Caspase-activation powers anti-Desmoglein 3-induced acantholysis in human epidermis.

Pemphigus vulgaris (PV) is a life-threatening autoimmune blistering disease caused by circulating autoantibodies against desmoglein (Dsg) 1 and Dsg 3. Whether acantholysis in PV results exclusively from antibody binding to Dsgs or involves additional factors remains controversial. Given that Fas-Ligand (FasL), an activator of apoptotic caspase-8, is increased in the serum and the skin of patients with PV, we investigated the role of caspases in anti-Dsg3-mediated acantholysis using both ex vivo and in vitro models. Our results demonstrated that anti-Dsg3 antibodies induced acantholysis ex vivo…

Exosomal export of activated CASPASE-8 is linked to the prevention of TNF-induced cytotoxicity.

Tumor Necrosis Factor (TNF) is a key pro-inflammatory cytokine whose sensing by TNFR1 triggers gene activation or cell death induction. While TNF cytotoxicity can be beneficial during infections by supporting effective immune responses, its chronic or excessive induction is harmful and promotes inflammatory pathologies. Protective brakes, known as cell death checkpoints, normally repress TNF cytotoxicity and therefore constitute crucial safeguards against these diseases. Death by TNF mainly proceeds upon inactivation of a checkpoint by microbial effector proteins or pathological mutations. We

Impaired NLRP3 inflammasome signaling diverts pyroptotic to apoptotic caspase activation in macrophages.

NLRP3 (NLR family pyrin domain-containing 3) inflammasome is a first line of defense of innate immunity, mediating caspase-1-dependent pyroptosis and cytokine release upon danger signaling. Intervention of NLRP3 innate surveillance may cause defects in this signaling pathway, while the host has evolved alternative ways to combat such intervention. Yet it remains incompletely understood whether NLRP3 sensing of danger signaling can divert pyroptosis to other forms of cell death in circumstances of impaired NLRP3 signaling. In this study, we adopted two macrophage models (delayed delivery of tri

TP53 mutations drive therapy resistance via post-mitochondrial caspase blockade.

Acute myeloid leukemia (AML) is a heterogeneous disease characterized by a broad spectrum of molecular alterations that influence clinical outcomes. TP53 mutations define one of the most lethal subtypes of acute myeloid leukemia (AML), driving resistance to nearly all available treatment modalities, including venetoclax plus azacitidine (VenAza). Yet, the molecular basis of this resistance, beyond affecting transactivation of BCL-2 family genes, has remained elusive. Here, we demonstrate that VenAza treatment leads to reduced transcriptional upregulation of the p53 signaling pathway in TP53 mu

Generation of Cell Models with Stable Expression of a Caspase-3 Activity Fluorescent Sensor.

A lentiviral vector was created for inserting the commercial fluorogenic protease biosensor for caspase-3, ZipGFP-Casp3, into cell cultures. Three cultures of mesenchymal stem cells (MSCs) from the liver and placenta, dermal fibroblasts, and colorectal carcinoma clones of HT-29 and Caco2 cell lines with knockout of the main cancer stem cell marker CD133, along with control clones of these lines, were selected as the target cultures for studying apoptosis. All cultures were transduced with the ZipGFP-Casp3 sensor. The transduced cells were sorted, and in some derived cultures, cells undergoing

Caspase-1 activation drives vascular inflammatory processes and hypoperfusion in intravascular hemolysis.

Intravascular hemolysis (IVH), a pathological process associated with various conditions, triggers inflammatory responses, yet the key molecular drivers of these responses are poorly defined, particularly within the vasculature. To explore the role of NLRP3 inflammasome- and caspase-1-dependent pathways in IVH-induced vascular dysfunction, we used in vivo models of acute and chronic IVH, alongside heme stimulation of endothelial cells, thereby isolating this disease mechanism from its etiological causes. IVH induced rapid inflammatory responses in C57BL/6J mice, including interleukin (IL)-1β r

Caspase-11 drives macrophage hyperinflammation in models of Polg-related mitochondrial disease.

Mitochondrial diseases (MtD) represent a significant public health challenge due to their heterogenous clinical presentation, often severe and progressive symptoms, and lack of effective therapies. Environmental exposures, such bacterial and viral infection, can further compromise mitochondrial function and exacerbate the progression of MtD. However, the underlying immune alterations that enhance immunopathology in MtD remain unclear. Here we employ in vitro and in vivo approaches to clarify the molecular and cellular basis for innate immune hyperactivity in models of polymerase gamma (Polg)-r

Dehydroascorbic acid impairs neurite growth through RIPK1-associated caspase activation.

Axonal and neurite loss is a common event in neurodegenerative diseases, such as Alzheimer's disease or amyotrophic lateral sclerosis, which are enhanced by oxidative damage and reactive oxygen species (ROS) production. In the central nervous system, vitamin C can be found as ascorbic acid (AA), its reduced form, or dehydroascorbic acid (DHA), its oxidized form. Vitamin C mainly acts as an antioxidant agent, and homeostasis in the brain is maintained through its recycling between neurons and astrocytes. However, DHA accumulation under pathophysiological conditions has been associated with chan

Molecular cloning and functional characterization of Caspase-8 in goldfish (Carassius auratus L.).

Apoptosis, a form of programmed cell death, is essential for maintaining homeostasis within the internal environment. Caspase-8, an initiator caspase, plays a pivotal role in activating the caspase cascade during the apoptotic process. This study cloned and expressed Caspase-8 from goldfish, aimed to investigate the role of Caspase-8 in the immune response of fish to bacterial infections, specifically those caused by Aeromonas hydrophila. The Gf-Casp8 gene consists of 1425 base pairs, encoding a protein of 474 amino acids with a molecular weight of 54.55 kDa. Quantitative real-time PCR analysi

Caspase-4 promotes metastasis and interferon-γ-induced pyroptosis in lung adenocarcinoma.

