Chromosomal instability of SV40-transformed human prostatic epithelial cell lines.

Three SV40-transformed derivatives (G1, T2, and T5) of human prostatic epithelial cells were analyzed karyotypically. For comparison, an SV40 derivative (TA) obtained from isogenic fibroblasts, was also studied. The chromosome complements of these cells, as well as a sub-clone of T2 isolated in soft agar (T2-A5), were analyzed using banding techniques. Numerical as well as structural changes were observed in all transformed cultures. Karyotypic changes in all cells at a given passage level appeared to be random. On the other hand, characteristic differences in modal chromosome number, and type

Recovery kinetics of radiation-induced chromosome aberrations in human G0 lymphocytes.

Declining yields of radiation-induced dicentric chromosomes in human G0 lymphocytes were observed in split-dose experiments with time intervals varied up to 8 h. In agreement with microdosimetric intratrack-intertrack interaction models, only the dose-squared yield component was reduced and approached an asymptotic value equal to one half of the corresponding single exposure yield. For 150 kV X-rays and 13 MeV electrons, at total doses up to 6 Gy, the time constant tau of the approximately exponential decline was practically dose- and quality-independent within a range of 100-180 min. For 10 k

Effects of chromosome constitution on growth and longevity of human skin fibroblast cultures.

Employing standardized cell-culture methods, 10 euploid and 22 constitutionally aneuploid human skin fibroblast strains were assessed in triplicate for total growth potential, growth rates, population-doubling times, and cloning. In addition, longitudinal growth rate studies were carried out with otherwise isogenic 45,X and 47,XXX clonal cultures derived from a mosaic parental strain. Growth rates and longevities were cell-strain specific and highly reproducible among sister cultures of a given strain. There was no systematic correlation between chromosome constitution and any of the measured

Some insights into X chromosome inactivation from studies of human cells.

The inactivation of all but one X chromosome in the somatic cells of mammalian females in an important mechanism for regulating X chromosomal genes. Although the molecular basis for the single active X remains to be elucidated, recent studies have provided some insights. Inactivation occurs about the time of implantation, at least in mouse embryos. On the other hand, germ cells from human embryos have two active X chromosomes and may never be subjected to inactivation. As a consequence of X inactivation, one X chromosome becomes the sole determinant of X specified characteristics of the cell,

Chromosome aberrations in in vitro irradiated lymphocytes from human cord blood.

An in vitro dose effect curve of dicentric chromosome aberrations in human cord blood lymphocytes has been obtained for 250-kV X-rays. This is compared with a curve prepared in an identical manner using blood from adults. The comparison shows a marginally higher dicentric yield in blood of newborns at doses above about 250 rads.

Successive spontaneous abortions with diverse chromosomal aberrations in human translocation heterozygote.

A patient who had 3 first-trimester spontaneous abortions (blighted ova) was found to be carrying a balanced 13/14 Robertsonian translocation. In the 2 cases cytogenetically analyzed, different chromosomal aberration were found (trisomy 16 and supernumerary D elements). Histologic examination of the placentas of all 3 abortions revealed hypovascular or avascular villi, hydropic degeneration, and occasional atypical stromal (Hofbauer-like) cells. In 2 cases the decidua was examined by light microscopy and was diffusely inflamed with a plasmolymphocytic infiltrate. The relation of the maternal t

TSPY, the candidate gonadoblastoma gene on the human Y chromosome, has a widely expressed homologue on the X - implications for Y chromosome evolution.

TSPY, a candidate gene for a factor that promotes gonadoblastoma formation (GBY), is a testis-specific multicopy gene family in the male-specific region of the human Y (MSY) chromosome. Although it was originally proposed that male-specific genes on the Y originated from a transposed copy of an autosomal gene (Lahn & Page 1999b), at least two male-specific genes (RBMY and SRY) descended from a formerly recombining X-Y identical gene pair. Here we show that a TSPY homologue with similar gene structure lies in conserved positions, close to SMCX, on the X chromosome in human (TSPX ) and mouse (Ts

HOX11L1, a gene involved in peripheral nervous system development, maps to human chromosome 2p13.1-->p12 and mouse chromosome 6C3-D1.

