Somatic cell hybrids are generated by fusion of two different parental cells. This technology has been used extensively in the production of monoclonal antibodies and has made significant contributions to the field of human genetics through its applications in gene expression, gene mapping, and positional cloning of human disease genes. In our laboratory, we have employed this technique in the positional cloning of several genes for human diseases associated with cytogenetic abnormalities (chromosomal disorders), including translocations. Somatic cell hybrids are constructed by fusing mouse th

Acute Myeloid Leukemia (AML) is a prevalent and aggressive hematologic cancer driven by leukemic stem cells and immune evasion. Among its subtypes, Mixed Lineage Leukemia-AF9 (MLL-AF9)- rearranged AML is exceptionally lethal, with B-cell lymphoma 2 (Bcl2), a key anti-apoptotic protein, supporting leukemic survival and immune dysfunction. Targeting Bcl2 in this context is crucial for dismantling oncogenic circuits and restoring immune activity. Here, we have synthesized, de novo, IL-3 receptor alpha chain (IL-3Rα, commonly known as cluster of differentiation 123 or CD123) targeted aptamer-tethe

The trimeric severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein (S) is the sole viral protein responsible for both viral binding to a host cell and the membrane fusion event needed for cell entry. In addition to facilitating fusion needed for viral entry, S can also drive cell-cell fusion, a pathogenic effect observed in the lungs of SARS-CoV-2-infected patients. While several studies have investigated S requirements involved in viral particle entry, examination of S stability and factors involved in S cell-cell fusion remain limited. A furin cleavage site at the border…

The rubivirus Rubella virus contains the two envelope glycoproteins E2 and E1 as a heterodimeric spike complex embedded in its lipid envelop … show that not only E1, but also E2, is able to mediate infectious viral entry. Furthermore, a cell-cell fusion assay was used to further clarify membrane-fusion activities of E2 and E1 as one of the early steps of infection. It was demonstrated that the capsid protein, when coexpressed in cis, enhances the degree of E2- and E1-mediated cell-cell fusion.

Monocyte-derived dendritic cells (DCs) from adult T cell leukemia are impaired in taking up exogenous antigens. To overcome this impairment, we fused unpulsed DCs to human T-lymphotropic virus type I (HTLV-I)-infected CD4(+) T cells (fusion DC-T cells). The efficiency of fusion was 50% and the fusion cells expressed higher HLA-ABC and CD86 Ags than HTLV-I-infected DCs. The fusion DC-T cells stimulated autologous CD4(+) and CD8(+) T cells, but DCs fused to itself or PHA-blasts did not stimulate any subsets. The functionally highest fusion DC-T cells was obtained when a DC and HTLV-I-infected T

Despite decades of clinical experience, lumbar interbody fusion (LIF) still leads to pseudoarthrosis in 5%-28% of patients. Although tissue-differentiation algorithms offer a powerful predictive framework, their application to spinal fusion modeling is currently limited. In this study, we present one of the first mechano-regulated tissue differentiation algorithms incorporating cell-phenotype-specific activities, with the goal to simulate the fusion process after posterior-LIF. The influence of the model input parameters (n = 23) was first assessed in 2D through fractional factorial analyses t

Soilborne fungal pathogens must locate host roots, stably attach to their surface and assemble a persistent mycelial network before invading the plant, yet how host perception is coupled to these cellular outputs remains poorly understood. Here, we characterize HAM-7, a GPI-anchored cell wall protein of Fusarium oxysporum and orthologue of the Neurospora crassa cell wall integrity sensor, and show that it is required for two mechanistically distinct processes essential for root colonization. Targeted deletion of ham-7 strongly impaired stable mycelial retention on tomato roots while leaving ge

B cell precursor acute lymphoblastic leukemia (BCP ALL) is driven by a combination of genetic alterations that promote invasion and expansion of undifferentiated B cells. Mice that express a NUP98::HOXD13 (NHD13) fusion gene in hematopoietic cells develop a wide range of hematologic malignancy, and when crossed to mice that are prone to develop copy number aberrations (CNA), develop BCP ALL, accompanied by homozygous mutations of Ptpn1. To determine if Ptpn1 deletion can be a causal event leading to BCP-ALL, we crossed NHD13 mice with Ptpn1-/- mice. Mice with NHD13 expression and homozygous Pt

