Taxonomic and functional analysis of type I sourdough microbiota after long-term cryopreservation and subsequent reactivation.

Type I sourdough microbiota may be subjected to taxonomic and functional shifts during traditional maintenance by continuous backslopping. This proof-of-concept study evaluated a standardized workflow for long-term cryopreservation (-80 °C for 12 months) and recovery of three artisanal sourdough microbiota differing for their starting microbial compositions (Pediococcus/Fructilactobacillus and Saccharomyces/Kazachstania as dominant taxa). Microbiota stability was assessed through culture-dependent analyses on both purified microbiota pellets and whole sourdough, and through culture-independent

Enhanced B-type starch granules proportion modulates starch-gluten interactions during the thermal processing of reconstituted doughs.

This work investigated structure-properties changes of reconstituted wheat A/B starch doughs under different ratios during dynamic thermal processing. Results indicated that a change in spatial conformation and aggregation structure of the starch-gluten system was induced with heating (30 °C-86 °C). Moderately increased B starch ratio can effectively fill the gluten network and improve starch-protein interactions, which promotes the free sulfhydryl group oxidation and results in the formation of more glutenin macropolymer; this contributes to a higher degree of cross-linking and stability to t

Detoxification of Wheat Gluten by Enzymatic Transamidation under Reducing Condition and Its Application in Typical Food Model.

SCOPE: Gluten, the primary network builder of wheat dough, is responsible for celiac disease or wheat allergy. Transamidation of gluten unde … apparent structure, digestive level, and rheological characteristics of resultant dough and steamed bread, as a typical food model. The results show that transamidation starts at the kneading stage, as evaluated by using R5 enzyme-linked immunoassay and rat basophils. The potential toxicity of celiac disease is reduced by about 83% when 1% microbial tra …

Studies on gliadin related peptides. I. Synthesis, purification and 1H n.m.r. characterization of the pentapeptide H-Tyr-(Gln)3-Pro-OH.

The pentapeptide H-Tyr-(Gln)3-Pro-OH has been recently postulated to be the basic repetitive unit of a sequential polypeptide contained in wheat bread alpha-gliadins, which are believed to be toxic factors in coeliac disease, gluten-dependent enteropathy. Solid-phase synthesis, purification and 1H n.m.r. characterization in water solution of this peptide are described.

A Bioinformatic Workflow for InDel Analysis in the Wheat Multi-Copy α-Gliadin Gene Family Engineered with CRISPR/Cas9.

The α-gliadins of wheat, along with other gluten components, are responsible for bread viscoelastic properties. However, they are also related to human pathologies as celiac disease or non-celiac wheat sensitivity. CRISPR/Cas was successfully used to knockout α-gliadin genes in bread and durum wheat, therefore, obtaining low gluten wheat lines. Nevertheless, the mutation analysis of these genes is complex as they present multiple and high homology copies arranged in tandem in A, B, and D subgenomes. In this work, we present a bioinformatic pipeline based on NGS amplicon sequencing for the anal…

Detailed analysis of the expression of an alpha-gliadin promoter and the deposition of alpha-gliadin protein during wheat grain development.

BACKGROUND AND AIMS: Alpha-gliadin proteins are important for the industrial quality of bread wheat flour, but they also contain many epitopes that can trigger celiac (coeliac) disease (CD). The B-genome-encoded alpha-gliadin genes, however, contain very few epitopes. Controlling alpha-gliadin gene expression in wheat requires knowledge on the processes of expression and deposition of alpha-gliadin protein during wheat grain development. METHODS: A 592-bp fragment of the promotor of a B-genome-encoded alpha-gliadin gene driving the expression of a GUS reporter gene was transformed into wheat.…

Presence, Tissue Localization, and Gene Expression of the Adiponectin Receptor 1 in Testis and Accessory Glands of Male Rams during the Non-Breeding Season.

