Mesoporous bioactive glass nanoparticles doped with magnesium: drug delivery and acellular in vitro bioactivity.

The effects of the magnesium doping of binary glass (Si-Ca) on particle texture, on the biomineralization process in simulated body fluid (SBF) as well as on drug loading and release were examined. For this purpose, magnesium-doped binary bioglass nanoparticles (85SiO2-(15 - x)CaO-xMgO, with x = 1, 3, 5 and 10 mol%) were prepared by a base catalysed sol-gel method. N2 adsorption isotherm analysis showed an enhancement in specific surface area as the Mg molar fraction increased. In addition, the FTIR spectra of the samples after soaking in SBF for various periods of time confirmed the presence …

Inhibition of CXCR4 inhibits the proliferation and osteogenic potential of fibroblasts from ankylosing spondylitis via the Wnt/β‑catenin pathway.

Ankylosing spondylitis (AS) is an autoimmune condition characterized by chronic inflammation and abnormal ossification as the primary featur … viral oncogene homolog, cyclin D1 and osteocalcin. Alizarin red staining demonstrated a decrease in biomineralization activity following the inhibition of CXCR4. These data support the hypothesis that inhibiting CXCR4 in patients with AS may suppress the ossification of fibroblasts.

Shape-preserving transformation of carbonate minerals into lead halide perovskite semiconductors based on ion exchange/insertion reactions.

Biological and bio-inspired mineralization processes yield a variety of three-dimensional structures with relevance for fields such as photonics, electronics and photovoltaics. However, these processes are only compatible with specific material compositions, often carbonate salts, thereby hampering widespread applications. Here we present a strategy to convert a wide range of metal carbonate structures into lead halide perovskite semiconductors with tunable bandgaps, while preserving the 3D shape. First, we introduce lead ions by cation exchange. Second, we use carbonate as a leaving group, fa

Imaging the nanostructure of bone and dentin through small- and wide-angle X-ray scattering.

X-ray scattering is a powerful nondestructive experimental method that is well suited to study biomineralized tissues such as bone. Small-angle X-ray scattering (SAXS) gives information about the size, shape, and predominant orientation of the nanometer-sized mineral particles in the bone. Wide-angle X-ray diffraction (WAXD) allows the characterization of structural parameters, describing size and orientation of the hydroxyapatite crystals. Furthermore, scanning an area with nano- or micrometer-sized X-ray beams allows one to extend this local information to map large bone or dentin sections.

Hierarchical scaffold design for mesenchymal stem cell-based gene therapy of hemophilia B.

Gene therapy for hemophilia B and other hereditary plasma protein deficiencies showed great promise in pre-clinical and early clinical trials. However, safety concerns about in vivo delivery of viral vectors and poor post-transplant survival of ex vivo modified cells remain key hurdles for clinical translation of gene therapy. We here describe a 3D scaffold system based on porous hydroxyapatite-PLGA composites coated with biomineralized collagen 1. When combined with autologous gene-engineered factor IX (hFIX) positive mesenchymal stem cells (MSCs) and implanted in hemophilic mice, these scaff

Solid-state P-31 NMR study of octacalcium phosphate incorporated with succinate.

Octacalcium phosphate (OCP) is an important model compound in the study of biomineralization. The octacalcium phosphate-succinate (OCPS) compound is prepared and characterized by (31)P solid-state NMR spectroscopy. Taking advantage of the fact that the crystal structures of OCP and OCPS are very similar, an NMR strategy based on the (31)P homonuclear double-quantum spectroscopy is developed to assign all the peaks observed in the (31)P magic-angle spinning spectrum of OCPS. On the basis of our experimental data, the molecular formula of OCPS is determined to be Ca(7.81)(HPO(4))(1.82)(PO(4))(3.…

Divisive effect of alcohol-water mixed solvents on growth morphology of calcium carbonate crystals.

