Mapping the alpha-globin genes in an Algerian HbH patient and his family.

The organization of alpha-globin genes in normal white European, normal Algerian, and alpha-thalassemic Algerian DNA was examined by restriction endonuclease mapping using HindIII, HpaI, Bamhi, EcoRI, BgIII, and PstI. The results for normal DNA confirm and add to the findings of Surrey et al. and Orkin; the two alpha-genes are approximately 3.0 kb apart. The restriction enzymes BgIII and HpaL cut between the two alpha-genes. Four PstL sites are located: two surrounding each alpha-gene. The physical maps for a number of Algerian controls (normal alpha- and beta-globin biosynthesis profiles) are

A maternal effect shapes early-life adaptive body size variation in house mice.

UNLABELLED: Almost all phenotypic variation is mediated by a combination of genetic and environmental effects. Importantly, the balance of these effects can fluctuate over an organism's lifespan, with the maternal environment being particularly important during early life, especially in mammals. Using house mice as a model, we combine behavioral, molecular, and quantitative genetic approaches to understand how genetic and maternal variation together shape phenotypic divergence in a classic example of morphological adaptation. Temperate house mice are larger than tropical house mice, conforming

Chondrodysplasia of Texel sheep - a new disease of suspected genetic aetiology (abstract).

A chondrodysplasia characterised by dwarfism and varus deformity of the forelimbs has been recognised over four seasons in Texel and Texel-cross lambs on three related properties. Affected lambs appear normal at birth, but by 2 weeks of age have reduced growth rate, shortened neck and legs, varus forelimb deformities and a progressive reluctance to walk. Most die within the first 4 months of life. Some affected lambs have had normal co-twins. Post mortem examination of two severely affected 3-month-old lambs revealed extensive loss of articular cartilage and exposure of subchondral bone on the

Shared genetic architecture between Crohn's disease and IgA nephropathy: implications for a gut-immune-kidney axis.

BACKGROUND: Inflammatory bowel disease (IBD) and IgA nephropathy (IgAN) share features of mucosal immune dysregulation, but the genetic basis of their relationship and the contribution of specific IBD subtypes remain unclear. METHODS: Using public genome-wide association study (GWAS) summary datasets, we performed bidirectional Mendelian randomization (MR) analyses in European and East Asian populations, together with generalized summary-data-based Mendelian randomization (GSMR), Causal Analysis Using Summary Effect Estimates (CAUSE), genetic correlation, enrichment, cross-trait locus analysi

Fetal Autopsy in Stillbirth: Its Acceptance and Role in Determining the Causes and Risk Factors in North India.

BACKGROUND: Stillbirth is a major global health issue. The fetal autopsy is considered one of the primal diagnostic tools to determine the cause of stillbirth over MRI, ultrasound, and genetic testing. It is often the way to obtain a definitive diagnosis and can be helpful for families who are grieving the loss of a pregnancy. However, its acceptance is limited due to cultural, emotional, and logistical challenges. This study aimed to evaluate the acceptance of fetal autopsy and its role in identifying causative and risk factors for stillbirth in north India. MATERIALS AND METHODS: This cross

The need for widely available genomic testing in rare eye diseases: an ERN-EYE position statement.

BACKGROUND: Rare Eye Diseases (RED) are the leading cause of visual impairment and blindness for children and young adults in Europe. This heterogeneous group of conditions includes over 900 disorders ranging from relatively prevalent disorders such as retinitis pigmentosa to very rare entities such as developmental eye anomalies. A significant number of patients with RED have an underlying genetic etiology. One of the aims of the European Reference Network for Rare Eye Diseases (ERN-EYE) is to facilitate improvement in diagnosis of RED in European member states. MAIN BODY: Technological adva

Profibrinolytic effect of the epigenetic modifier valproic acid in man.

AIMS: The aim of the study was to test if pharmacological intervention by valproic acid (VPA) treatment can modulate the fibrinolytic system in man, by means of increased acute release capacity of tissue plasminogen activator (t-PA) as well as an altered t-PA/Plasminogen activator inhibitor -1 (PAI-1) balance. Recent data from in vitro research demonstrate that the fibrinolytic system is epigenetically regulated mainly by histone deacetylase (HDAC) inhibitors. HDAC inhibitors, including VPA markedly upregulate t-PA gene expression in vitro. METHODS AND RESULTS: The trial had a cross-over desi

A chimeric CYP11B1/CYP11B2 gene in glucocorticoid-insuppressible familial hyperaldosteronism.

Although a chimeric gene combining the 11beta-hydroxylase gene (CYP11B1) and the aldosterone synthase gene (CYP11B2) explains the pathophysiology of familial hyperaldosteronism (FH) type I, the contribution of this abnormality to FH type II has not been tested. We screened genomic DNA from a Japanese family with FH type II for the CYP11B1/CYP11B2 gene. The index patient was a 27-year-old woman with hypertension. Hypokalaemia, elevated plasma aldosterone and suppressed plasma renin activity suggested primary aldosteronism. Though computed tomography failed to reveal an adrenal tumour, left adre

Reciprocal longitudinal associations between symptoms of body dissatisfaction and disordered eating, self-harm and suicidal ideation.