OBJECTIVES: To evaluate the effectiveness and safety of non-medical switching from reference to biosimilar etanercept in adult patients with rheumatoid arthritis (RA), psoriatic arthritis (PsA) or axial spondyloarthritis (axSpA) using different information strategies before switching. METHODS: Data of adult patients with RA, PsA or axSpA who had received reference etanercept were retrospectively analysed. Whether or not patients were informed about the switch from reference to biosimilar etanercept was left to the discretion of the treating rheumatologist. Disease activity and function were r

Consistent breakage between consensus recombinase heptamers of chromosome 9 DNA in a recurrent chromosomal translocation of human T cell leukemia.

Chromosomal translocations in lymphoid tumors frequently result from recombination between a normally rearranging antigen receptor gene and a normally non-rearranging second locus. The possibility that the lymphocyte recombinase apparatus plays a role in determining the position of breakage at the second locus has been a matter of controversy because of the inconsistent presence of heptamer-like recognition sequences adjoining breakpoints at this site. To further investigate this issue, sites of DNA recombination were analyzed in both the der(9) and der(7) products of t(7;9)(q34;q32), a recurr

Isolation and characterisation of a novel human gene (C9orf11) on chromosome 9p21, a region frequently deleted in human cancer.

The chromosome 9p21 region has been described to be frequently deleted in several neoplasias. The cyclin dependent kinase inhibitor 2A (CDKN2A or P16) gene was cloned in this region and identified as a tumour suppressor gene. However, much evidence indicates the existence of another tumour suppressor gene located proximal to the CDKN2A gene, which could be involved in cutaneous malignant melanoma (CMM) initiation. In the present report we have further investigated this 9p21 chromosomal region and cloned and characterised a novel gene within it (C9orf11). This gene shares no similarities to any

New variants of the human and rat nuclear hormone receptor, TR4: expression and chromosomal localization of the human gene.

TR4 is a new member of the nuclear hormone receptor family. This receptor is highly conserved in rat and human, but an in-frame insertion of 19 amino acid residues in the amino-terminal (A/B) region was found in the human homolog, which we refer to as hTR4alpha1. By reverse transcription-PCR (RT-PCR) we have identified a human TR4 mRNA (hTR4alpha2) that is analogous in size and sequence to the reported rat TR4. RT-PCR analysis using total RNA derived from various rat tissues revealed a new rat TR4 transcript, referred to as rTR4alpha1, which is homologous to hTR4alpha1 since it contains the ex

A novel zinc finger gene on human chromosome 1qter that is alternatively spliced in human tissues and cell lines.

DNA-binding proteins that share the conserved C2-H2 zinc finger motif have been shown to have important roles as transcriptional regulators of gene expression and have been implicated in several hereditary human diseases. In order to define potential candidate genes for inherited disorders characterized by aberrant gene expression, we utilized Kruppel-related sequences to isolate zinc finger-containing cDNAs. We isolated and characterized two novel zinc finger-encoding cDNAs from a human hepatoblastoma cell line, which demonstrate DNA sequence homology to a recently described human Kruppel-rel

Chromosome and gene rearrangements in immortalized human lymphocytes infected with human T-lymphotropic virus type I.

Karyotypes of 26 human lymphocyte cultures, infected or noninfected with human T-lymphotropic virus type I (HTLV-I), with or without Epstein-Barr virus infection, were analyzed by G-banding. Hypodiploidy and structural abnormalities were seen more frequently in HTLV-I-infected cultures (30%) than in virus noninfected cultures (10%) propagated for less than 200 days. In all of six immortalized cell lines infected with HTLV-I alone or with both HTLV-I and Epstein-Barr virus, clonal chromosomal abnormalities characteristic for each cell line were observed in nearly 100% of cells examined after pr

Microcell-mediated chromosome transfer between non-identical human iPSCs.

Microcell-mediated chromosome transfer (MMCT) is anticipated as a unique strategy to manipulate numbers of chromosomes, including the generation of hyperaneuploidy syndrome models with human induced pluripotent stem cells (hiPSCs). Mouse A9/Chinese hamster ovary (CHO) cell libraries of human monochromosomal hybrids as chromosome donor cells frequently exhibit chromosomal rearrangement in the components. The generation of a new A9/CHO library is time-consuming and laborious. Here, we developed an MMCT method using hiPSCs as chromosome donor and recipient cells, through micronucleation using pac
