Extracellular vesicles (EVs) are key mediators of communication in fungal populations, but the mechanisms underlying lipid exchange through EVs remain poorly characterized. Here, we describe an experimental approach to visualize lipid transfer between fungal cells using the lipophilic dye FM 1-43. This styryl dye efficiently stained membranous structures in multiple fungal species, including Cryptococcus neoformans, Candida albicans, Aspergillus fumigatus, Saccharomyces cerevisiae, and Sporothrix brasiliensis. Staining of the acapsular C. neoformans strain Cap67 with FM 1-43 generated fluoresc

BACKGROUND: Lectins are carbohydrate-binding proteins that are involved in fundamental intra- and extracellular biological processes. They occur ubiquitously in nature and are especially abundant in plants and fungi. It has been well established that certain higher fungi produce lectins in their fruiting bodies and/or sclerotia as a part of their natural resistance against free-living fungivorous nematodes and other pests. Despite relatively high diversity of the glycan structures in nature, many of the glycans targeted by fungal lectins are conserved among organisms of the same taxon and some

Recent studies in pathogenic yeasts reinforce our appreciation of the influence of metal homeostasis on the fungal cell surface. To illustrate this influence, we focus on recent studies on Cryptococcus neoformans, a fungal pathogen with a complex surface of a cell wall with embedded melanin and an attached polysaccharide capsule. Copper and iron are essential yet toxic metals, and current efforts demonstrate the importance of these metals for modulating the surface structure of C. neoformans cells in ways that contribute to fungal-host interactions during disease in vertebrate hosts. In this r

Extracellular polymeric substances (EPS), which envelop on fungal hyphae surface, interact strongly with minerals and play a crucial role in the formation of nanoscale minerals during biomineralization in nature environments. However, it remains poorly understood about the molecular mechanisms of nanominerals (i.e., iron nanominerals) formation in fungal EPS halos during fungus-mineral interactions. This process is vital because fungi typically grow attached to various mineral surfaces in nature. According to the changes of thickness of the fungal cell and EPS layers during the Trichoderma gui

Amidst the escalating demand for biocompatible, biodegradable, and ecologically responsible materials in clinical biomaterial science, filamentous fungal hyphae have emerged as a compelling and underexplored resource for the development of medical sutures. This review consolidates and critically evaluates the utilization of fungal mycelial networks, primarily from filamentous fungi within the Zygomycota phylum, as structural frameworks for biofunctional surgical sutures. The hyphal architecture, characterized by its hierarchical organization, tensile robustness, and inherent biodegradability,

Fungal urinary tract disease has been poorly studied in veterinary medicine. While it is an uncommon condition, identification of fungal cystitis or pyelonephritis raises many clinical questions about optimal management practices. To better understand optimal management approaches and inform guideline development, a scoping review was to identify available evidence pertaining to treatment of fungal urinary tract disease in dogs and cats. After de-duplication and relevance screening, 28 studies were included in the synthesis). Twenty-six (93 %) were single case reports and 2 (7.1 %) were case s

Aspergillus fumigatus is responsible for a disproportionate number of invasive mycosis cases relative to other common filamentous fungi. While many fungal factors critical for infection establishment are known, genes essential for disease persistence and progression are ill defined. We propose that fungal factors that promote navigation of the rapidly changing nutrient and structural landscape characteristic of disease progression represent untapped clinically relevant therapeutic targets. To this end, we find that A. fumigatus requires a carbon catabolite repression (CCR) mediated genetic net

In an attempt to discover a new generation of triazole antifungal agents, a series of triazole-thiazolidinedione hybrids were designed and synthesized by molecular hybridization of the antifungal agent fluconazole and rosiglitazone (an antidiabetic). Most of the target compounds showed good to excellent inhibitory activity against a variety of clinically important fungal pathogens. In particular, compounds (Z)-5-(2,4-dichlorobenzylidene)-3-(2-(2,4-difluorophenyl)-2-hydroxy-3-(1H-1,2,4-triazol-1-yl)propyl)thiazolidine-2,4-dione) (15 c), (Z)-3-(2-(2,4-difluorophenyl)-2-hydroxy-3-(1H-1,2,4-triazo

Due to the limit of available treatments and the emergence of drug resistance in the clinic, invasive fungal infections are an intractable problem with high morbidity and mortality. The cell wall, as a fungi-specific structure, is an appealing target for the discovery and development of novel and low-toxic antifungal agents. In an attempt to accelerate the discovery of novel cell wall targeted drugs, this Perspective will provide a comprehensive review of the progress made to date on the development of fungal cell wall inhibitors. Specifically, this review will focus on the targets, discovery

Caspase recruitment domain-containing protein 9 (CARD9) is a critical adaptor molecule triggered by the interaction of C-type lectin receptors (CLRs) with carbohydrate motifs found in fungi. Consequently, clinical and animal studies indicate that CARD9 is an important regulator of protective immunity against fungal pathogens. Previous studies suggest that CARD9 is important for the induction of protection against Cryptococcus neoformans, an opportunistic fungal pathogen that causes life-threatening infections of the central nervous system in immunocompromised patients. However, the effect of C

UNLABELLED: The substantial increase in fungal diseases, the limited availability of antifungal drugs, and rising resistance to existing treatments highlight the urgent need for developing novel antifungal drugs. Natural products, known for their diverse bioactivities and unique structures, remain critical sources for antifungal drug discovery. In this study, we aimed to identify novel antifungal compounds from soil microbes and isolated a novel 28-membered polyene macrolide, compound 1, from Kitasatospora melanogena. This compound demonstrated potent antifungal activity, with MICs of 4-16 μg/

Fungi are essential members of ecosystems, playing key roles in nutrient cycling, agriculture, and medicine. Their classification into proper species helps us to understand their biodiversity, allowing us to leverage their ecological and practical benefits. A new hybrid deep learning-based technique has been proposed, merging the Vision Transformer and Swin Transformer models with transfer learning frameworks like MobileNetV2, DenseNet121, and EfficientNetB0 for Fungi multiclass classification. This study utilized a publicly available dataset containing 9115 images of five fungal species from

This protocol describes an assay for Photodynamic Inactivation of the plant-pathogenic filamentous fungal species Botrytis cinerea. Grey mold (B. cinerea) occurs worldwide and affects over 200 different crops. Several resistant strains of the species have been collected on fields in past years. Here, mycelial spheres are used as a three-dimensional model to mimic the mold's hyphal structures. The protocol is intended for comparative evaluation of antifungal efficacy of different variables, such as different photosensitizers, concentration dependence, and illumination parameters.

This article illustrates and discusses the applications and value of magnetic resonance imaging (MRI) in the evaluation of sinonasal disease. There are several clinical scenarios where MRI can add value over conventional computed tomography (CT) evaluation of the sinonasal spaces. Specifically, MRI can provide insight through better depiction of the anatomy of certain sinonasal sub-sites including the olfactory structures. It can aid in evaluating anosmia, sinusitis (fungal sinusitis and complications), benign and malignant lesions, CSF leaks and pathology extending into sinonasal spaces.

Mucormycosis is a saprophytic fungal disease. In the region of the head, it has been recognized to invade nasal cavity, paranasal sinus, orbit, or intracranial structures. However, unique involvement of the middle ear, with facial paralysis, still remains unreported. An unusual experience with a diabetic who developed this disease is described. Surgical debridement and amphotericin B administration resulted in near-total cure. The significance of this report is to add a new site involved by mucormycosis, that is, the tympanic cavity.
