Carcinogenic, mutagenic and teratogenic risks associated with vinyl chloride.

The data presented demonstrate clearly that vinyl chloride (VC) is related to a significant excess of mortality from cancer of the liver, lung and brain among workers occupationally exposed to VC. The risk of dying from cancer of the lymphatic and hematopoietic system also appears to increase with an increase in latency. These cancer sites could have been predicted by the animal bioassay conducted by Maltoni. With regard to the liver, even the histophthologic type of cancer (angiosarcoma) was observed first in experimental animals. A study of cancer mortality among populations residing proxima

An umbrella review of the evidence associating occupational carcinogens and cancer risk at 19 anatomical sites.

Occupational exposure to carcinogens of increasing cancer risk have been extensively suggested. A robust assessment of these evidence is needed to guide public policy and health care. We aimed to classify the strength of evidence for associations of 13 occupational carcinogens (OCs) and risk of cancers. We searched PubMed and Web of Science up to November 2022 to identify potentially relevant studies. We graded the evidence into convincing, highly suggestive, suggestive, weak, or not significant according to a standardized classification based on: random-effects p value, number of cancer cases…

Pilot studies evaluating the nongenotoxic rodent carcinogens phenobarbital and clofibrate in the rat Pig-a assay.

The Pig-a assay is an emerging and promising in vivo method to determine mutagenic potential of chemicals. Since its development in 2008, remarkable progress has been made in harmonizing and characterizing the test procedures, primarily using known mutagenic chemicals. The purpose of the present study was to evaluate specificity of the Pig-a assay using two nongenotoxic and well-characterized rodent liver carcinogens, phenobarbital and clofibrate, in male F344/DuCrl rats. Daily oral administration of phenobarbital or clofibrate at established hepatotoxic doses for 28 days resulted in substanti…

Presence of unreported carcinogens, Aflatoxins and their hydroxylated metabolites, in industrialized Oaxaca cheese from Mexico City.

Aflatoxins (AFs) are toxic secondary metabolites of the fungi Aspergillus flavus, A. parasiticus and A. nomius. The fungi produce these AFs in cereals, oilseeds and spices. AFs have damaging effects on all organisms, including humans, and their symptoms can be classified as acute (vomiting, hemorrhage and death) or chronic (immunodepression, Reye syndrome, Kwashiorkor, teratogenesis, hepatitis, cirrhosis, and various cancers). Basic AFs (AFB1, AFB2, AFG1, and AFG2) are metabolized in the liver or by microbes that produce hydroxylated metabolites (AFM1, AFM2, and AFP1) and aflatoxicol (AFL), so

Transplacental exposure to environmental carcinogens: Association with childhood cancer risks and the role of modulating factors.

Biological responses to carcinogens from environmental exposure during adulthood are modulated over years or decades. Conversely, during transplacental exposure, the effects on the human foetus change within weeks, intertwining with developmental mechanisms: even short periods of transplacental exposure may be imprinted in the organism for a lifetime. The pathways leading to childhood and juvenile cancers, such as leukaemias, neuroblastoma/brain tumours, hepatoblastoma, and Willm's tumour involve prenatally-induced genomic, epigenomic and/or non-genomic effects caused by xenobiotics. Pregnant …

Disruption of spindle checkpoint function in rats following 28 days of repeated administration of renal carcinogens.

We previously reported that 28-day exposure to hepatocarcinogens that facilitate cell proliferation specifically alters the expression of G1/S checkpoint-related genes and proteins, induces aberrant early expression of ubiquitin D (UBD) at the G2 phase, and increases apoptosis in the rat liver, indicating G1/S and spindle checkpoint dysfunction. The present study aimed to determine the time of onset of carcinogen-specific cell-cycle disruption after repeated administration of renal carcinogens for up to 28 days. Rats were orally administered the renal carcinogens nitrofurantoin (NFT), 1-amino-…

Effect of nutritional status on mutagenicity of urine excreted by rats treated with standard/experimental carcinogens.

