Co-existence of Paragonimus harinasutai and Paragonimus bangkokensis metacercariae in fresh water crab hosts in central Viet Nam with special emphasis on their close phylogenetic relationship.

During our epidemiological surveys for Paragonimus species in central Viet Nam, we found four morphologically different Paragonimus metacercariae in mountainous crabs. They were identified as metacercariae of Paragonimus westermani, P. bangkokensis, P. proliferus, and P. harinasutai in the order of their prevalence in crab hosts. This is the first discovery of P. harinasutai in Viet Nam, co-inhabiting with P. bangkokensis and other species. Metacercariae of P. harinasutai were given orally to a cat to obtain adult worms. Then, ITS2 and CO1 sequences of metacercariae and adults of P. harinasuta

The first occurrence of Trichinella murrelli in wild boar in Iran and a review of Iranian trichinellosis.

Trichinella larvae isolated from the thigh muscle of a wild boar, Sus scrofa, captured from Gilan Province, northern Iran, was processed for DNA analysis. Polymerase chain reaction (PCR) for amplification of the 5S rDNA fragment demonstrated a 700 bp band on agarose gel. Analysis of DNA sequencing by BLAST confirmed the isolate as T. murrelli. This report constitutes the first recorded occurrence of T. murrelli in Asia, and also the first occurrence in a wild boar host.

Isolation of Wuchereria bancrofti microfilariae from archived stained blood slides for use in genetic studies and amplification of parasite and endosymbiont genes.

Information on change in genetic diversity of Wuchereria bancrofti is important in view of the launching of the Global Lymphatic Filariasis Elimination Programme, as it may have important consequences on the control operations and on the potential resurgence after their withdrawal. Since attention was not paid to generate such information when the programme was launched, use of archived parasite material will provide an opportunity to derive this information in a prospective manner. In this paper a simple and effective technique is reported for isolation of microfilariae of W. bancrofti from d

Genetic variation in Taenia solium.

Neurocysticercosis is a major zoonotic larval cestode infection that has a worldwide distribution and is of significant public health importance. Knowledge of the genetic structure of Taenia solium can be applied to the epidemiology and transmission of this disease, since genetic variants may differ in infectivity and pathogenicity. Molecular epidemiological approaches can also enable detailed studies of transmission. On a global scale, mitochondrial markers have differentiated between T. solium isolates from Asia and isolates from Africa/Latin America. Intraspecific variation in T. solium has

Scalable assembly of Ascaris mitogenomes from whole-genome data reveals a novel clade.

The genus Ascaris is an important group of giant parasitic roundworms, infecting over 700 million people globally and causing substantial economic losses in domestic pigs. Whilst species of Ascaris are morphologically indistinguishable, analysis of mitochondrial loci has revealed three clades (A, B, C) broadly associated with host species and geographic distribution. The diversity within these lineages may expand with the addition of further genomic data. Here, we present a bioinformatic framework for de novo assembly of complete mitochondrial genomes (mitogenomes) from low-coverage whole-geno

Monorchiids of silverbiddies (Gerreidae) from Queensland, Australia, including two new genera and species.

Five species of monorchiids are known from fishes of the family Gerreidae, of which one is from Australian waters. Here, we report it and two new monorchiids from three species of Gerres Quoy & Gaimard, 1824 from off Lizard Island, northern Great Barrier Reef, and Moreton Bay in south-eastern Queensland: Gerres oyena (Forsskål), Gerres oblongus Cuvier and Gerres subfasciatus Cuvier. One of the new species, found only in G. oblongus at Lizard Island, conforms most closely to the concept of Proctotrema Odhner, 1911. However, it differs from species of Proctotrema in oral sucker shape and locatio

Taxonomic inflation or hidden diversity: unveiling the fact surrounds Lytocestus indicus (Cestoda: Caryophyllidea).

