Spidroin-based nanoparticles derived from spider aggregate gland glue for efficient peptide drug delivery.

Spiders can produce a wide variety of silk types for daily functions, each exhibiting unique mechanical properties and biological roles. Among all silk types, aggregate silk is distinctive in that it is a viscoelastic, amorphous, and wet adhesive material, rather than a solid fibrous structure. However, unlike other spider fibroins (spidroins), studies on the biomedical applications of aggregate spidroin (AgSp) remain scarce, particularly in the context of drug delivery systems. Here, we designed and produced three recombinant AgSp (rAgSp) proteins (rAgSp2R, rAgSp4R, and rAgSp6R) with differen…

The engineered bladder patch with a three-layer structure promotes the regeneration and functional recovery of the bladder in a rabbit model.

The engineered bladder patch with a three-layer structure was constructed from a bi-layer silk fibroin scaffold (BSFS) and a methacrylated bladder acellular matrix hydrogel (BAMMAH) that carried induced vascular endothelial cells (VECs) and induced nerve cells (NCs). The patch was developed for bladder augmentation in a rabbit model. The prepared BSFS combined the rigidity of silk fibroin film with the elasticity of silk fibroin sponge, providing a support framework for the construction of engineered bladder. The synthesized BAMMAH exhibited a rapid gelling property, and 2% BAMMAH showed good

Surface modification of polyurethane towards promoting the ex vivo cytocompatibility and in vivo biocompatibility for hypopharyngeal tissue engineering.

Polymeric substrates with good biocompatibility have been widely employed to create a living construct with the complexities of tissue histology and function in the field of tissue engineering. In this study, poly(ester-urethane) (58213, NAT022) was used to be substrate due to its good physical and chemical properties. Proteins like gelatin or silk fibroin were covalently bonded on its surface using method of diamine aminolysis and glutaraldehyde crosslinking, which had been setup in our group in order to improve poly(ester-urethane)'s hydrophilicity and biocompatibility. The modification was

Multi-tissue transcriptomics of the black widow spider reveals expansions, co-options, and functional processes of the silk gland gene toolkit.

BACKGROUND: Spiders (Order Araneae) are essential predators in every terrestrial ecosystem largely because they have evolved potent arsenals … 647 silk gland-specific transcripts, including the few known silk fiber components (e.g. six spider fibroins, spidroins). Silk gland specific transcripts are enriched compared to the entire transcriptome in several functions, including protein degradation, inhibition of protein degradation, and oxidation-reduction. Phylogenetic analyses of 37 gene families …

Surface Functionalization of Thermoplastic Polyurethane Scaffolds to Enhance Microparticle Attachment.

While the use of polymer micro- and nanoparticles for drug delivery has been demonstrated for diverse biomedical applications, not all techniques have allowed for the precise localization of the drug that is needed to support surrounding tissues. Herein, we report a biocompatible thermoplastic polyurethane silk fibroin composite drug delivery platform for localized drug release. The work leverages two distinct materials, combining the biocompatibility of silk fibroin protein particles as a drug carrier with the modularity of a thermoplastic polyurethane scaffold, whose mechanical properties ar

Silk proteins in archaeological soils: mass transfer, structural evolution, and an identification framework.

This study investigated the retention behavior of silk-derived proteins in archaeological soils and seeks to establish an analytical framework for the identification of silk residues preserved in burial environments. Silk fabric ageing and degradation were simulated using soil collected from the Tianzifen Site in Wulong, Chongqing. A multi-dimensional analytical framework combining multiple testing methods was used to characterize the transfer and preservation of silk proteins in soil. The results showed that silk degradation followed a clear staged trajectory. In the intermediate stage, fiber

Biomaterial based implants caused remote liver fatty deposition through activated blood-derived macrophages.

Understanding the biocompatibility of biomaterials is a prerequisite for the prediction of its clinical application, and the present assessments mainly rely on in vitro cell culture and in situ histopathology. However, remote organs responses after biomaterials implantation is unclear. Here, by leveraging body-wide-transcriptomics data, we performed in-depth systems analysis of biomaterials - remote organs crosstalk after abdominal implantation of polypropylene and silk fibroin using a rodent model, demonstrating local implantation caused remote organs responses dominated by acute-phase respon

Dopamine-dependent functions of hyaluronic acid/dopamine/silk fibroin hydrogels that highly enhance N-acetyl-L-cysteine (NAC) delivered from nasal cavity to brain tissue through a near-infrared photothermal effect on the NAC-loaded hydrogels.

Hyaluronic acid/silk fibroin (HA/SF or HS) hydrogels with remarkable mechanical characteristics have been reported as tissue engineering biomaterials. Herein, the addition of dopamine/polydopamine (DA/PDA) to HS hydrogels to develop multifunctional HA/PDA/SF (or HDS) hydrogels for the delivery of drugs such as N-acetyl-L-cysteine (NAC) from nasal to brain tissue is examined. Herein, DA-dependent functions of HDS hydrogels with highly adhesive forces, photothermal response (PTR) effects generated by near infrared (NIR) irradiation, and anti-oxidative effects were demonstrated. An in-vitro study

Wild Silkworm Cocoon Waste Conversion into Tough Regenerated Silk Fibers by Solution Spinning.

