Human exposure to pet prescription medications.

Veterinarians commonly prescribe medications to treat a variety of companion animal clinical conditions. Many pet medications are pharmacologically the same product prescribed for human use. We report 2 cases in which pet medications were used for self poisoning. Obtaining a pet history may provide critical information in the assessment of a patient with a suspected overdose.

Potent contact allergen in the rubber plant guayule (Parthenium argentatum).

Chemical and dermatotoxicological investigations of the natural and processed resin of the Mexican rubber plant, guayule (Parthenium argentatum), has established the presence of a sesquiterpene cinnamic acid ester (guayulin A) that is a potent elicitor of allergic contact dermatitis in experimental animals. The guayule contact allergen is comparable to the poison ivy skin allergens as an elicitor of dermatitis in sensitized guinea pigs.

Construction of a nontoxigenic Clostridium botulinum strain for food challenge studies.

Clostridium botulinum produces the most poisonous natural toxin known and is a perennial concern to the food industry and to regulatory agencies due to the potential threat of food-borne botulism. To ensure the botulinal safety of foods, rigorous food challenge testing to validate food-processing conditions and food formulations has been routinely performed. Detection of the botulinum neurotoxin is performed by using a mouse bioassay and/or in vitro assays. There has been considerable interest by the food industry and regulatory agencies in minimizing or even replacing the use of animals in th

Antagonism of barbiturate by DNA pyrimidines and allied compounds.

The mortality caused by barbiturate poisoning in mice is reduced by the combined action of the DNA pyrimidines thymine and 5-hydroxymethylcytosine (5-HMC) and the structurally related vitamins, pyridoxal, nicotinamide and thiamine, given intraperitoneally. The acute hypnotic effect of sodium pentobarbital at moderate to high dosages is also antagonized by these compounds. All five compounds are required for activity. The structural similiarity between these antagonist components and the barbiturates had suggested that the former's reactions as cofactors or as the constituents of more complex m

Effects of acute carbon monoxide poisoning on liver damage and comparisons of related oxygen therapies in a rat model.

Acute carbon monoxide (CO) poisoning may cause liver damage and liver dysfunction. Therefore, in this study, we aimed to compare the efficiency of normobaric oxygen (NBO) and high-flow nasal cannula oxygen (HFNCO) treatments on liver injury. For that purpose, 28 male Wistar albino rats were divided into four groups (Control, CO, CO + NBO, and CO + HFNCO). The control group was allowed to breath room air for 30 min. Acute CO poisoning in CO, CO + NBO, CO + HFNCO was induced by CO exposure for 30 min. Thereafter, NBO group received 100% NBO with reservoir mask for 30 min. HFNCO group received hi

Efficacy of the antinicotinic compound MB327 against soman poisoning - Importance of experimental end point.

Medical countermeasures for acute poisoning by organophosphorus nerve agents are generally assessed over 24h following poisoning and a single administration of treatment. At 24h, the antinicotinic bispyridinium compound MB327 (1,10-(propane-1,3-diyl)bis(4-tert-butylpyridinium)) dimethanesulfonate is as effective as the oxime HI-6 against poisoning by soman, when used as part of a treatment containing atropine and avizafone. In this study, we hypothesised that an earlier endpoint, at 6h, would be more appropriate for the pharmacokinetics and mechanism of action of MB327 and would therefore resu

[The pathogenesis, clinical picture and treatment of poisonings by organic solvents, derivatives of chlorinated hydrocarbons (trichloroethylene, carbon tetrachloride)].

Clinical studies and experiments on laboratory animals covered mechanism of trichloroethylene toxicity. The chemical and its metabolites cause nonspecific toxic effects in membranes and therefore induce energy metabolism disorder that is proved to be a trigger of pathologic process in the intoxication. Experimental studies failed to disclose and explain mechanism of compromised calcium metabolism and its role in cardiac manifestations seen in trichloroethylene poisoning.

Morbity and mortality in Texel sheep following feed-based monensin overdose in Brazil.

Monensin is an antibiotic ionophore used as feed additive in production animals, however, overdose with monensin is associated with poisoning in livestock. This study describes a mortality outbreak in sheep associated with ingestion of a feed supplement containing elevated concentrations of monensin. The objectives were characterize pathological lesions from monensin toxicosis, correlate these findings with serum creatine kinase myocardial band (CKMB) to estimate the extent of myocardial injury, as well as with electrocardiogram to evaluate the progression and severity of cardiac damage. An experimental study of the effects of lead acetate on hearing. Cochlear microphonics and action potential of the guinea pig.

Guinea pigs were poisoned with repeated i.p. injections of 1% lead acetate. After 5 weeks, the animals were examined electrophysiologically by using cochlear microphonics (CM) and action potential (AP). The thresholds of maximum voltage of N1 in the AP of the animals injected with a total of 100 mg lead acetate were elevated about 15 dB and increased N1 latency was also observed. However, no significant changes in those of CM were found. The results suggest that lead acetate not only induces damage to the peripheral nerves, but also to the cranial nerves.

Activated Charcoal Haemoperfusion in the Treatment of Experimental Amitriptyline Poisoning in Pigs - The Effect on Amitriptyline Plasma Concentration and Haemodynamic Parameters.