Caspase-4 (CASP4) is a member of the inflammatory caspase subfamily and promotes inflammation. Here, we report that CASP4 in lung adenocarcinoma cells contributes to both tumor progression via angiogenesis and tumor hyperkinesis and tumor cell killing in response to high interferon (IFN)-γ levels. We observe that elevated CASP4 expression in the primary tumor is associated with cancer progression in patients with lung adenocarcinoma. Further, CASP4 knockout attenuates tumor angiogenesis and metastasis in subcutaneous tumor mouse models. CASP4 enhances the expression of genes associated with an

Caspase-like activity is associated with bacterial infection of the urine in urinary tract diseases.

Urinary tract infections (UTIs) are a serious public health problem. They can be caused by a number of pathogens, but the most common are Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, Enterococcus faecalis and Staphylococcus saprophyticus. Bacterial infection is diagnosed by examining a urine sample. The presence of bacteria or white blood cells is determined under a microscope or a urine culture is performed. In this study, we used a panel of chromogenic substrates for the qualitative determination of specific enzyme activity in the urine of patients with confirmed bacterial inf

Histone H3 activates caspase-1 and promotes proliferation and metastasis in hepatocellular carcinoma.

Background: As a component of nucleosomes, histone H3 plays an important role in chromosome structure and gene expression. Current studies have mostly focused on the role of histones in epigenetics, but in addition to this, the role of histones themselves in tumor development and microenvironment have been less explored. Methods: Western blot and immunofluorescence were carried out to detect the content and localization of histone H3 in hepatocellular carcinoma. The changes of histone H3 were observed in hypoxia treatment cells, the specific action mechanism of histone H3 was studied by CoIP a

Cyclosporine A (CsA) prevents synaptic impairment caused by truncated tau by caspase-3.

During Alzheimer's (AD), tau protein suffers from abnormal post-translational modifications, including cleaving by caspase-3. These tau forms affect synaptic plasticity contributing to the cognitive decline observed in the early stages of AD. In addition, caspase-3 cleaved tau (TauC3) impairs mitochondrial dynamics and organelles transport, which are both relevant processes for synapse. We recently showed that the absence of tau expression reverts age-associated cognitive and mitochondrial failure by blocking the mitochondrial permeability transition pore (mPTP). mPTP is a mitochondrial comple

An oral caspase inhibitor as monotherapy or with antibiotics eradicates MRSA skin infections in mice.

Staphylococcus aureus is the leading cause of skin and soft tissue infections. With the emergence of antibiotic-resistant bacteria, there is an unmet clinical need to develop immune-based therapies to treat skin infections. Previously, we have shown pan-caspase inhibition as a potential host-directed immunotherapy against community-acquired methicillin-resistant S aureus (CA-MRSA) and other bacterial skin infections. Here, we evaluated the role of irreversible pan-caspase inhibitor emricasan as a monotherapy and an adjunctive with a standard-of-care antibiotic, doxycycline, as potential host-d

Endothelial Caspase-8 prevents fatal necroptotic hemorrhage caused by commensal bacteria.

Caspase-8 transduces signals from death receptor ligands, such as tumor necrosis factor, to drive potent responses including inflammation, cell proliferation or cell death. This is a developmentally essential function because in utero deletion of endothelial Caspase-8 causes systemic circulatory collapse during embryogenesis. Whether endothelial Caspase-8 is also required for cardiovascular patency during adulthood was unknown. To address this question, we used an inducible Cre recombinase system to delete endothelial Casp8 in 6-week-old conditionally gene-targeted mice. Extensive whole body v

Regulatory Roles of Caspase-11 Non-Canonical Inflammasome in Inflammatory Liver Diseases.

An inflammatory response consists of two consecutive steps: priming and triggering, to prepare and activate inflammatory responses, respectively. The cardinal feature of the triggering step is the activation of intracellular protein complexes called inflammasomes, which provide a platform for the activation of inflammatory signaling pathways. Despite many studies demonstrating the regulatory roles of canonical inflammasomes in inflammatory liver diseases, the roles of newly discovered non-canonical inflammasomes in inflammatory liver diseases are still largely unknown. Recent studies have repo

The Role of Caspase Family in Acute Brain Injury: The Potential Therapeutic Targets in the Future.

The caspase family is commonly involved in the pathophysiology of acute brain injury (ABI) through complex apoptotic, pyroptotic, and inflammatory pathways. Current translational strategies for caspase modulation in ABI primarily focus on caspase inhibitors. Because there are no caspase-inhibiting drugs approved for clinical use on the market, the development of caspase inhibitors remains an attractive challenge for researchers and clinicians. Therefore, we conducted the present review with the aim of providing a comprehensive introduction of caspases in ABI. In this review, we summarized the …

Mortality of spontaneous intracerebral haemorrhage patients and high serum caspase-8 concentrations.

PURPOSE: Apoptotic changes in brain samples have been found in haematoma areas of patients with spontaneous intracerebral haemorrhage (SIH) undergoing surgical haematoma evacuation. However, circulating caspase-8 concentrations in SIH patients have not been described. Thus, we carried out this study with the aim to explore whether there is an association of circulating caspase-8 concentrations and mortality in patients with SIH. METHODS: We included patients with severe and supratentorial SIH. We established that the SIH was severe if Glasgow Coma Scale (GCS) was lower than 9. Intensive Care