The tapeworms belonging to the order Caryophyllidea (Platyhelminthes, Eucestoda) represent a widespread group of intestinal parasites found in cypriniform and siluriform fishes. This group comprises four families (Balanotaeniidae, Capingentidae, Caryophyllaeidae, Lytocestidae), 42 genera and approximately 130 described species. Additionally, over a hundred new taxa have been reported from India. While many of these taxa are endemic and have unique evolutionary importance, a considerable number of putative new taxa were also invalidated because the quality and reliability of the published data

Identification of Fasciola spp. based on ITS-2 reveals the Fasciola gigantica infection in buffaloes in Nanning city, South China.

Fasciolosis is harmful to ruminant husbandry worldwide. Given the superficial survey on Fasciolosis infection in Guangxi, the main buffalo breeding area in China, an in-depth investigation in the infection of buffaloes in Nanning, the capital of Guangxi Zhuang Autonomous Region, with Fasciola (Platyhelminthes: Trematoda: Digenea) species will provide a theoretical support for the control and prevention of Fasciolosis infection in buffaloes. Five water buffalo livers were collected from an abattoir in Nanning every 2 weeks from June 2018 to April 2019, and a total of 101 livers were obtained. A

Description and molecular characterisation of a new nematode species parasitic in the lungs of Strongylopus grayii (Smith) (Anura: Pyxicephalidae) in South Africa.

Rhabdias delangei n. sp. (Nematoda: Rhabdiasidae) is described from the lungs of the clicking stream frog Strongylopus grayii (Smith) in the Western Cape Province of South Africa. The new species is characterised by the nerve-ring being located just posterior to the mid-length of the oesophagus, a pre-equatorial vulva, as well as a thin-walled and apparently underdeveloped rectum. Pairwise analyses based on ITS-28S and partial cox1 gene fragments showed significant differences between R. delangei n. sp. and its congeners from the Afrotropical Realm. Rhabdias delangei n. sp. is the 12th species

Tm-Shift Detection of Dog-Derived Ancylostoma ceylanicum and A. caninum.

To develop a Tm-shift method for detection of dog-derived Ancylostoma ceylanicum and A. caninum, three sets of primers were designed based on three SNPs (ITS71, ITS197, and ITS296) of their internal transcribed spacer 1 (ITS1) sequences. The detection effect of the Tm-shift was assessed through the stability, sensitivity, accuracy test, and clinical detection. The results showed that these three sets of primers could distinguish accurately between A. ceylanicum and A. caninum. The coefficient of variation in their Tm values on the three SNPs was 0.09% and 0.15% (ITS71), 0.18% and 0.14% (ITS197

Morphological and molecular descriptions of Moniliformis saudi sp. n. (Acanthocephala: Moniliformidae) from the desert hedgehog, Paraechinus aethiopicus (Ehrenberg) in Saudi Arabia, with a key to species and notes on histopathology.

A new acanthocepohalan species, Moniliformis saudi sp. n. is described from the desert hedgehog, Paraechinus aethiopicus (Ehrenberg), in central Saudi Arabia. Fourteen other valid species of Moniliformis Travassos, 1915 are recognised. The new species of Moniliformis is distinguished by having a small proboscis (315-520 µm long and 130-208 µm wide) with two apical pores, 14 rows of 8 hooks each and small hooks, thre largest being 25-31 µm long anteriorly. Distinguishing features are incorporated in a dichotomous key to the species of Moniliformis. The description is augmented by scanning elect

Immunopathological mechanisms underlying the time-course of Trichinella spiralis cardiomyopathy in rats.

The present study shows that isolated, perfused hearts from rats orally infected with Trichinella spiralis have a reduced left ventricular developed pressure (LVDP), heart rate (HR) and coronary flow (CF). This reduction is considerably enhanced by a single bolus (100 pM) of PAF (platelet activating factor, an eosinophil activator), especially at 21 days post-infection (d.p.i.), which is the time of the maximum increase in blood and tissue eosinophilia. Helminthic DNA analysis shows that, from 21 d.p.i. onwards, the morphological and functional changes in the myocardium cannot be ascribed to t

Estimating Prevalence and Infection Intensity of Soil-Transmitted Helminths Using Quantitative Polymerase Chain Reaction and Kato-Katz in School-Age Children in Angola.