Wild silkworm silk fibers have garnered attention owing to their softness, natural color, lightweight, and excellent mechanical properties. Because most wild silkworm cocoons obtained are pierced or dirty after the eclosion process, it is difficult to reel the long filament from the pierced cocoons to use as textile materials. Therefore, damaged wild silkworm cocoons are typically removed during the industrial process. Artificial silk spinning has been developed to transform domesticated silkworm silk solutions into regenerated silk fibers. However, regenerated fibers derived from wild silkwor

Microenvironment Rheology Modulates the Effect of the Anticancer Peptide CIGB300 on 3D Head and Neck Tumoroids.

3D cell systems for in vitro experimental studies are able to mimic the in vivo efficacy of drugs before they are tested on animals. However, many studies are still needed in order to mimic the physiological environment with 3D cell-growth systems. The mechano-physical properties of the microenvironment are relevant for the invasiveness of cancer cells and for their drug resistance. In this study, 3D tumoroids of human oral squamous cell carcinoma (OSCC) CAL27 cells of different stiffnesses were produced using a tunable PEG-silk fibroin hydrogel (PSF), and the antitumor activity of the peptide

Implantation of injectable SF hydrogel with sustained hydrogen sulfide delivery reduces neuronal pyroptosis and enhances functional recovery after severe intracerebral hemorrhage.

Hydrogen sulfide (H2S), an important endogenous signaling molecule, plays an important neuroprotective role in the central nervous system. However, there is no ideal delivery material or method involving the sustained and controlled release of H2S for clinical application in brain diseases. Silk fibroin (SF)-based hydrogels have become a potentially promising strategy for local, controlled, sustained drug release in the treatment of various disorders. Here, we show a silk fibroin (SF)-based hydrogel with sustained H2S delivery (H2S@SF hydrogel) is effective in treating brain injury through ste

Smart textiles in wound care: functionalization of cotton/PET blends with antimicrobial nanocapsules.

Management of infected wounds is one of the most costly procedures in the health care sector. Burn wounds are of significant importance due to the high infection risk that can possibly lead to severe consequences such as sepsis. Because antibiotic wound treatments have caused increasing antibiotic resistance in bacteria, there is currently a strong need for alternative strategies. Therefore, we developed new antimicrobial wound dressings consisting of pH-responsive human serum albumin/silk fibroin nanocapsules immobilized onto cotton/polyethylene terephthalate (PET) blends loaded with eugenol,

A Dopamine Driven Dual-Amplifier Strategy for High Performance Bioorganic Piezoelectrics via Secondary-Structure Modulation and Synergistic Molecular Dipoles.

Bio-proteins with intrinsic piezoelectricity are highly promising for bioelectronics due to their tunable conformations, biocompatibility, and biodegradability. However, their practical applications are severely constrained by difficulties in regulating piezoelectric domains and inherent mechanical brittleness. In this study, we propose a small-molecule dual-amplifier strategy to achieve high performance piezoelectric biomaterials. By introducing a dopamine-mediated modulation approach, a silk fibroin-based film with an enhanced piezoelectric coefficient of up to 12.7 pC N- 1 is obtained. This

Injectable Shear Thinning Hydrogels from Functionalized Silk Proteins for 3D Mammalian Cell Culture.

Silk fibroin hydrogels are a promising platform for tissue engineering due to their biocompatibility and tunability, yet efficient cell recovery from gels remains a challenge. This work synthesizes reversible hydrogels by electrostatically-crosslinking carboxylated silk with linear and branched polyamine. Solvent choice (water, saline, or medium) and protein concentration impacted gel stiffness, mesh size, degradability, and function as an in vitro culture substrate. Hydrogels flowed as a liquid and recovered their viscoelastic properties when cycled under shear. Unlike enzymatically-crosslink

Skin derived precursor Schwann cell-generated acellular matrix modified chitosan/silk scaffolds for bridging rat sciatic nerve gap.

Extracellular/acellular matrix has been attracted much research interests for its unique biological characteristics, and ACM modified neural scaffolds shows the remarkable role of promoting peripheral nerve regeneration. In this study, skin-derived precursors pre-differentiated into Schwann cells (SKP-SCs) were used as parent cells to generate acellular(ACM) for constructing a ACM-modified neural scaffold. SKP-SCs were co-cultured with chitosan nerve guidance conduits (NGC) and silk fibroin filamentous fillers, followed by decellularization to stimulate ACM deposition. This NGC-based, SKP-SC-d

Lightweight, elastic and bioactive scaffolds with enhanced vascularization and anti-inflammation for post-enucleation orbital reconstruction.

Ocular destructive surgery serves as the definitive treatment for irreversible severe damage to intraorbital structures. However, complications such as chronic inflammation, tissue compression, and displacement caused by traditional orbital implant materials significantly impair the life quality of patients in the long term. In this study, we developed a silk fibroin super-elastic sponge (SF-SEA) orbital implant featuring a microporous spherical structure, designed to provide intraorbital structural support and promote tissue regeneration. The orbital implant incorporates a double- crosslinkin
