Self-propelled double-silicon-shell-coated Janus nanomotors integrating PTT/CDT/GT therapy to enhance synergistic ferroptosis in cancer therapy.

The effective induction of ferroptosis represents a novel strategy for cancer treatment. This study developed a near-infrared (NIR)-responsive Janus nanomotor (CP@SiO2@RSiO2&PMO@SNP) that synergistically amplifies ferroptosis through dual glutathione (GSH) depletion. The nanomotor comprises a rough silica head incorporating copper-doped Prussian blue (CP) and a mesoporous organosilica tail loaded with sodium nitroprusside (SNP). Under 808 nm irradiation, the photothermal effects of the CP core induce a cascade reaction that enhances Cu2 +-catalyzed ·OH production, depletes GSH, and inhibits gl

Cerium-doped Prussian blue biomimetic nanozyme as an amplified pyroptosis inhibitor mitigate Aβ oligomer-induced neurotoxicity in Alzheimer's disease.

Antioxidant enzyme therapy shows promise for treating Alzheimer's disease (AD), but significant challenges remain in achieving effective blood-brain barrier (BBB) penetration and sustained therapeutic effects. We developed a novel neutrophil membrane (NM)-coated cerium-doped Prussian blue biomimetic nanozyme (NM@PB-Ce) that demonstrates outstanding enzymatic properties and targeted therapeutic efficacy. Extensive physicochemical characterization using transmission electron microscopy, X-ray photoelectron spectroscopy, and dynamic light scattering confirmed the successful synthesis of uniform n

Detection of Intraplaque Hemorrhage in Mouse Atherosclerotic Lesions.

Intraplaque hemorrhage is defined as the presence of fresh or lysed erythrocytes, iron deposits in macrophages, and/or a fibrin clot in an atherosclerotic plaque. These features can be detected by hematoxylin and eosin, Martius scarlet Blue, and Perl's iron histological stainings. It is noteworthy that intraplaque hemorrhage is only present in murine atherosclerotic plaques after additional interventions or additional genetic traits affecting matrix degradation or thrombosis. In this chapter, we describe methods to detect intraplaque hemorrhage in mouse atherosclerotic lesions.

Amperometric detection of morphine at a Prussian blue-modified indium tin oxide electrode.

In this work, the electrocatalytic oxidation of morphine (MO) at an optically transparent indium tin oxide (ITO) electrode modified by an electrodeposited Prussian blue (PB) thin film is first demonstrated, and the amperometric detection of MO was then investigated. Experimental results showed that the thin film on the ITO surface, confined to the PB/Berlin green (BG) redox pair, can serve as an excellent mediator which facilitates electron transfer and considerably lowers the overpotential required, as compared to a bare ITO electrode. Thus, PB can be regarded as a promising artificial peroxi

Polycyclodextrin as a linker for nanomedicine fabrication and synergistic anticancer application.

Polycyclodextrin (denoted PCD) composed of cyclodextrin monomer units and 1,3-diethoxypropan-2-ol containing many hydroxyl groups with lone pairs of electrons, easily coordinated with transition metals with empty orbitals. The CD unit can also provide host-guest binding sites for functional molecules. This article utilizes this feature of PCD for the first time as a "linker" to combine transition metal nanomaterials with synergistic functional molecules. We synthesized PCD with 50% CD monomer by epichlorohydrin cross-linking method. Utilizing the coordination effect of the hydroxyl group in PC

Copper-Enriched Prussian Blue Nanomedicine for In Situ Disulfiram Toxification and Photothermal Antitumor Amplification.

In situ toxification of less toxic substance for the generation of effective anticarcinogens at the specific tumor tissue has been a novel paradigm for combating cancer. Significant efforts have been recently dedicated to turning clinical-approved drugs into anticancer agents in specific tumor microenvironment by chemical reactions. Herein, a hollow mesoporous Prussian blue (HMPB)-based therapeutic nanoplatform, denoted as DSF@PVP/Cu-HMPB, is constructed by encapsulating alcohol-abuse drug disulfiram (DSF) into the copper-enriched and polyvinylpyrrolidone (PVP)-decorated HMPB nanoparticles to

Magnetic resonance imaging of stem cell migration.

Noninvasive cellular imaging allows the real-time tracking of grafted cells as well as the monitoring of their migration. In this review, we will focus on cell tracking using MRI, since MRI is noninvasive, clinically transferable, and displays good resolution, ranging from 50 μm in animal experiments up to 300 μm using whole body clinical scanners. In addition to information about grafted cells, MRI provides information about the surrounding tissue (i.e., lesion size, edema, inflammation), which may negatively affect graft survival or the functional recovery of the tissue. Transplanted cells a

Benzene exposure--an experimental machinery for induction of myelodysplastic syndrome: stem cell and stem cell niche analysis in the bone marrow.

Human epidemiologic studies of highly exposed occupational cohorts have demonstrated that inhalation/exposure to benzene can cause several blood disorders, like non-lymphocytic leukemia, pre leukemic stage, aplastic anemia, and other related syndromes collectively considered as bone marrow failure syndromes. Like many other agents [e.g. chemotherapeutics etc] benzene selects the bone marrow as an important target but the exact location and the mechanism of damage is yet unexplored. The present study aimed at delineating benzene induced myelodysplasia and related disorders in an experimental mo

Tumor-targeted near-infrared/ultraviolet-triggered photothermal/gas therapy nanoplatform for effective cancer synergistic therapy.

