Semaphorins are a family of axonal guidance molecules that, by virtue of their chemorepulsive or chemoattractive actions, may be the important factors in determining the success or failure of axonal regeneration in the mature nervous system after injury. Here, we have used two adult mouse models of nervous system injury to evaluate the neuronal expression of Semaphorin3C (Sema3C) in regenerating (facial motoneurons) and non-regenerating (rubrospinal) neurons following axonal injury. Using in situ hybridization (ISH), we observed that uninjured facial motoneurons express Sema3C mRNA and, follow

BACKGROUND: Cortical spreading depression (CSD) is an electrophysiological event underlying migraine aura. Traditional CSD models are invasive and often cause injuries. The aim of the study was to establish a minimally invasive optogenetic CSD model and identify the active networks after CSD using whole-brain activity mapping.
METHODS: CSD was induced in mice by light illumination, and their periorbital thresholds and behaviours in the open field, elevated plus-maze and light-aversion were recorded. Using c-fos, we mapped the brain activity after CSD. The whole brain was imaged, reconstructed

PURPOSE: Of all nonauditory sensory systems, only the somatosensory system seems to be related to tinnitus (eg, temporomandibular joint syndrome and whiplash). The purpose of this study is to describe the distinguishing characteristics of tinnitus associated with somatic events and to use these characteristics to develop a neurological model of somatic tinnitus.
MATERIALS AND METHODS: Case series.
RESULTS: Some patients with tinnitus, but no other hearing complaints, share several clinical features including (1) an associated somatic disorder of the head or upper neck, (2) localization of th

In the present report, we discussed the case of a 57-year-old man with unilateral masticatory muscle weakness, nystagmus, skew deviation and facial hypesthesia due to pontine tegmental infarction. Trigeminal motor neuropathy attributed to brain infarction is very rare. Brain magnetic resonance imaging revealed a small dot-like infarction lesion in the pontine tegmentum. Masticatory muscle weakness was confirmed by an electrophysiological study performed on the day after admission in which there was an incomplete interference pattern without spontaneous denervation activity, suggesting that the

Surgical technique and functional results of implantation of the acoustic implant of the brain stem (BS) are described for a 16-year-old patient with bilateral vestibular shvannoma (neurofibromatosis of type 2). Initially, a large (IV degree) tumor was removed from the left cerebellopontile angle using the translabyrinthine-transcochlear approach. The function of the facial nerve recovered completely. The BS implant was implanted a year later in the course of removing shvannoma from the right cerebellopontile angle using the above approach. The patient received 22-channel implant Nucleus-CI22M

A localized pontine lesion affecting the abducens nucleus or paramedian pontine reticular formation (PPRF), the medial longitudinal fasciculus (MLF), and the facial nerve fascicle results in eight-and-a-half syndrome, a rare neurological condition. It manifests as ipsilateral peripheral facial nerve palsy, internuclear ophthalmoplegia (INO), and horizontal gaze palsy. This condition typically indicates a minor but well-placed infarct in the dorsal pons. We diagnosed eight-and-a-half syndrome in a patient who presented with dizziness and double vision, with a background history of hypertension

Peripheral facial paralysis refers to the involvement of the facial nerve (VII cranial nerve) at any point along its path, which starts from its nucleus, located in the pons, and extends to its most distal branches. The etiology is heterogeneous, including viral infections, bacterial infections, trauma, and neoplasms, among others. However, in the majority of cases, the cause is idiopathic, commonly referred to as Bell's palsy. The diagnosis is therefore one of exclusion, based in particular on the physical examination. Naturally, the diagnosis is decisive in directing the therapeutic approach

Recently, the neural mechanism of respiratory control in the brainstem has been extensively analyzed mainly in vitro. A neuronal group in the ventrolateral medulla, the ventral respiratory group (VRG), is important in respiratory rhythm and pattern generation. A small region in the rostral VRG, the pre-Bötzinger Complex (pre-BötC), is the kernel of respiratory rhythmogenesis. A novel region ventrolateral to the facial nucleus, the para-facial respiratory group (pFRG), was found and has been considered to also generate respiratory rhythm. These two oscillators, pre-BötC and pFRG, are coupled an…

We identified p53-activated gene 608 (PAG608) as a specifically induced gene in striatal tissue of L-DOPA (100mg/kg)-injected hemi-parkinsonian rats using differential display assay. In the present study, we further examined morphological distribution of PAG608 in the central nervous system of L-DOPA-treated hemi-parkinsonian rats. PAG608 expression was markedly induced in fibers and neuronal cells of the lateral globus pallidus and reticular thalamic nucleus adjacent to internal capsule, specifically in the parkinsonian side of L-DOPA-treated models. The protein was also constitutively expres

