5-fluorouracil: new applications in complicated retinal detachment for an established antimetabolite.

Long-term reattachment of the retina following the development of proliferative vitreoretinopathy is often prevented by the occurrence of cellular reproliferation. 5-fluorouracil, a synthetic pyrimidine analog, is a potent inhibitor of fibroblast proliferation in cell culture and an animal model of tractional retinal detachment. Doses of up to 1.0 mg, when administered intravitreally to rabbits, result in no demonstrable retinal toxicity by microscopic and electrophysiologic criteria. The first 22 consecutive patients with advanced forms of proliferative vitreoretinopathy were treated with a c…

[Vitamin A acid for the topical management of epithelial neoplasms. Combination with 5-fluorouracil].

15 patients with superficial basaliomas or premalignant epithelial neoplastic disorders (actinic keratosis, leukoplakia, Bowen's disease) were treated locally over three weeks with retinoic acid (RA) alone or in combination with 5-fluorouracil (5-FU). In 11 cases the lesions disappeared clinically, however, only 5 patients proved to be cured by histological examination. 4 of them were treated with RA combined with 5-FU. There was a decrease of the 3H-index after the first week and an increase after the third week of treatment. These findings suggest, that RA inhibits the proliferation of neopl…

Ocular Toxicity of Systemic 5-Fluorouracil Chemotherapy: Role of Lacrimal Gland Transporters.

PURPOSE: 5-Fluorouracil (5-FU) is a commonly used antimetabolite in cancer therapy. Patients (25% to 38%) undergoing chronic systemic 5-FU chemotherapy are affected by ocular toxicity and require cessation of therapy to manage ocular complications. However, the mechanism by which 5-FU crosses the blood-tear barrier is not well understood. The current study investigates the role of organic anion transporters (OAT2 and 3) in the blood-tear barrier of the lacrimal gland, in trafficking 5-FU into the ocular surface. METHOD: Computer simulations were performed to determine the substrate specificit…

Repurposed cAMP-Modulating Agents Enhance 5-Fluorouracil Response Through Membrane-Dependent Mechanisms.

Despite its established role as a cornerstone of chemotherapy for solid tumors, 5-Fluorouracil (5-FU) clinical efficacy remains limited by chemoresistance and heterogeneous drug response. Traditional explanations have focused on intracellular metabolism and genetic determinants; however, increasing evidence identifies the plasma membrane as a critical regulatory interface controlling drug availability, signaling integration, and cell fate. Here, we propose a membrane-centered framework in which compartmentalized cAMP/PKA signaling, modulated by repurposed vasoregulatory agents-levosimendan, mi…

Genetic and transcriptomic alterations underlying aggressiveness in 5-fluorouracil-resistant HCT116 cells.

Resistance to 5-fluorouracil (5-FU) remains an obstacle to effective colorectal cancer (CRC) treatment. Genetic alterations play significant roles in the development of drug-induced chemoresistance, leading to tumor recurrence and increased aggressiveness. This study sought to characterize the transcriptomic profiles of 5-FU-resistant CRC cells. HCT116 colon cancer cells were progressively exposed to increasing concentrations of 5-FU to induce resistance. Chemoresistance and aggressiveness were investigated using MTT, clonogenic, and scratch assays. Transcriptomic alterations were analyzed wit…

5-Fluorouracil induces hair loss by inhibiting β-catenin signaling and angiogenesis.

Chemotherapy-induced alopecia (CIA) is a side effect of the anticancer drug 5-fluorouracil (5-FU). However, the mechanism of action in hair follicle cells is unclear. This study investigated the mechanism of action of 5-FU on the hair cycle and growth in vitro and in vivo. Intraperitoneal injection of 5-FU into C57BL/6 mice delayed anagen initiation, resulting in small hair follicles. 5-FU inhibited angiogenesis by reducing cluster of differentiation 31+ cells, vascular endothelial growth factor, and fetal liver kinase-1 expression in mouse skin tissue and rat vibrissa dermal papilla (rDP) cel…

Salidroside enhances 5-fluorouracil sensitivity against hepatocellular carcinoma via YIPF5-induced mitophagy.

Hepatocellular carcinoma (HCC) is a major medical challenge due to its high incidence and poor prognosis. 5-Fluorouracil (5-FU), although extensively studied in the treatment of HCC and other solid tumors, has limited application as a first-line therapy for HCC due to its resistance and significant inter-patient variability. To address these issues, researchers have explored drug repurposing. One of our key findings in this endeavour was the potent anti-HCC effect of the natural product Salidroside (Sal) when co-administered with 5-FU. Sal was found to inhibit mitosis and promote cellular sene…

Association between Weight Gain and Sex-Related Differences through 5-Fluorouracil Administration.

Research on sex differences has increased across various fields, including cancer and its treatment domains. Reports have indicated sex differences in cancer incidence, survival rates, and the efficacy of anticancer drugs. However, such reports are limited, and in-depth assessments of the underlying mechanisms are still in progress. Although various chemotherapeutic regimens are applicable for breast cancer treatment, reports have surfaced regarding weight gain in female patients undergoing fluorouracil, epirubicin, cyclophosphamide (FEC) or cyclophosphamide, methotrexate, fluorouracil (CMF) t…

Chemotherapeutic agent 5-fluorouracil increases survival of SOD1 mouse model of ALS.

Amyotrophic lateral sclerosis (ALS) is a lethal motor neuron disease with no cure. Currently there are only two ALS drugs approved by the FDA, both with a limited therapeutic effect. In the search for drug candidates for ALS, we studied the effect of known stem cell mobilizing agents (treatment) and antimetabolite 5-fluorouracil (5-FU) (anti-treatment) in SOD1G93A model of ALS. Surprisingly, we found that anti-cancer drug 5-FU increases lifespan, delays the disease onset and improves motor performance in ALS mice. Although we were not able to demonstrate the mechanistic basis of the beneficial…