Quantitative polymerase chain reaction (qPCR) is gaining recognition in soil-transmitted helminth (STH) diagnostics, especially for Strongyloides stercoralis and differentiating hookworm species. However, sample preservation and DNA extraction may influence qPCR performance. We estimated STH prevalence and infection intensity by using qPCR in schoolchildren from Huambo, Uige, and Zaire, Angola, and compared its performance with that of the Kato-Katz technique (here termed Kato-Katz). Stool samples from 3,063 children (219 schools) were preserved in 96% ethanol and analyzed by qPCR, of which 2,

PREVALENCE OF LYMPHATIC FILARIASIS, MALARIA AND SOIL TRANSMITTED HELMINTHIASIS IN A COMMUNITY OF BARDIYA DISTRICT, WESTERN NEPAL.

Lymphatic filariasis (LF), malaria and soil transmitted helminthiasis (STH) cause major health problems in Nepal, but in spite of this very few stud- ies have been carried out on these parasitic infections in Nepal. A cross sectional survey of all three categories of parasitic infections was carried out in Deuda- kala Village of Bardiya District, western Nepal. A total of 510 individuals aged 5 years and above were examined from finger prick blood for circulating filarial antigen (CFA), malaria antigen using a rapid diagnostic test (RDT), and malaria DNA using a PCR-based assay. Characterisation of ITS2 secondary structures for selected allocreadiid species (Digenea: Plagiorchiida: Gorgoderoidea) and their use in taxonomy and systematics.

Secondary structures of the internal transcribed spacer 2 (ITS2) region of ribosomal RNA (rRNA) were reconstructed for 46 species belonging to 12 genera of the family Allocreadiidae. Most structures exhibited the classical four-domain conformation typical of ITS2, with some notable variants. The C-domains of Bunodera acerinae and Allocreadium sp. 2 were divided into three branches. Structures of Creptotrema funduli and Pseudoparacreptotrema macroacetabulata showed unique conformations, with unusually variable A and B domains. Ten-nucleotide sequence variation at the 5' end of the ITS2 region c

Molecular identification of cestodes from rodents in the Mazury Lake District region of Poland.

Rodents serve as hosts for many species of parasites, including cestodes, but the taxonomy of rodent-infecting tapeworms, especially in Central Europe, is still understudied. Therefore, the current study was conducted to identify larval and adult cestodes in free-living rodents from Northeastern Poland. The fieldwork was conducted between 2014 and 2023 in three forest sites. Cestodes were collected from five rodent species (Clethrionomys glareolus, Alexandromys oeconomus, Microtus agrestis, Apodemus agrarius and A. flavicollis, and were identified through morphology and by several genetic mark

Comparative study of Taqman-based qPCR assay for the detection of Anisakis simplex and Pseudoterranova decipiens.

Anisakidosis is a foodborne parasitic infection caused by the consumption of raw or uncooked seafood that contains third stage larvae from the Anisakidae family. This infection has been observed across the globe, with a particularly high prevalence in South Korea and Japan. Consequently, there is a necessity to compare and analyze the optimal detection methods with a view to preventing Anisakis outbreaks. In this study, a species-specific Taqman-based qPCR method was developed for the detection of the internal transcribed spacer region and mtDNA genes of Anisakis simplex and Pseudoterranova de

Identification of Lepidapedon oregonense as the current world's deepest trematode.

The deepest recorded depth for trematodes currently stands at approximately 6200 m. This depth record was achieved solely through sequence datasets of Lepidapedon sp. obtained from a gastropod. Given that trematodes of this genus typically use fish as definitive hosts, the origin of the trematode sequence was thought to be larval stages. However, the specific species remained unclear owing to the absence of reported adult-stage sequences. In the present study, we definitively identified the deepest trematode as Lepidapedon oregonense by comparing 28S ribosomal DNA sequences from adult worms fr
