A Statistical Review: Why Average Weighted Accuracy, not Accuracy or AUC?

Sensitivity and specificity are key aspects in evaluating the performance of diagnostic tests. Accuracy and AUC are commonly used composite measures that incorporate sensitivity and specificity. Average Weighted Accuracy (AWA) is motivated by the need for a statistical measure of diagnostic yield that can be used to compare diagnostic tests from the medical costs and clinical impact point of view, while incorporating the relevant prevalence range of the disease as well as the relative importance of false positive versus false negative cases. We derive the variance/covariance estimators and tes

Global Odds Model with Proportional Odds and Trend Odds Applied to Gross and Microscopic Brain Infarcts.

Medical and epidemiological researchers commonly study ordinal measures of symptoms or pathology. Some of these studies involve two correlated ordinal measures. There is often an interest in including both measures in the modeling. It is common to see analyses that consider one of the measures as a predictor in the model for the other measure as outcome. There are, however, issues with these analyses including biased estimate of the probabilities and a decreased power due to multicollinearity (since they share some predictors). These issues create a necessity to examine both variables as simul

Sentinel-lymph-node resection compared with conventional axillary-lymph-node dissection in clinically node-negative patients with breast cancer: overall survival findings from the NSABP B-32 randomised phase 3 trial.

BACKGROUND: Sentinel-lymph-node (SLN) surgery was designed to minimise the side-effects of lymph-node surgery but still offer outcomes equivalent to axillary-lymph-node dissection (ALND). The aims of National Surgical Adjuvant Breast and Bowel Project (NSABP) trial B-32 were to establish whether SLN resection in patients with breast cancer achieves the same survival and regional control as ALND, but with fewer side-effects. METHODS: NSABP B-32 was a randomised controlled phase 3 trial done at 80 centres in Canada and the USA between May 1, 1999, and Feb 29, 2004. Women with invasive breast ca

Annual and seasonal variation of tick-borne encephalitis virus (TBEV) prevalence in ticks in selected hot spot areas in Germany using a nRT-PCR: results from 1997 and 1998.

The prevalence of tick-borne encephalitis virus (TBEV) in Ixodes ricinus tick populations in endemic areas of Germany with the highest TBE risk is unknown. Annual and seasonal differences in TBEV prevalence have also not been studied. Against this background, in May 1997 we started a systematic virus surveillance programme in ticks collected in locations known to have a high incidence of autochthonous TBE cases. These were 5 locations in Baden-Württemberg (Black Forest) and 8 locations in Bavaria (surrounding Passau). Field-collected ticks were randomly assigned to pools of 10 adults or 20 nym

A longitudinal Bayesian framework for estimating causal dose-response relationships.

Existing causal methods for time-varying exposure and time-varying confounding focus on estimating the average causal effect of a time-varying binary treatment on an end-of-study outcome, offering limited tools for characterizing marginal causal dose-response relationships under continuous exposures. We propose a scalable, nonparametric Bayesian framework for estimating marginal longitudinal causal dose-response functions with repeated outcome measurements. Our approach targets the average potential outcome at any fixed dose level and accommodates time-varying confounding through the generaliz

Best subset selection meets resampling: tackling selection instability at very low numbers of events per variable.

Trust is a basic necessity for translating machine learning models from bench to bedside. To foster clinicians' trust in predictive models, these models should be interpretable or explainable. A major prerequisite for interpretability or explainability is that feature subsets are stable, i.e., that predictors returned by feature or best subset selection approaches do not change dramatically when the data is perturbed only slightly. However, prior research has shown that stable selections are usually not possible when the number of labels in the smaller class per feature, or outcome events per

Estimating relapse time distribution from longitudinal biomarker trajectories using iterative regression and continuous time Markov processes.

Biomarker measurements obtained by blood sampling are often used as a non-invasive means of monitoring tumour progression in cancer patients. Diseases evolve dynamically over time, and studying longitudinal observations of specific biomarkers can help to understand patients response to treatment and predict disease progression. We propose a novel iterative regression-based method to estimate changes in patients status within a cohort that includes censored patients, and illustrate it on clinical data from myeloma cases. We formulate the relapse time estimation problem in the framework of Piece

Scalable inference for spatial generalized mixed models for use in multiplexed imaging analysis.

Understanding how cells interact to coordinate tissue function and respond to disease remains a central challenge in biology. Recently developed multiplex bioimaging technologies [multiplex immunofluorescence (MxIF)] can quantify multiple protein markers at a single-cell resolution in spatially preserved tissue, offering the opportunity to understand how cellular organization relates to disease mechanisms and treatment response. Spatial generalized linear mixed models for cell co-localization offer a flexible framework for these analyses, accommodating cell-, image-, or subject-level predictor

A monotone single-index modal regression powered by deep neural networks for non-Gaussian periodontal data.

Pocket depth (PD) is a widely used biomarker for diagnosing risk of periodontal disease (PrD). However, PD typically exhibits skewness and heavy-tailedness, and its relationship with clinical risk factors is often nonlinear. Motivated by PrD studies, this paper develops a robust single-index modal regression framework for analyzing skewed and heavy-tailed data. Our method has the following novel features: (i) a flexible 2-piece scale Student-$ \boldsymbol{t} $ error distribution that generalizes both normal and 2-piece scale normal distributions; (ii) a neural network with guaranteed monotonic

Dynamic case-control sampling for rapid estimation of vaccine effectiveness against an emerging infectious disease variant.

New SARS-CoV-2 variants arise frequently with different viral properties that can impact the effectiveness of the vaccines. Updating estimates of vaccine effectiveness (VE) in public health surveillance can be limited by the necessity of conducting a distinct study that entails analysis of prospective cohort data or using a test-negative design. We introduce a method for dynamically updating estimates of VE using data that accumulate in real time. Our method uses dynamic case-control sampling to estimate VE against a newly emerging variant relative to a previous variant. Dynamic case-control s

Statistical method for pooling categorical biomarker data from multi-center matched/nested case-control studies.

Pooled analyses that aggregate data from multiple studies are becoming increasingly common in collaborative epidemiologic research to increase sample size and population diversity. Many biomarkers, such as vitamin D, are routinely analyzed using clinically defined categories rather than their original continuous scales, yet statistical methods for pooling biomarker data across studies have largely focused on continuous biomarkers. Biomarker measurements from different studies are subject to systematic measurement errors and directly pooling them for analyses may lead to biased estimates of the

Multivariate functional linear discriminant analysis with feature selection: an application to inflammatory bowel disease classification.

Inflammatory Bowel Disease (IBD), including Crohn's Disease (CD) and Ulcerative Colitis (UC), presents significant public health challenges due to its complex etiology. Motivated by the IBD study of the Integrative Human Microbiome Project, our objective is to identify microbial pathways that distinguish between CD, UC, and non-IBD over time. Most current research relies on simplistic analyses that examine 1 variable or time point at a time, or address binary classification problems, limiting our understanding of the dynamic interactions within the microbiome over time. To address these limita

Addressing the influence of unmeasured confounding in observational studies with time-to-event outcomes: a semiparametric sensitivity analysis approach.

In this paper, we develop a semiparametric sensitivity analysis approach designed to address unmeasured confounding in observational studies with time-to-event outcomes. We target estimation of the marginal distributions of potential outcomes under competing exposures using influence function-based techniques. We derive the non-parametric influence function for uncensored data and map the uncensored data influence function to the observed data influence function. Our methodology is motivated by and applied to an observational study evaluating the effectiveness of radical prostatectomy (RP) ver