Controlling the process of crystal growth is of importance to the biomineralization and materials science. In this work, some novel morphology of calcium carbonate (CaCO3) was precipitated in an ethanol-water binary solvent (EWBS) with a CaCl2/Na2CO3 reaction system. For the solutions of CaCl2/Na2CO3 in EWBS, the alcoholization and hydration of Ca2+ and CO3(2-) were discussed from the radial distribution functions by molecular dynamics simulations, and the number density profiles of water molecules around and approximately 15 A away from CO3(2-) were employed to reveal the distribution of wate…

Nucleation of apatite crystals in vitro by self-assembled dentin matrix protein 1.

Bones and teeth are biocomposites that require controlled mineral deposition during their self-assembly to form tissues with unique mechanical properties. Acidic extracellular matrix proteins play a pivotal role during biomineral formation. However, the mechanisms of protein-mediated mineral initiation are far from understood. Here we report that dentin matrix protein 1 (DMP1), an acidic protein, can nucleate the formation of hydroxyapatite in vitro in a multistep process that begins by DMP1 binding calcium ions and initiating mineral deposition. The nucleated amorphous calcium phosphate preci

Millisecond-time-scale controlled freeze-quench for solute-intermediate analysis by solid-state NMR.

Crystallization is a fundamental process in biomineralization, yet its complexity increases significantly in non-classical pathways of nucleation and growth, where numerous intermediates exist between free ions in solution and the final crystalline phase. Characterization of such intermediates can be delicate, as some processes can proceed very fast, inducing a short temporal window for their characterization. The problem is particularly true for the so-called prenucleation species, for which this difficulty is made worse due to their solubility, nanometric size and highly dynamic nature. In t…

Electrogenic HfO2/TiO2 Living Biointerface Dismantles Metabolic Radioresistance for Amplified Radio-Immunotherapy.

Radiotherapy (RT) efficacy in aggressive solid tumors is hindered by lactate-driven metabolic radioresistance and an immunosuppressive microenvironment. Here, we engineer a living biointerface (SHT) via in situ biomineralization of HfO2/TiO2 onto an electrogenic Shewanella oneidensis chassis to address this multifaceted challenge. This structural integration bridges the bacterial metabolism with interfacial radiocatalysis. The TiO2 shell acts as an efficient electron acceptor to accelerate bacterial extracellular electron transfer, driving rapid intratumoral lactate depletion, while the high-Z…

Polymer-mediated mineralization in hydrogels: design principles, hybrid materials, and network visualization.

Polymer-mediated mineralization in hydrogels offers a versatile route to inorganic-organic hybrids in which mineral formation is coupled to network chemical affinity, precursor transport, spatial confinement, and mechanics. This review first distills the biomineralization concepts most relevant to hydrogel design, including classical and nonclassical nucleation, precursor stabilization, and charge-regulated ion binding. We then organize hydrogel-mineral systems around four experimentally useful variables: chemical affinity, precursor transport, spatial confinement, and network reorganization k…

Photocatalytic degradation of methylene blue by CdS quantum dots biosynthesized by cysteine synthetase TtCsa1 from Tetrahymena thermophila.

H2S is a crucial endogenous gaseous signaling molecule involved in various metabolic pathways and is associated with the biological response to heavy metal stress. The biomineralization of CdS serves as a critical mechanism for responding to cadmium stress and removing Cd2+ in different organisms. CdS is also a widely utilized semiconductor material for the photocatalytic degradation of dyes. In this study, we found that cysteine synthetase 1 (TtCsa1) is involved in CdS formation under cadmium stress in Tetrahymena thermophila. TtCsa1 also catalyzed CdS formation in vitro, and the synthesized …

Adult-onset hypophosphatasia diagnosed after consecutive tooth loss during orthodontic treatment: a case report.

BACKGROUND: Adult hypophosphatasia is an uncommon inherited disorder of mineral homeostasis affecting bone. It arises from mutations within the Alkaline Phosphatase, Biomineralization Associated (ALPL) gene, which encodes tissue-nonspecific alkaline phosphatase. Because of its low prevalence and non-specific clinical manifestations, underdiagnosis and misdiagnosis are frequent, particularly in Asian populations. CASE PRESENTATION: We present a case of a 38-year-old Japanese male diagnosed with adult hypophosphatasia following consecutive tooth loss during orthodontic treatment. Genetic analys…