Urine samples, collected from Sprague Dawley rats treated with extracts of tobacco/masheri, benzo (a) pyrene, N'-nitrosonornicotine, N'-nitrosodiethylamine and maintained on semi-synthetic diets sufficient or deficient in Vitamin A, B and protein were tested for mutagenicity using Salmonella/microsome assay. The mutagenic activity of urine or various treated groups was in the order deficient diet greater than standard laboratory diet greater than nutritionally sufficient diet. Present results confirmed the earlier observations that nutritionally deficient animals are likely to have more exposu

Investigations into the metabolism and mode of action of the colon carcinogens 1,2-dimethylhydrazine and azoxymethane.

Colon cancer can be induced reliably in rodents with 1,2-dimethylhydrazine and azoxymethane (AOM). Our studies deal with the mode of action of these compounds and their organotropism. A partial summary of our previous work on the metabolism of 1,2-dimethylhydrazine and its inhibition by disulfiram, carbon disulfide and other thiono-sulfur compounds is presented. On-going studies with AOM-14C indicate that in male F-344 rats, this carcinogen is rapidly metabolized to 14CO2 (37%, 48 hours), and to methylazoxymethanol-14C (MAM) (0.6--1%), which, along with other metabolites, appears in the urine.

Investigating the carcinogenic and non-carcinogenic health hazards of heavy metal ions in Spinacia oleracea grown in agricultural soil treated with biochar and humic acid.

This study addressed the bioaccumulation and human health risk among the consumption of Spinacia oleracea grown in agricultural soil treated with humic acid (189-2310 ppm) and biochars (0.00-5.10%.wt). The biochars came from two local feedstocks of rice-husk (RH) and sugar-beet-pulp (SBP) pyrolyzed at temperatures 300 and 600 °C. Total concentrations of Cu, Cd, and Ni found in both the soil and biomass/biochar exceeded global safety thresholds. The bioaccumulation levels of HMs in spinach leaves varied, with Fe reaching the highest concentration at 765.27 mg kg-1 and Cd having the lowest conce

Heavy Metal Contamination in Commercial Coffee Products in Saudi Arabia: Prevalence, Carcinogenic and Non-Carcinogenic Risk Assessment, and Implications for Diabetes and Cardiovascular Disease.

Coffee is deeply embedded in Saudi culture and is among the most consumed beverages worldwide. However, contamination by toxic heavy metals may represent an underrecognized public health concern, particularly in regions already burdened by diabetes mellitus and cardiovascular disease. This study evaluated concentrations of lead (Pb), cadmium (Cd), arsenic (As), chromium (Cr), and nickel (Ni) in 21 commercial coffee samples representing seven product categories marketed in Saudi Arabia. Samples were prepared using EPA Method 3050B acid digestion and analyzed by inductively coupled plasma mass s

Probabilistic approach for assessing water quality, carcinogenic and non-carcinogenic risks of drinking water in a semi-arid region: A correlation and clustering by AI approach.

Global industrialization and urbanization have raised concerns about widespread contamination of water resources. This study evaluated the water quality index (WQI) and health risk assessment in a region with a hot semi-arid climate using a probabilistic method. In addition, a correlation matrix and hierarchical cluster analysis (HCA), a machine learning technique, were employed to investigate the origins of the contaminants. Around 1.75 % of electricity conductivity (EC), 7.98 % of nitrate (NO3), 6.03 % of magnesium (Mg), 4.69 % of sulfate (SO4), 3.5 % of calcium (Ca), 18.29 % of alkalinity (

Cancer incidence and mortality among persons in the Finnish Register of Employees Exposed to Carcinogens, 1979-2018.

INTRODUCTION: It has been estimated that 2% to 8% of cancers in high-income countries are linked to carcinogenic exposures in workplaces. In Finland, employers are obliged by law to provide data annually on selected carcinogens and carcinogenic working processes to the Finnish Register of Employees Exposed to Carcinogens (ASA). In this study, we expand the follow-up period of a previous study on cancer incidence in exposed employees by 16 years and add estimates of cancer mortality. METHODS: The study population included all 142,204 workers registered in ASA from 1979 to 2018. For the cancer …
