No detection of c-kit gene mutations in exons 9, 11, 13 and 17 and low CD117 expression in plaque-stage mycosis fungoides.

The growth factor receptor c-kit (CD117) is expressed in immature T-cells and in some advanced forms of mycosis fungoides. c-kit gene mutation results in unrestricted neoplastic proliferation. We aimed to detect by PCR the most frequent exon mutations in seventeen plaque-stage MF patients, in their perilesional skin and in healthy skin donors. We secondarily evaluated CD117 expression by immunohistochemistry in plaque-stage and tumor-stage MF. We detected no mutation in c-kit gene and low CD117 expression was confirmed on atypical cells in one patient. Complete c-kit exon and intron sequences

Two case reports of inherited antithrombin deficiency: a novel frameshift mutation and a large deletion including all seven exons detected using two methods.

An inherited antithrombin deficiency is an autosomal dominant thrombotic disorder. We identified two pedigrees of inherited type I antithrombin deficiency and two responsible mutations in each. A novel 21-22delAA appeared to have caused a frameshift with a premature termination at amino acid +63 in one patient and a large deletion including all seven exons was identified by multiplex ligation-dependent probe amplification in the other. Some asymptomatic relatives of the second patient had the same mutation. The present findings support the value of using more than one method of gene analysis a…

[Family study allows more optimistic prognosis and genetic counselling in a child with a deletion of exons 50-51 of the dystrophin gene].

INTRODUCTION: In frame deletions of exons encoding the central rod domain of dystrophin have been associated with a highly variable phenotype, including asymptomatic individuals. The lack of family history impairs accurate genetic counselling. OBSERVATION: We report on a 4-year-old child suffering from transient episodes of limping at the age of 2 and several episodes of fall since the age of 3. Clinical examination did not show muscle weakness. CPK levels were increased (1300 UI). EMG was normal. Muscle histology showed a rhabdomyolysis without features of muscular dystrophy. Immunolabelling…

Pseudohypoparathyroidism type Ib is not caused by mutations in the coding exons of the human parathyroid hormone (PTH)/PTH-related peptide receptor gene.

Pseudohypoparathyroidism type Ib (PHP-Ib) is thought to be caused by a PTH/PTH-related peptide (PTHrP) receptor defect. To search for receptor mutations in genomic DNA from 17 PHP-Ib patients, three recently isolated human genomic DNA clones were further characterized by restriction enzyme mapping and nucleotide sequencing across intron/exon borders. Regions including all 14 coding exons and their splice junctions were amplified by polymerase chain reaction, and the products were analyzed by either temperature gradient gel electrophoresis or direct nucleotide sequencing. Silent polymorphisms w…

Case Report With Biallelic Variants in GCNT2 Implicates Exon 1B in Congenital Cataracts.

GCNT2-related cataracts is a disorder characterized by bilateral congenital cataracts (CC) of various types (with or without the adult i blood phenotype) and is caused by biallelic variants in GCNT2, which has 3 major isoforms, differentiated by alternative splicing of the first exon (known as exon 1A, B, and C). While the transcript that includes exon 1C is thought to be key for the adult i blood phenotype, it is not clear which transcript(s) are clinically relevant for the CC disease phenotype. We report a proband with CC and a truncating variant in exon 1B (NM_001491.3: c.760dup p.H254Pfs*2

PLCG2 exon-skipped variants: insights into their potential role in chronic lymphocytic leukemia.

Bruton tyrosine kinase inhibitors (BTKi) are successful in the treatment of chronic lymphocytic leukemia (CLL), yet acquired resistance remains a challenge. In our study, REC-1 cells that acquired resistance to the BTKi tirabrutinib by long-term treatment gained a splice site mutation (SSM) c.2236-1G>T in phospholipase C gamma 2 (PLCG2). Notably, a deletion spanning c.2236-23_2238 at the same splice site was identified in CLL from a patient who had disease progression during ibrutinib therapy. These alterations resulted in the exon-skipped variant Δ21, which exhibited enhanced phospholipase ac

DNA end-resection is stimulated by an interaction between BRCA1 exon 11 and TOPBP1.