This study describes the short-term effects of intracaudate microinjections of carbachol in temporarily isolated and restrained Java monkeys. The monkeys were found to display a series of motor disturbances including blepharoptosis, facial twitches, tongue protrusions, ear flattening, torticollis, and compulsive alternations of rapid flexions and extensions of the extremities. In general, carbachol was found to produce consistent effects as far as it concerns its ability to elicit motor disturbances. Three of the five tested monkeys had previously received another series of carbachol injection

Following facial nerve resection in the mouse, a substantial number of neurons reside in an atrophied state (characterized by cell shrinkage and decreased ability to uptake Nissl stain), which can be reversed by re-injury. The mechanisms mediating the reversal of neuronal atrophy remain unclear. Although T cells have been shown to prevent neuronal loss following peripheral nerve injury, it was unknown whether T cells play a role in mediating the reversal of axotomy-induced neuronal atrophy. Thus, we used a facial nerve re-injury model to test the hypothesis that the reversal of neuronal atroph

INTRODUCTION: The average nerve graft length utilized in cross-face nerve grafting for reconstruction of facial nerve palsy is 20-22 cm. While the graft length is thought to be one of the greatest determinants of muscle strength, the mechanism through which this happens remains unknown. We studied changes in axonal regeneration along the length of a 2 cm cross-face nerve graft in a rat model. The hypothesis was that axon count would decrease along the length of the graft.
METHODS: A 2 cm nerve graft (sciatic nerve) was used as a cross-face nerve graft in 16 adult female, 210-250 g, Sprague Da

Spinocerebellar ataxia type 4 (SCA4), also known as 'hereditary ataxia with sensory neuropathy', represents a very rare, progressive and untreatable form of an autosomal dominant inherited cerebellar ataxia (ADCA). Due to a lack of autopsy cases, no neuropathological or clinicopathological studies had yet been performed in SCA4. In the present study, the first available cerebellar and brainstem tissue of a clinically diagnosed and genetically-confirmed German SCA4 patient was pathoanatomically studied using serial thick sections. During this systematic postmortem investigation, along with an o

A 5-month-old male cynomolgus monkey (Macaca fascicularis) with facial deformation and proptosis of the left eye was euthanized due to a rapidly-growing peribulbar and retro-orbital mass. The mass invaded the soft tissue behind the right eye and extended ventro-medially through the lacrimal, ethmoid, maxillary and frontal bones. Caudo-dorsal extention of the mass through the left frontal bone and into the meninges and parenchyma of the frontal cerebral lobe was observed. Histologically, the cell population making up the mass was variable, but most of the neoplastic cells were blast-like with a

Spinocerebellar ataxia type 2 (SCA2) is a progressive autosomal dominantly inherited cerebellar ataxia and is assigned to the CAG repeat or polyglutamine diseases. Recent morphological studies characterized the pathoanatomical features in heterozygous SCA2 patients and revealed severe neuronal loss in a large variety of cerebellar and extra-cerebellar brain sites. In the present study, we examined the brain pathoanatomy of a monozygous twin of a large Hungarian SCA2 family with pathologically extended CAG repeats in both SCA2 alleles. This unique patient was in the initial clinical stage of SC

BACKGROUND: Non-invasive stimulation of the vagus nerve has been proposed as a new neuromodulation therapy to treat primary headache disorders, as the vagus nerve is hypothesized to modulate the headache pain pathways in the brain. Vagus nerve stimulation can be performed by placing an electrode on the ear to stimulate the tragus nerve, which contains about 1% of the vagus fibers. Non-invasive vagus nerve stimulation (nVNS) conventionally refers to stimulation of the cervical branch of the vagus nerve, which is made up entirely of vagal nerve fibers. While used interchangeably, most of the res

The ability to differentiate between rewarding and aversive stimuli is crucial for survival, yet the underlying neural mechanisms allowing animals to make such a distinction remain elusive. Here, using in vivo two-photon calcium imaging, we uncovered how the activity of dopamine-sensitive neurons in the medial nucleus of the central amygdala (CeM) is associated with appetitive and aversive experiences. We found that cocaine and sucrose strongly activated DRD1(+) neurons while suppressing DRD2(+) activity, whereas the aversive quinine stimulus predominantly engaged DRD2(+) neurons, particularly
