Novel pneumocystis antigen discovery using fungal surface proteomics.

Pneumonia due to the fungus Pneumocystis jirovecii is a life-threatening infection that occurs in immunocompromised patients. The inability to culture the organism as well as the lack of an annotated genome has hindered antigen discovery that could be useful in developing novel vaccine- or antibody-based therapies as well as diagnostics for this infection. Here we report a novel method of surface proteomics analysis of Pneumocystis murina that reliably detected putative surface proteins that are conserved in Pneumocystis jirovecii. This technique identified novel CD4(+) T-cell epitopes as well

Intelligent Batch Epitope Identification and Partitioning: an intelligent tool for the identification of B cell dominant epitopes.

Identifying B cell dominant epitopes helps to improve vaccine design and better understand immune evasion of pathogens. Herein, we present the Intelligent Batch Epitope Identification and Partitioning (IBEIP), an intelligent tool for identifying B cell dominant epitope regions based on antigen-neutralizing antibody (Ag-nAb) complex data. IBEIP can accurately map the epitopes on any appointed Ag-nAb complex by analyzing antigen-antibody interactions at a molecular level. Combined with a hierarchical iterative merging model, IBEIP can intelligently merge and analyze mapped epitopes to identify B

OMP16-targeting monoclonal antibodies H4 and E6 reveal essential linear epitopes for Brucella immunoprotection.

Brucellosis, a global zoonosis, faces significant challenges from lacking safe vaccines and therapies. OMP16, a key Brucella virulence factor essential for bacterial survival, also functions as a protective antigen capable of activating host immunity. However, not all OMP16-targeting antibodies confer immunoprotection; antibodies against non-protective epitopes may consume immune resources and even exacerbate infection. To validate the protective potential of two OMP16-targeting monoclonal antibodies (H4/E6) and identify their corresponding epitopes, we established in vitro models showing both

Computational design of experimentally validated multi-epitopes vaccine against hepatitis E virus: An immunological approach.

Hepatitis E virus (HEV) is one of the leading acute liver infections triggered by viral hepatitis. Patients infected with HEV usually recover and the annual death rate is negligible. Currently, there is no HEV licensed vaccine available globally. This study was carried out to design a multi-epitope HEV peptide-based vaccine by retrieving already experimentally validated epitopes from ViPR database leading to epitope prioritization. Epitopes selected as potential vaccine candidates were non-allergen, immunogenic, soluble, non-toxic and IFN gamma positive. The epitopes were linked together by AA

Reverse vaccinology-based prediction of a multi-epitope SARS-CoV-2 vaccine and its tailoring to new coronavirus variants.

The genome feature of SARS-CoV-2 leads the virus to mutate and creates new variants of concern. Tackling viral mutations is also an important challenge for the development of a new vaccine. Accordingly, in the present study, we undertook to identify B- and T-cell epitopes with immunogenic potential for eliciting responses to SARS-CoV-2, using computational approaches and its tailoring to coronavirus variants. A total of 47 novel epitopes were identified as immunogenic triggering immune responses and no toxic after investigation with in silico tools. Furthermore, we found these peptide vaccine

MimoPro: a more efficient Web-based tool for epitope prediction using phage display libraries.

BACKGROUND: A B-cell epitope is a group of residues on the surface of an antigen which stimulates humoral responses. Locating these epitopes on antigens is important for the purpose of effective vaccine design. In recent years, mapping affinity-selected peptides screened from a random phage display library to the native epitope has become popular in epitope prediction. These peptides, also known as mimotopes, share the similar structure and function with the corresponding native epitopes. Great effort has been made in using this similarity between such mimotopes and native epitopes in predicti

Mining for the antibody-antigen interacting associations that predict the B cell epitopes.

BACKGROUND: Predicting B-cell epitopes is very important for designing vaccines and drugs to fight against the infectious agents. However, due to the high complexity of this problem, previous prediction methods that focus on linear and conformational epitope prediction are both unsatisfactory. In addition, antigen interacting with antibody is context dependent and the coarse binary classification of antigen residues into epitope and non-epitope without the corresponding antibody may not reveal the biological reality. Therefore, we take a novel way to identify epitopes by using associations bet
