Desiccation tolerance as a driver of environmental persistence and transmission in nosocomial bacterial pathogens.

Hospital-acquired infections (HAIs) represent a major global health burden, leading to increased patient morbidity, mortality, and healthcare costs. These infections frequently occur following patient contact with microbial pathogens persisting on indwelling devices and other abiotic surfaces in the hospital environment. A key driver of this persistence is the ability to survive the total loss of water, a phenomenon known as desiccation tolerance. While desiccation tolerance has been studied across various kingdoms of life, comparatively little is known about the molecular mechanisms that enab

Ocular streptococcal infections: A clinical and microbiological review.

Streptococcus is a diverse bacterial genus that is part of the ocular surface microbiome implicated in conjunctivitis, keratitis, endophthalmitis, dacryocystitis, and orbital cellulitis that can lead to decreased visual acuity and require surgical intervention. The pathophysiology of S. pneumoniae is well-established and the role of the polysaccharide capsule, pneumolysin, neuraminidases, and zinc metalloproteinases in ocular infections described. Additionally, key virulence factors of the viridans group streptococci such as cytolysins and proteases have been outlined, but there is a paucity o

Capsule null locus meningococci: typing of antigens used in an investigational multicomponent meningococcus serogroup B vaccine.

The investigational multicomponent meningococcus serogroup B vaccine (4CMenB) targets the antigenetically variable population of serogroup B meningococci. Forty-one strains of capsule null locus (cnl) meningococci, which are frequent among healthy carriers, were selected from nine sequence types (ST), which belong to four clonal complexes (cc), and three countries. They were antigen sequence typed and analyzed for antigen expression to predict whether these strains harbor the genes and express the four vaccine antigens of 4CMenB as measured by the meningococcal antigen typing system (MATS). Influence of modified natural or synthetic surfactant preparations on growth of bacteria causing infections in the neonatal period.

Connatal bacterial pneumonia is common in neonates. Animal studies and initial clinical reports indicate that surfactant dysfunction is involved in the pathophysiology of severe neonatal pneumonia. Since respiratory distress syndrome and connatal pneumonia may be difficult to differentiate in the first hours of life, neonates with respiratory failure due to bacterial infections might receive surfactant. Under such conditions surfactant components might be catabolized by bacteria and promote bacterial growth. We therefore investigated the influence of three modified natural (Curosurf, Alveofact

Impact of Docosahexaenoic Acid Therapy on Subgingival Plaque Microbiota.

BACKGROUND: Oral docosahexaenoic acid (DHA) + aspirin therapy has been shown to reduce periodontal probing depth (PD) and local inflammatory mediators in gingival crevicular fluid (GCF) among patients with untreated chronic periodontal disease. Whether DHA + aspirin therapy influences specific bacterial burden in this setting is unknown. Thus, the aim of this study is to evaluate the impact of DHA with low-dose aspirin therapy on periodontal bacterial profile in patients with periodontitis. METHODS: Fifty-five adults with moderate-to-severe periodontitis were enrolled in a randomized, 3-month

Structural insights into the polytopic Escherichia coli phosphoglycosyl transferase WbaP.

Host-pathogen interactions frequently depend on key components of the bacterial cell surface, such as capsules and lipopolysaccharides in Gram-negative bacteria. The first step in the synthesis of lipid-linked polysaccharides is the substitution of a uridine diphosphate (UDP)-sugar by a lipid monophosphate catalysed by a phosphoglycosyl transferase (PGT). We report the 3.0 Å cryo-electron microscopy apo-structure of the PGT enzyme WbaP from Escherichia coli, a UDP-galactose:undecaprenolphosphate galactose-1-phosphoryl transferase. The structure is a dimer with each monomer formed of four N-ter

Targeted inhibition of pathobiont virulence factor mitigates alcohol-associated liver disease.

Alcohol-associated liver disease poses a global health burden with high mortality. Imbalances in the gut microbiota are important for disease progression. Using metagenomic sequencing of fecal samples from a multicenter, international cohort of patients with alcohol-associated hepatitis, we found that the presence of virulence factor KpsM, encoded in the genome of Escherichia coli (E. coli), correlated with patient mortality. Functional studies using gnotobiotic mouse models and genetic manipulation of bacteria demonstrated that kpsM-positive E. coli exacerbate ethanol-induced liver disease. T

Splenic red pulp macrophages eliminate the liver-resistant Streptococcus pneumoniae from the blood circulation of mice.