Coated activated charcoal haemoperfusion (CAC-HP) is a well-known treatment modality. Case reports have revealed conflicting results about the efficacy of CAC-HP in the treatment of amitriptyline (AT) poisoning, and no randomized clinical trials have been identified in the literature. This study aimed at quantifying the efficacy of modern CAC-HP as an adjunctive treatment of AT intoxication compared with standard care alone. Fourteen female Danish landrace pigs were randomized to either standard care or standard care plus 4 hr of CAC-HP. The pigs were anaesthetized, and vital parameters were c

Sodium Nitrite and Sodium Thiosulfate Are Effective Against Acute Cyanide Poisoning When Administered by Intramuscular Injection.

STUDY OBJECTIVE: The 2 antidotes for acute cyanide poisoning in the United States must be administered by intravenous injection. In the out-of-hospital setting, intravenous injection is not practical, particularly for mass casualties, and intramuscular injection would be preferred. The purpose of this study is to determine whether sodium nitrite and sodium thiosulfate are effective cyanide antidotes when administered by intramuscular injection. METHODS: We used a randomized, nonblinded, parallel-group study design in 3 mammalian models: cyanide gas inhalation in mice, with treatment postexpos

Should phenytoin or barbiturates be used as second-line anticonvulsant therapy for toxicological seizures?

INTRODUCTION: Seizures are a common sequela of self-poisoning. However, their mechanism differs from seizures of other etiologies. Toxicological seizures result from alterations in the excitatory and inhibitory balance of otherwise normal neurons. In contrast, idiopathic or trauma related seizures usually start with a focus of abnormal neurons. For both forms of seizures, benzodiazepines are recommended as first-line therapy; however, there is debate about the use of phenytoin or barbiturates for second-line therapy. METHODS AND RESULTS: In this article, we systematically review the evidence

[Toxicology of acetonitrile].

Acetonitrile is a high-polarity organic solvent widely used in various chemical industries and laboratories. It was once used in consumer goods such as cosmetics. Acetonitrile is readily absorbed through the skin, by inhalation and by ingestion, and acute poisoning and even fatal effects are possible via these routes. The oral LD50 of acetonitrile in mice, which are one of the most susceptible animals to acetonitrile, is 170-520 mg/kg, and LC50 is about 2,700 ppm after one hour of inhalation. The toxic effects of acetonitrile are attributable to the metabolic release of cyanide, but the sympto

Microbial food borne pathogens. Salmonella.

All food animals are susceptible to infection with Salmonella, a genus of gram negative, nonspore-forming, usually motile, facultative anaerobic bacilli belonging to the family Enterobacteriaceae. Salmonella are differentiated into over 2200 serologically distinct types (serotypes) based on differences in somatic, flagellar, and capsular antigens. Infection with Salmonella may or may not lead to a sometimes fatal salmonellosis, a disease that can remain localized in the gastrointestinal tract as gastro-enteritis, or become generalized as a septicemia and affect several organ systems. Infected

Discovery of Protein-Derived Candidate Anticancer Peptides from Toad Poison (ChanSu) Using an Integrated Proteomics and Bioinformatics-Guided Strategy.

Toad poison (ChanSu), a traditional animal-derived medicine, has long been used in East Asian medical systems for the treatment of inflammatory conditions and tumor-related disorders. While bufadienolides have been extensively investigated as its major bioactive constituents, the contribution of peptide components to its antitumor effects remains largely unexplored. Here, we identified protein-derived candidate antiproliferative peptides from toad poison using an integrated proteomics and bioinformatics-guided strategy. Proteomic analysis identified 135 proteins, from which 2117 peptide sequen

The combination of cobinamide and sulfanegen is highly effective in mouse models of cyanide poisoning.

CONTEXT: Cyanide is a component of smoke in residential and industrial fires, and accidental exposure to cyanide occurs in a variety of industries. Moreover, cyanide has the potential to be used by terrorists, particularly in a closed space such as an airport or train station. Current therapies for cyanide poisoning must be given by intravenous administration, limiting their use in treating mass casualties. OBJECTIVE: We are developing two new cyanide antidotes--cobinamide, a vitamin B(12) analog, and sulfanegen, a 3-mercaptopyruvate prodrug. Both drugs can be given by intramuscular administr

Copper Poisoning, a Deadly Hazard for Sheep.

Copper (Cu) is an essential microelement for animals. However, sheep are particularly susceptible to Cu intoxication, a deadly disease reported worldwide. The risk of developing this poisoning is higher in vulnerable breeds and in intensively managed lambs or milk sheep. Two types of Cu intoxication can occur depending on the chronic or acute exposure to Cu. In chronic Cu poisoning (CCP), the most common form, Cu is accumulated in the liver during a subclinical period. A low intake of Cu antagonists (molybdenum, sulphur, iron, or zinc) favours Cu accumulation. The sudden release of Cu into the

[Theriac: medicine and antidote].

Theriac was an ancient multi-ingredient preparation; originating as a cure for the bites of serpents, mad dogs and wild beasts, it later became an antidote to all known poisons. The name theriac (treacle), (Greek theriake, Latin theriaca, French thériaque) was derived from the Greek for wild beast - theriakos. The first formula was created by Mithridates Vl, King of Pontus, a skillful ruler but a monster of cruelty, who, living in such a fear of being poisoned, took a great interest in toxicology. In the 1st century AD, Nero's personal physician Andromachus improved the formula of Antidotum Mi
