Astroimmunology: the effects of spaceflight and its associated stressors on the immune system.

As humans embark on longer and deeper missions into space, it is crucial to understand how spaceflight impacts the immune system. Decades of discoveries, bolstered by recent multiomic analyses, have identified key immune processes that are affected by the spaceflight environment. These findings form the foundations of the emerging field of 'astroimmunology'. Spaceflight stressors - such as microgravity and galactic cosmic radiation - and other mission-associated variables, including psychological stress and abnormal circadian rhythms, can disrupt or adversely affect immune cell biology. Structural damage to the rat eye following long-term simulated weightlessness.

To better perform space missions and develop human spaceflights, the eye health of astronauts is receiving increasing attention from researchers. In this study, we used prolonged tail suspension to simulate microgravity cephalad fluid shift in space to observe intraocular pressure (IOP) changes, retinal structure, and optic nerve damage in rats. We observed significant choroidal thickening and optic nerve demyelination lesions in the rats in each experimental group. At the cellular level, retinal ganglion cells (RGCs) survival was significantly reduced, optic nerve oligodendrocytes were reduce…

Russian Countermeasure Systems for Adverse Effects of Microgravity on Long-Duration ISS Flights.

The system of countermeasures for the adverse effects of microgravity developed in the USSR supported the successful implementation of long-duration spaceflight (LDS) programs on the Salyut and Mir orbital stations and was subsequently adapted for flights on the International Space Station (ISS). From 2000 through 2010, crews completed 26 ISS flight increments ranging in duration from 140 to 216 d, with the participation of 27 Russian cosmonauts. These flights have made it possible to more precisely determine a crew-member's level of conditioning, better assess the advantages and disadvantages

Carcinogens in spacecraft air.

The health effects of long-term exposure to ionizing radiation during spaceflight are a major concern to NASA, especially for missions beyond low-Earth orbit. Experiments involving astronauts will be conducted during the next few years to improve the risk assessment for exposures to ionizing radiation during flights; however, concomitant exposure to certain airborne chemical carcinogens during these experiments could confound the results of radiation experiments. Carcinogens can reach the spacecraft atmosphere from leaking thrusters, from off-gassing materials, from chemical experiments and fr…

Optic nerve cerebrospinal fluid drainage via lymphatic vessels in its sheath.

Cerebrospinal fluid (CSF) surrounding the optic nerve is essential for nutrient transport, waste removal, and optic nerve function. Optic disc edema in astronauts during long-duration spaceflight-Spaceflight Associated Neuro-Ocular Syndrome (SANS)-may result from impaired optic nerve CSF drainage, but the drainage pathway remains unclear. Given that the optic nerve sheath is an extension of the cranial meninges, we hypothesized that its CSF drains via lymphatic vessels rather than refluxing into the cranium. We assessed the pressure relationship between intracranial and optic nerve CSF and tra…

Blinded by the depths: A case of acute loss of vision in a scuba diver.

Decompression illness (DCI) is a well-known medical emergency among scuba divers, aviators, astronauts, and compressed-air workers. It includes two diseases: decompression sickness (DCS) and arterial gas embolism (AGE). Vision changes are one of the symptoms that can happen in patients with DCI, but acute complete loss of vision is extremely rare. We report a 28-year-old male patient, a novice scuba diver who presented to our hospital with complete loss of vision while he was ascending from 18 m depth. The diagnosis of AGE was made depending on the history, clinical examination, and radiologic…

Neuropsychological considerations for long-duration deep spaceflight.

The deep space environment far beyond low-Earth orbit (LEO) introduces multiple and simultaneous risks for the functioning and health of the central nervous system (CNS), which may impair astronauts' performance and wellbeing. As future deep space missions to Mars, moons, or asteroids will also exceed current LEO stay durations and are estimated to require up to 3 years, we review recent evidence with contemporary and historic spaceflight case studies addressing implications for long-duration missions. To highlight the need for specific further investigations, we provide neuropsychological con…

Platelets in Wound Healing: What Happens in Space?

Beyond their fundamental role in hemostasis, platelets importantly contribute to other processes aimed at maintaining homeostasis. Indeed, platelets are a natural source of growth factors and also release many other substances-such as fibronectin, vitronectin, sphingosine 1-phosphate-that are important in maintaining healthy tissues, and ensuring regeneration and repair. Despite rare thrombotic events have been documented in astronauts, some in vivo and in vitro studies demonstrate that microgravity affects platelet's number and function, thus increasing the risk of hemorrhages and contributin…

Physiological, pharmacokinetic, and pharmacodynamic changes in space.

Medications have been taken since the first Mercury flight in 1967 and, since then, have been used for several indications such as space motion sickness, sleeplessness, headache, nausea, vomiting, back pain, and congestion. As the duration of space missions get longer, it is even more likely that astronauts will encounter some of the acute illnesses that are frequently seen on Earth. Microgravity environment induces several physiological changes in the human body. These changes include cardiovascular degeneration, bone decalcification, decreased plasma volume, blood flow, lymphocyte and eosino…

Dietary instead of pharmacological management to counter the adverse effects of physiological adaptations to space flight.