The integration of photothermal therapy (PTT) and gas therapy (GT) on a nanoplatform shows great potential in cancer treatment. In this paper, a tumor-targeted near-infrared/ultraviolet (NIR/UV) triggered PTT/GT synergistic therapeutic nanoplatform, PB-CD-PLL(NF)-FA, was designed based on Prussian blue (PB) nanoparticles, 5-chloro-2-nitrobenzotrifluoro (NF)-grafted polylysine (PLL(NF)), and folic acid (FA). PB serves as a core to load PLL(NF) through host-guest interaction and can further modify FA. PB-CD-PLL(NF)-FA can be enriched in tumor tissues by passive targeting with enhanced permeabili

Zwitterion-functionalized hollow mesoporous Prussian blue nanoparticles for targeted and synergetic chemo-photothermal treatment of acute myeloid leukemia.

Multifunctional drug delivery systems combining two or more therapies have a wide-range of potential for high efficacy tumor treatment. Herein, we designed a novel hollow mesoporous Prussian blue nanoparticles (HMPBs)-based platform for targeted and synergetic chemo-photothermal treatment of acute myeloid leukemia (AML). The HMPBs were first loaded with the anticancer drugs daunorubicin (DNR) and cytarabine (AraC), and were subsequently coated with polyethylenimine (PEI) through electrostatic adsorption. Then, zwitterionic sulfobetaine (ZS) and CXCR4 antagonist peptide E5 were modified onto th

Co-Delivery of Chemo and Immune Drugs by Prussian Blue Nanocubes for Combinational Therapy and T1-T2W MR Imaging of MDA-MB-231.

Herein, we present Prussian blue nanocube (PBNC)-based nanoformulation, co-loaded with programmed death ligand 1 (PD-L1)/programmed death protein 1 (PD-1) inhibitor (JQ1) and a chemotherapeutic drug (doxorubicin), to improve triple-negative breast cancer (TNBC) cell treatment through combining immuno-, chemo-, and phototherapy. Mechanistically, the synthesized nanoformulation induces robust pyroptotic cell death (∼86%) with a low IC50 (∼15.71 μg/mL) through the CASP3/NF-κβ /IL-1β signaling pathway and remodels the immunosuppressive microenvironment by restoring extracellular and intracellular

Manganese-enriched prussian blue nanohybrids with smaller electrode potential and lower charge transfer resistance to enhance combination therapy.

Prussian blue (PB) is authenticated in clinical treatment, while it generally exhibits unfavorable chemodynamic therapy (CDT) performance. Herein, we developed manganese-doped prussian blue (PBM) nanoparticles to significantly enhance both CDT and photothermal therapy (PTT) effect. The lower redox potential of Mn3+/2+ (0.088 V) in PBM against that of Fe2+/3+ (0.192 V) in PB leads to favorable electron transfer of PBM with respect to PB. Besides, PBM has a lower charge-transfer resistance (Rct) of 2.98 Ω than 4.83 Ω of PB. Once PBM entering the tumor microenvironment (TME), Mn3+ may be readily

Hyaluronic acid-coated Cu/Mn Prussian blue nanocubes amplify cuproptosis and cGAS-STING signaling for synergistic prostate cancer therapy.

Prostate cancer is a prevalent immune-cold malignancy in which hypoxia and glutathione (GSH) enrichment hinder redox-dependent therapies and cuproptosis. A hyaluronic-acid-coated copper-manganese Prussian blue nanomedicine (Cu/Mn-PB@HA) was developed to remodel the tumor microenvironment (TME) and enhance mitochondria-associated cuproptosis. The multivalent Cu/Mn centers enable catalase-like O2 generation, GSH-oxidase-like depletion of GSH, and Fenton-like ·OH production, which together enhance chemodynamic therapy (CDT) and create a favorable window for cuproptosis. In acidic and GSH-rich con

MRI detects white matter reorganization after neural progenitor cell treatment of stroke.

We evaluated the effects of neural progenitor cell treatment of stroke on white matter reorganization using MRI. Male Wistar rats (n = 26) were subjected to 3 h of middle cerebral artery occlusion and were treated with neural progenitor cells (n = 17) or without treatment (n = 9) and were sacrificed at 5-7 weeks thereafter. MRI measurements revealed that grafted neural progenitor cells selectively migrated towards the ischemic boundary regions. White matter reorganization, confirmed histologically, was coincident with increases of fractional anisotropy (FA, P < 0.01) after stroke in the ischem

Prussian Blue Nanoparticle-Induced Alteration of the Polarization State of Tumor-Associated Macrophages as a Substantial Antitumor Mechanism Against Oral Squamous Cell Carcinoma (OSCC).

INTRODUCTION: Oral squamous cell carcinoma (OSCC) has a poor prognosis due to its immunosuppressive tumor microenvironment (TME), in which tumor-associated macrophages (TAMs) play a pivotal role in promoting disease progression and therapeutic resistance. This study examines whether Prussian blue nanoparticles (PB NPs) could reprogram TAMs and block tumor-stroma communication in OSCC. METHODS: PB NPs were synthesized using polyvinylpyrrolidone-assisted coprecipitation and characterized by transmission electron microscopy, dynamic light scattering, and UV-Vis spectroscopy. In vitro, their effe