BACKGROUND: Eating disorders are severe psychiatric conditions associated with high mortality rates, particularly among young people. These disorders often co-occur with self-harm and suicidal ideation, yet the temporal dynamics between these variables remain poorly understood. AIMS: This study aims to elucidate the longitudinal associations between symptoms of body dissatisfaction and disordered eating, self-harm and suicidal ideation using structural equation modelling. METHOD: Repeated measures of these phenotypes were used to construct a hypothetical model that includes cross-path analys

Phenological Development, Environmental Effects, and Selection of Soybean Lines from Crosses with Contrasting Relative Maturity Groups.

Soybean is the main oilseed crop cultivated worldwide and is important for human and animal nutrition. The duration of flowering, the reproductive period, and maturity is determined by genetic factors and influenced by environmental conditions, including temperature, photoperiod, precipitation, soil type, and crop management, which affect yield and regional adaptation. However, important gaps remain in understanding how soybean lines from different relative maturity groups respond to these conditions, particularly regarding the effects of temperature and precipitation on phenological developme

Cross-sectional imaging: current status and future potential in adult celiac disease.

Celiac disease (CD), triggered by exposure to gluten in genetically susceptible individuals, is an immune-mediated small bowel disease affecting about 1% of the population worldwide. But the prevalence of CD varies with age, sex, and location. A strict gluten-free diet remains the primary treatment for CD, currently. Most of patients with CD respond well to gluten-free diet with good prognosis, while some patients fail to get symptomatic relief or histological remission (e.g., nonresponsive or refractory CD). Because of heterogeneous clinical appearance, the diagnosis of CD is difficult. Moreo

Sex-allocation trade-offs and their genetic architecture revealed by experimental evolution.

The theory of sex allocation is hailed as one of evolutionary biology's great successes. But while it has successfully predicted strategies of resource allocation to male versus female offspring under a wide range of conditions in species with separate sexes, it has been difficult to verify the fundamental assumption of a direct genetic trade-off between male and female allocations when both sexes are expressed together in the same individuals. This trade-off assumption supposes that mutations that increase an individual's male allocation reduce its female allocation, and vice versa. Here, we

The International Consortium for Arthrogryposis: A Collaborative Framework for Early Detection, Care, Research, and Education.

Arthrogryposis multiplex congenita (AMC) is a group of rare congenital conditions, characterized by multiple joint contractures but may involve any body system including central nervous system. AMC is etiologically heterogeneous, with over 400 genetic and many non-genetic causes implicated in its prenatal development. AMC requires lifelong, multidisciplinary management, a challenge compounded by their rarity and phenotypic heterogeneity. Our understanding of AMC is hampered by diagnostic delays, scarce trained specialists, and variable clinician expertise. Research networks are paramount to ha

From Farm to Community: Dispersal of Potentially Pathogenic Staphylococcus and Mammaliicoccus Species and Antimicrobial Resistance Across Shared Environments.

Staphylococcaceae are a diverse bacterial family with important implications for human and animal health. This study highlights the One Health relevance of their environmental dispersal, particularly, by identifying closely related or genetically identical strains circulating between farm and community environments. Environmental Staphylococcaceae strains were isolated from animal farms and interconnected areas within a university setting, both influenced by anthropogenic activities. Genetic diversity analysis via GTG5-PCR revealed that specific strains circulated across these environments, li

Living with Marfan syndrome: coping with stigma.

Social stigmatization can disrupt the ability of individuals with genetic conditions to successfully adapt to their situation. We offer data on perceptions of stigma from a cross-sectional survey of 174 adults with Marfan syndrome by self-report. Fifty-six respondents (32%) reported feeling discriminated against or socially devalued because of having Marfan syndrome. Endorsement of discrimination was significantly correlated with having depressive symptoms, low self-esteem, the view Marfan syndrome has had significant negative consequences on one's life, striae, and perceptions of workplace di

Detection of Salivary miRNAs Reflecting Chronic Periodontitis: A Pilot Study.

The purpose of this cross-sectional pilot study was to find salivary microRNAs (miRNAs) reflecting periodontal condition in chronic periodontitis. One hundred and twenty chronic periodontitis patients (mean age, 68.4 years) participated in the study, from whom unstimulated whole saliva was collected. A multiphase study was conducted to explore salivary miRNAs as biomarkers of periodontitis. At first, a polymerase chain reaction (PCR) array was performed to compare salivary miRNAs profiles in no and mild (no/mild) and severe periodontitis patients. Next, the relative expression of salivary miRN