The development of pharmacological counter-measures to the adverse effects of physiological adaptation to microgravity has received relative … moral defenses against orthostatic hypotension. It is proposed that these simple alterations in the astronauts' diets may be easily implemented to lessen the adverse effects of physiological adaptations to space flight, and to enhance or even replace prescribed pharmacological counter-measures.

Management of corneal injuries in spaceflight and recommendations for planetary missions.

In February 1968, NASA purchased 400 antigravity ballpoint pens from the Fisher Pen Company for the Apollo Program to prevent potential harm to astronauts and equipment. Mechanical pencils previously used in microgravity posed risks like eye injuries from floating fragments penetrating the cornea. The cornea is vulnerable to abrasions, perforations, and chemical burns in such environments, affecting crewmembers aboard the International Space Station (ISS). While they undergo extensive training for emergency situations, there are inherent complexities when faced with eye injuries. In this chall…

Recording of blood pressure, heart rate and aortic nerve activity during parabolic flight in the rat via radio-telemetry.

Exposure to microgravity induces cardiovascular deconditioning characterized by orthostatic hypotension when astronauts return to the earth. In order to understand the mechanism of cardiovascular deconditioning, it is necessary to clarify the changes in hemodynamics and the cardiovascular regulation system over the period of space flight. The telemetry system applied to freely moving animals will be a useful and appropriate technique for this kind of long term study of the cardiovascular system in the conscious animal during space flight. The purpose of the present study is twofold: firstly, t…

A predictive test for space motion sickness.

Eye torsion was examined in 13 astronaut subjects, tested during repeated episodes of 0 G and 1.8 G in parabolic flight aboard NASA's KC-135. Four findings are included. 1) A strong relationship between previous history of SMS and ocular torsional disconjugacy in novel gravitational states. 2) Responses were unchanged in 4 subjects retested a year later. 3) Ocular torsional disconjugacy scores increased as exposure to 0 and 1.8 G increased. This was particularly evident in subjects who had had SMS. 4) Torsional studies during 10 to 20 parabolas are required to accurately predict SMS. The hypot

Unilateral centrifugation of the otoliths as a new method to determine bilateral asymmetries of the otolith apparatus in man.

Ten healthy subjects were eccentrically rotated with constant speed on a Barany chair. Setting of a luminous line (LL) to the subjective vertical and ocular counter-roll (OCR) were evaluated. During eccentric position rotation subjects consistently reported illusory rotation and set the LL to an angle correlating to centrifugal force. At the same time an OCR of opposite direction was measured. In one patient, labyrinthectomized on the right side, only counterclockwise rotation of the luminous line was observed. Differences between "inner" and "outer" eye were evident for luminous line settings

Hypertension and orthostatic hypotension in applicants for spaceflight training and spacecrews: a review of medical standards.

The inauguration of NASA of the position of Payload Specialists for SHUTTLE-SPACELAB flights has broken the tradition of restrictive medical physical standards in several ways: by reducing physical requirements and extensive training; by permitting the selection of older individuals and women; by selecting individuals who may fly only one or several missions and do not spend an entire career in space activities. Experience with Payload Specialists to be gained during the forthcoming SPACELAB missions, observing man in spaceflight step by step on an incremental basis, will provide valuable data

Motion sickness: part I--a theory.

Early reports on space exploration suggested that cosmonauts and astronauts sustained "motion sickness" symptoms described as "dizziness, nausea, vomiting, flashes of light, formed hallucinations or illusions of inversion of image in space." Hallucinations may be due to many causes but most of the above symptoms were similar to those experienced by some patients with expanding intracranial lesions whose symptomatology was referable to the temporoparieto-occipital cortex of the brain. On the basis of our observations, it is suggested that the term "motion sickness" might be applied to earthly s…

Molecular Mechanism of Microgravity-Induced Intestinal Flora Dysbiosis on the Abnormalities of Liver and Brain Metabolism.

Space flight has many adverse effects on the physiological functions of astronauts. Certain similarities have been observed in some physiological processes of rodents and astronauts in space, although there are also differences. These similarities make rodents helpful models for initial investigations into space-induced physiological changes. This study uses a 3D-Clinostat to simulate microgravity and explores the role of microgravity in space flight-induced liver and brain abnormalities by comparing changes in the gut microbiota, serum metabolites, and the function and physiological biochemis…

Promethazine Effects on Motion Sickness During Altered Gravity Induced by Parabolic Flight.

INTRODUCTION: Astronauts commonly experience space motion sickness, which can impair astronaut performance and safety. Pharmaceuticals are frequently used to reduce space motion sickness symptoms but have not been extensively studied in partial gravity. In this research effort, we investigated the impact of oral promethazine on motion sickness during parabolic flight. METHODS: We collected motion sickness scores from 12 subjects (6 women) during parabolic flight. Each subject participated in two flights-one 0-G flight and one partial g flight-experiencing 10 parabolas at 0 g, 0.25 g, 0.50 g, …