Approximately 60% of the tumour suppressor protein BRCA1 is encoded by a single exon. Many tumours carry mutations in this exon, often resulting in exon skipping and thus a protein of a severely reduced size. Although none of the well-described protein domains of BRCA1 are encoded by this exon 11, the isoform lacking this part shows a hypomorphic activity in homologous recombination. To better understand the function of this large exon, we performed proteomic analyses to identify interaction partners via this part of the protein. Here, we report a DNA damage- and phospho-dependent interaction

Prolonged Survival With Homozygous Deletion of Exon 9 in Perlman Syndrome: A Case Report.

Perlman syndrome is a rare autosomal recessive overgrowth disorder characterized by macrosomia, nephromegaly, renal dysplasia, and characteristic facial features. It has both similarities and differences to other more common overgrowth syndromes. Pathogenic homozygosity is extremely rare in nonconsanguineous relationships. Survival is predicted by differences among various germline mutations in the DIS3L2 gene on chromosome 2q37.1, with prolonged survival documented in heterozygous mutations allowing partial exoribonuclease function. Although homozygous deletions of exon 9 are rare and have be

The First Molecular Profiling of JAK2 Exon 12 and Flanking Intronic Sequences in Indonesia.

OBJECTIVE: Data on JAK2 Exon 12 mutations in Indonesia are not available. This study aimed to explore the presence of known JAK2 exon 12 coding mutations in V617F-negative myeloproliferative neoplasm (MPN) patients in Indonesia and to describe any novel intronic changes downstream of exon 12. METHODS: A total of 198 V617F-negative MPN samples were screened using allele-specific polymerase chain reaction (AS-PCR) for four commonly reported canonical exon 12 mutations. Of these, 57 cases were selected at random or for atypical PCR band patterns and underwent bidirectional Sanger sequencing of a

Routine sequencing of BRAF exon 15 improves therapeutic stratification for metastatic melanoma.

OBJECTIVE: BRAF V600E/K mutations are significant drivers of metastatic melanoma, and therapies that selectively target these mutations have significantly improved patient long-term survival. Emerging evidence suggests that tumors harboring BRAF non-V600E/K mutations are susceptible to MEK-targeting therapies, but these mutations are not always assessed in clinical workup. To assess whether patient stratification for targeted therapy can be improved, we characterized the prevalence and nature of BRAF non-V600E/K mutations in a cohort of patients diagnosed with metastatic melanoma. METHODS: We

Amivantamab Plus Chemotherapy in Japanese Patients With EGFR Exon 20 Insertions NSCLC.

Epidermal growth factor receptor (EGFR) exon 20 insertion mutations (EGFR Exon 20ins) are the third most common mutations in non-small cell lung cancer (NSCLC) and are associated with a poorer prognosis and resistance to conventional EGFR-tyrosine kinase inhibitors. This subpopulation analysis of the open-label phase 3 trial (PAPILLION) evaluates the efficacy and safety of amivantamab-chemotherapy versus chemotherapy among Japanese patients with locally advanced or metastatic NSCLC with EGFR Exon 20ins mutation (ClinicalTrials.gov, NCT04538664). Patients were randomized 1:1 to either intraveno

Valproic Acid Improves Antisense-Mediated Exon-Skipping Efficacy in mdx Mice.

Duchenne muscular dystrophy (DMD) is a severe genetic disorder characterized by the progressive degeneration of skeletal and cardiac muscles due to the absence of dystrophin. Exon-skipping therapy is among the most promising approaches for treating DMD, with several antisense oligonucleotides (ASO) already approved by the FDA; however, their limited efficacy highlights substantial potential for further improvement. In this study, we evaluate the potential of combining ASO with valproic acid (VPA) to enhance dystrophin expression and improve functional outcomes in a murine model of DMD. Our res

Transcript Imbalance from TENM4 Exon Skipping: Effects on Epilepsy and Genetic Pleiotropy.

TENM4 is a transmembrane protein belonging to the teneurin family. It is localized in neurons and oligodendrocytes, where it regulates oligodendrocyte maturation and myelination. While missense variants of TENM4 were reported to cause essential tremor and schizophrenia, we identified a novel variant of uncertain significance at the exon 10-intron 10 junction, c.1255+2T>C, which was segregated in affected patients with a pedigree of intellectual disability and epilepsy. Minigene assay confirmed that the variant caused an in-frame skipping of exon 10 (ΔE10). To clarify the etiology of the novel