Invasive infections by encapsulated bacteria are the major cause of human morbidity and mortality. The liver resident macrophages, Kupffer cells, form the hepatic firewall to clear many encapsulated bacteria in the blood circulation but fail to control certain high-virulence capsule types. Here we report that the spleen is the backup immune organ to clear the liver-resistant serotypes of Streptococcus pneumoniae (pneumococcus), a leading human pathogen. Asplenic mice failed to control the growth of the liver-resistant pneumococci in the blood circulation. Immunologic and genetic analyses ident

The syndrome of post-splenectomy fulminant sepsis. Case report and review of the literature.

Three years after splenectomy, a middle-aged woman had two separate episodes of fulminant bacterial sepsis. She recovered each time with prompt and appropriate treatment. Her immunologic system was examined and found to be normal with respect to antibody formation against diphtheria and tetanus toxoid, granulocyte killing of staphylococci and serum opsonizing activity. Granulocytes, bursa-equivalent (B) and thymus-dependent (T) lymphocytes and serum immunoglobulins were quantitatively normal. She produced antibody against subcutaneously injected polyvalent pneumococcal vaccine. She demonstrate

Complementary Role of CD4+ T Cells in Response to Pneumococcal Polysaccharide Vaccines in Humans.

Bacterial pathogens expressing capsular polysaccharides are common causes of mucosal infections (pneumonia, intestinal), as well as often fatal, invasive infections (meningitis, bloodstream infections) in children and adults worldwide. These chemically simple but structurally complex carbohydrate structures on the bacterial surface confer resistance to recognition and clearance by the immune system through a range of mechanisms. Such recognition of capsular polysaccharides may be reduced by their limited ability to directly stimulate B cells and the T cells that may facilitate these humoral re

A Multinuclear 1H and 31P NMR Approach for Quantification of Pneumococcal Cell-Wall Polysaccharide in Vaccine Development.

Capsular polysaccharides are the principal constituents of S. pneumoniae capsules and major determinants of bacterial virulence. As key antigenic targets, they represent the foundation for pneumococcal vaccine development. Cell wall polysaccharides (CWPSs) commonly persist as a process-related impurity during fermentation and purification and therefore require reliable quantification throughout vaccine manufacturing. In this work, a multinuclear quantitative NMR strategy based on 1H and 31P detection was developed for the determination of CWPSs in purified pneumococcal polysaccharide preparati

Emergence, identification, and characterization of a novel Streptococcus agalactiae CPS type Ia ST7 strain (Ia-2021) causing large scale mortalities in tilapia aquaculture.

In 2021 streptococcosis caused by Streptococcus agalactiae capsular polysaccharide (CPS) type Ia ST7 (referred to as Ia-2021) emerged in Mexico and spread to other regions in Central and South America devastating the industries in the affected countries. To address this emergent issue, the present research aimed to: (1) document case reports of the disease, (2) reproduce clinical signs under laboratory conditions, (3) use comparative genomics to investigate the evolutionary history of the emergent strain, and (4) develop an assay to discriminate from other S. agalactiae. Experimental injection

Effects of Capsular Polysaccharide amount on Pneumococcal-Host interactions.

Among the many oral streptococci, Streptococcus pneumoniae (Spn) stands out for the capacity of encapsulated strains to cause invasive infection. Spread beyond upper airways, however, is a biological dead end for the organism, raising the question of the benefits of expending energy to coat its surface in a thick layer of capsular polysaccharide (CPS). In this study, we compare mutants of two serotypes expressing different amounts of CPS and test these in murine models of colonization, invasion infection and transmission. Our analysis of the effect of CPS amount shows that Spn expresses a caps

Non-typeable pneumococci circulating in Portugal are of cps type NCC2 and have genomic features typical of encapsulated isolates.

BACKGROUND: Pneumococcus is a major human pathogen and the polysaccharide capsule is considered its main virulence factor. Nevertheless, strains lacking a capsule, named non-typeable pneumococcus (NT), are maintained in nature and frequently colonise the human nasopharynx. Interest in these strains, not targeted by any of the currently available pneumococcal vaccines, has been rising as they seem to play an important role in the evolution of the species. Currently, there is a paucity of data regarding this group of pneumococci. Also, questions have been raised on whether they are true pneumoco

[Multiplex amplification test system for the identification and differentiation of Bacillus anthracis].

The multiplex amplification test system for the identification of Bacillus anthracis with primers to plasmid cya (pX01), capC (pX02) genes and chromosomal sap gene were developed. The primers to sap gene were selected by the authors and, after being tested on 72 microbial strains of the genus Bacillus, proposed as more specific in comparison with the known primers to chromosomal locus Ba 813. The proposed test system permitted the simultaneous identification of B. anthracis of all plasmid variants, the evaluation of their potential virulence and the differentiation of B. anthracis nonplasmid s
