyzes the metabolism of an important amino acid in the brain, with the release acetate serving as the source for fatty acid biosynthesis. Defects in this enzyme lead to a loss of activity and the symptoms of a fatal neurological disorder called Canavan disease. Extensive evidence, including deglycosylation studies, differential activity upon eukaryotic host expression and site directed mutagenesis, have supported the presence of a glycan that plays an essential role in the stability and catalytic activity of mammalian aspartoacylase. However, the structure of this enzyme did

Inhibition of peripheral FAAH depresses activities of bladder mechanosensitive nerve fibers of the rat.

PURPOSE: FAAH degrades endocannabinoids and fatty acid amides. FAAH inhibition reduces micturition frequency and counteracts bladder overactivity in rats. We studied the effects of the peripherally active selective FAAH inhibitor URB937, and the CB1 and CB2 receptor antagonists rimonabant and SR144528, respectively, on single unit afferent activity of primary bladder afferents in rats. MATERIALS AND METHODS: Female Sprague Dawley® rats were anesthetized. Single unit afferent activity of Aδ or C-fibers from the L6 dorsal roots was recorded during bladder filling before and after URB937 adminis
