Evaluation of cochlear function in an acute endolymphatic hydrops model in the guinea pig by measuring low-level DPOAEs.

During and after microinjection of artificial endolymph into scala media of the guinea pig, the 2f1- f2 -DPOAE at 4.5 kHz generated by low-level primaries was recorded. Reproducible changes were measured when 1.1 microl of artificial endolymph was injected at a rate of 1.65 nl/s (1.53-1.83). This volume corresponds with an acute endolymphatic hydrops of 23%. After the onset of injection the inner ear pressure immediately increased to a mean higher level of 22 Pa, whereas the 2f1- f2 -amplitude and -phase did not change for about 1 min. Thereafter, the amplitude decreased 2.6 dB (+/- 0.7) on av…

Induction of endolymphatic hydrops in the guinea pig by perisaccular deposition of sepharose beads carrying and not carrying immune complexes.

We tried to induce endolymphatic hydrops in guinea pig cochleas by unilateral, perisaccular deposition of sepharose beads carrying immune complexes. Controls consisted of the deposition of sepharose beads without immune complexes and the contralateral, untreated ear. The effects of the treatment were studied by light microscopy and electrophysiological recordings of the gross cochlear potentials 1, 2, and 6 weeks after treatment. Each condition included six animals. Analysis of variance of the morphometric data concerning the ears treated with deposition of the beads showed a statistically sig…

Degenerative changes in the organ of Corti and lateral cochlear wall in experimental endolymphatic hydrops and human Menière's disease.

The pathogenesis of sensorineural hearing loss in Menière's disease and experimental endolymphatic hydrops is not fully understood. At the light microscopic level, there is poor correlation between the histopathology and loss of sensitivity and speech discrimination. The results of electron microscopic investigation of histopathology and alterations in immunoreactivity in the organ of Corti and lateral cochlear wall in the hydropic guinea pig are presented. Loss of outer and inner hair cells and spiral ganglion cells, particularly in the apical turn was evident by light microscopy. By electron…

Adenylate cyclase modulation of ion permeability in the guinea pig cochlea: a possible mechanism for the formation of endolymphatic hydrops.

The pathophysiological mechanisms leading to endolymphatic hydrops in Meniere's disease are unknown. Changes in ionic permeability of the cellular membranes between the endolymph and the perilymph, which alter the composition and osmolarity of the inner ear fluid, may be a major factor in the etiology of endolymphatic hydrops. To determine the possible involvement of adenylate cyclase in the formation of endolymphatic hydrops, we measured the endolymphatic K+, Na+, Cl- activities (AK, ANa, ACl) and the endocochlear potential (EP) by means of ion-selective microelectrodes while inner ear adenyl…

Ionic activities of the inner ear fluid and ionic permeabilities of the cochlear duct in endolymphatic hydrops of the guinea pig.

Ionic activities (K+, Na+, and Cl-) of the perilymph and endolymph of the basal turn were measured using ion-selective microelectrodes in experimentally induced endolymphatic hydrops of the guinea pig. Three months following the obstruction of the endolymphatic duct and sac, the endocochlear potential (EP) of hydroptic ears was measured at 59.7 +/- 9.6 mV (N = 12) which was significantly lower than the EP of the contralateral control ears (84.4 +/- 2.8 mV, N = 12). A paired t-test (P greater than 0.05) showed no significant differences of ion concentrations of the inner ear fluid between the h…

Characterizing nystagmus using a portable eye movement recorder in patients with Meniere's disease and MRI-confirmed endolymphatic hydrops.

OBJECTIVE: Meniere's disease (MD) causes episodic vertigo and fluctuating hearing loss. During attacks, nystagmus may appear either toward (i.e., irritative) or away from (i.e., paralytic) the affected ear, but it is difficult to observe these patterns in real-time. This study captured nystagmus using a portable eye movement recorder (PEMR) at the onset of attacks and analyzed its characteristics in patients with endolymphatic hydrops (EH) confirmed on contrast-enhanced magnetic resonance imaging (CE-MRI). METHODS: This study enrolled 54 patients with MD diagnosed using the Japan Society for …

Optical coherence tomography imaging demonstrates endolymphatic hydrops in the lateral and posterior semicircular canals in noise-exposed mice.

Optical Coherence Tomography (OCT) has been used to characterize cochlear endolymphatic hydrops (ELH) with distention of Reissner's membrane in mice after noise exposure. Noise exposure has been correlated with vestibular dysfunction, so we hypothesized that noise exposure can lead to ELH in the vestibular system. Little work has been performed using OCT to image the membranous labyrinth in the lateral and posterior semicircular canals (SCCs). We show that OCT with 12.5 µm axial resolution and 13.2 µm lateral resolution can image the SCCs and delineate the membranous labyrinth in anesthetized …

Computer-aided detection and quantification of endolymphatic hydrops within the mouse cochlea in vivo using optical coherence tomography.

Diseases that cause hearing loss and/or vertigo in humans such as Meniere’s disease are often studied using animal models. The volume of endolymph within the inner ear varies with these diseases. Here, we used a mouse model of increased endolymph volume, endolymphatic hydrops, to develop a computer-aided objective approach to measure endolymph volume from images collected <italic<in vivo</italic< using optical coherence tomography. The displacement of Reissner’s membrane from its normal position was measured in cochlear cross sections. We validated our computer-aided measurements with manual m…