Cell-specific detection of aberrant mRNA in blood is essential for diagnosing and treating hematological malignancies. However, current sensors are unable to function in unprocessed whole blood due to limitations in chemical stability and cell-targeting capability. Here, we engineer a cell-resolved ultrastable sensor for hematology (CRUSH) via spherical-nucleic-acid (SNA) technology, which enables live-cell detection of leukemia fusion transcripts in unprocessed whole blood. CRUSH employs stoichiometrically controlled thiol protector to achieve a defect-free thiol monolayer on AuNPs. This desi

Anaplastic lymphoma kinase-rearranged renal cell carcinoma (ALK-RCC) is a rare, molecularly defined entity in the fifth edition of the World Health Organization classification of tumors. The varied presentation and histomorphology of ALK-RCC requires pathologists to have familiarity and a high degree of suspicion for this entity when faced with unusual renal tumors. Here, we present a patient diagnosed with ALK-RCC, which was clinically suspected to be urothelial carcinoma. Histomorphologic features required consideration and work-up of a broad differential diagnosis, including mesenchymal and

Renal cell carcinoma associated with anaplastic lymphoma kinase rearrangement (ALK-RCC) is a rare subtype within the category of molecularly defined renal cell carcinomas. Due to its low incidence and highly heterogeneous clinicopathological features, ALK-RCC poses a significant diagnostic challenge. The implementation of advanced molecular techniques has facilitated the identification of various fusion partners, which may influence clinical behavior and guide targeted therapies. We report a case of ALK-RCC harboring a novel fusion partner, RAB1A::ALK. This report describes the clinicopatholog

ETV6::RUNX1 fusion is one of the most common genetic alterations among children with precursor B-cell acute lymphoblastic leukemia (B-ALL). This fusion generally results from a t(12;21), which fuses the 5' region of ETV6 to the 3' region of RUNX1. Here we report a novel atypical fusion in a 2-year-old male patient with B-ALL. Chromosome, FISH, and microarray were first performed to define a genetic subtype. Chromosome analysis showed three copies of chromosome 21. FISH results revealed ETV6::RUNX1 fusion. However, the signal pattern was atypical with the two fusion signals sitting on two diffe

Multiple primary malignant neoplasms (MPMNs) refers to the concurrent or sequential development of at least two distinct primary malignant tumors within the body, arising from the same organ or different organs. The current manuscript discusses the unique clinical case of a female patient diagnosed with concomitant metastatic ALK-positive NSCLC and del 5q-MDS and was treated for both entities with oral systemic treatments only with a remarkable response for both diseases.

Comprehensive molecular profiling is now routinely performed in newly diagnosed non-small cell lung carcinomas (NSCLCs) to identify actionable genomic alterations. Although numerous molecular abnormalities have been described in lung carcinomas, rare and unexpected gene fusions may create significant diagnostic challenges, particularly when they are characteristically associated with tumours of different lineages. To our knowledge, this is the first reported case of a primary lung squamous cell carcinoma harbouring an in-frame PAX8::PPARγ fusion with a concurrent FGFR2 exon 7 missense mutation

Extracellular vesicles (EVs) are endogenous vesicles secreted by cells. Exosomes (30-150 nm), are a subset of EVs playing key roles in intercellular communication. Exosomes show promise as cancer chemotherapeutic drug delivery vehicles given their low immunogenicity and cell-specific cytosolic delivery of their contents. However, inefficient drug loading limits their therapeutic application. To address this, methods for the fusion of EVs with therapeutic drug-loaded synthetic liposomes have been developed. While more efficient than passive incubation of EVs with liposomes, these risk either da

The spindle cell variant of papillary thyroid carcinoma (PTC) is exceptionally rare and poses significant diagnostic challenges due to its morphological overlap with other spindle cell lesions of the thyroid. We report a novel case of spindle cell variant PTC in a 66-year-old woman presenting with a TI-RADS 4 thyroid nodule, initially classified as Bethesda III on fine-needle aspiration. Histopathological examination revealed a biphasic tumor composed predominantly of bland spindle cells arranged in solid sheets and fascicles, admixed with entrapped thyroid follicles. Both components demonstra