Adiponectin (ADIPOQ) is a member adipocytokines, and its actions are supported by two receptors, ADIPOQ receptor 1 and -2, respectively (ADIPOR1 and -R2). Our study was performed to evaluate the ADIPOR1 presence and location and its gene expression in reproductive tissues of the male ram, during its non-breading season. The different portions of the male ram reproductive system (testis, epididymis, seminal vesicle, ampoule vas deferens, bulb-urethral gland) were collected in a slaughterhouse. Immunohistochemistry showed ADIPOR1 positive signals in the cytoplasm of all the glandular epithelial …

Alleviating and exacerbating foods in hidradenitis suppurativa.

While dietary triggers have been investigated in acne and other inflammatory follicular dermatoses, there is a paucity of data on diet and hidradenitis suppurativa (HS). We sought to identify exacerbating and alleviating foods in HS patients. An anonymous survey was distributed via HS Facebook support groups and in person at HS specialty clinics. Participants were asked to select all that apply from a list to indicate foods that worsen and make HS better including sweet foods, breads and pasta, red meat, chicken, fish, canned foods, fruits, vegetables, dairy, high-fat foods, I do not know, and…

Genome-wide association study identifies candidate genes for agronomic traits under heat- and combined recurrent drought-and heat-stress in wheat.

Combined heat and drought stress, mainly during sensitive growth stages, considerably limits wheat yields. Herein, we report on an experiment conducted to analyze and predict the phenology, architecture, and kernel yield component (PAKyC) traits of a diverse bread wheat population grown under heat stress and fully irrigated conditions (HSFIC), and combined recurrent drought and heat stress (CRDHS). 17,711 high-quality single-nucleotide polymorphisms (SNPs) were used to perform a GWAS for 13 PAKyC traits on 187 genotypes, a subset of 234 genotypes, using the BLINK model. Nine constitutive SNPs,…

Precision Nutrition in NAFLD: Effects of a High-Fiber Intervention on the Serum Metabolome of NAFD Patients-A Pilot Study.

Non-alcoholic fatty liver disease (NAFLD) is associated with dysfunction of the intestinal microbiota and its metabolites. We aimed to assess whether replacing bread with high-fiber buns beneficially changes the metabolome in NAFLD patients. This study involved 27 adult patients with NAFLD validated by FibroScan® (CAP ≥ 234 dB/m). Patients were asked to replace their existing bread for two meals with high-fiber buns. In this way, the patients ate two rolls every day for 2 months. The following parameters were analysed (at the beginning and after 2 months): the anthropometric data (BIA), eating…

Looking for immunotolerance: a case of allergy to baker's yeast (Saccharomyces cerevisiae).

We describe one case of baker's yeast true allergy in a boy with previously diagnosed mite-allergy and atopic dermatitis. At the age of 6, being atopic dermatitis and rhinitis well controlled by drugs, he began to experience generalized urticaria and asthma after eating pizza and bread, but only fresh from the oven. The diagnostic workup revealed single sensitization to baker's yeast (Saccharomyces cerevisiae), and a severe systemic reaction also occurred during the prick-by-prick procedure. After discussing with parents, no special dietary restriction was suggested but the use of autoinjectab…

Oesophageal manometry during eating in the investigation of patients with chest pain or dysphagia.

Dysphagia is a frequent cause of referral for oesophageal manometry although the motor response to eating is not routinely studied. We examined symptoms and oesophageal motor patterns in response to eating bread in 30 patients with either gastro-oesophageal reflux (n = 20), or normal oesophageal function tests (n = 10). No patient experienced symptoms while swallowing water but one complained of heartburn and one developed symptomatic oesophageal 'spasm' during eating. In eight further patients, pain or dysphagia which occurred with swallowing bread was associated with aperistalsis. Comparing …

Wheat GT-1-like Transcription Factor Boosts Cutin Biosynthesis.

Cutin matrices in the cuticle cover plant epidermis, facilitating plant adaptation to stressful environments. Although cutin biosynthesis is extensively explored in the model plant Arabidopsis thaliana, the molecular mechanism governing cutin biosynthesis in the agriculturally important crop bread wheat (Triticum aestivum L.) remains largely unknown. The aim of the study is the characterization of the function and transcriptional regulation of a wheat gene involved in cutin biosynthesis. Long-chain acyl-CoA synthetase TaLACS2 was identified as an essential component of the wheat cutin biosynth…
