OBJECTIVES: Dysbiosis of intestinal microbiota has been implicated in ulcerative colitis (UC). Fucosyltransferase (FUT) 2 and FUT3 determine expression of histo-blood group antigens in the gut and may affect the intestinal microbiota. We investigated the association between FUT2 and FUT3 polymorphisms and UC in Chinese patients.
METHODS: We genotyped FUT2 (rs281377, rs1047781 and rs601338) and FUT3 (rs28362459, rs3745635 and rs3894326) in 485 UC patients and 580 healthy controls using SNaPshot. We also evaluated expression of Lewis a and b antigens in the sigmoid colon of 7 UC patients and 7

Formation of the kidney requires reciprocal signaling among the ureteric tubules, cap mesenchyme and surrounding stromal mesenchyme to orchestrate complex morphogenetic events. The protocadherin Fat4 influences signaling from stromal to cap mesenchyme cells to regulate their differentiation into nephrons. Here, we characterize the role of a putative binding partner of Fat4, the protocadherin Dchs1. Mutation of Dchs1 in mice leads to increased numbers of cap mesenchyme cells, which are abnormally arranged around the ureteric bud tips, and impairment of nephron morphogenesis. Mutation of Dchs1 a

We have investigated by immunochemistry the distribution of H Type 3/4 chains of the ABO histo-blood group system in human submandibular gland using a monoclonal anti-H MBr1 antibody specific for H Type 3/4 chains, and have found the expression of H Type 3/4 chains was mainly in the serous cells. Serous cells from secretors were stained by MBr1 but not by anti-A and anti-B antibodies, whereas serous cells from nonsecretors exhibited a negative reaction with MBr1. Mucous cells were not stained by MBr1. Only a few striated duct cells showed a weak reaction with anti-H MBr1. These results suggest

Alterations on the cell surface of the oligosaccharide portion of glycoproteins and glycolipids are thought to play a role in tumorigenesis. Sialyltransferase catalyzes the incorporation of sialic acid to the carbohydrate group of glycoconjugates. Sialyltransferase has been found elevated in different tumour tissues and in the serum of cancer patients. In the present study we have examined the expression of the beta-galactosyl alpha 2,6-sialyltransferase requiring epitope CDw75, with the monoclonal antibody HH2. 142 breast lesions were included. 21% of the carcinomas in situ and 35% of the inv

The epicardium contributes to cardiac development and regeneration, primarily through paracrine signalling. However, the impact of severe hypoxic stress, such as occurs immediately following myocardial infarction, on the activity of the epicardial secretome remains poorly defined. Here we investigated the angiogenic potential of the epicardial secretome under normoxic and hypoxic conditions. In contrast to expectation, the results demonstrate that the angiogenic effect of epicardial cells on vascular endothelium is hampered under severe hypoxia. Mechanistically, we found that hypoxia-mediated

Fusobacterium nucleatum is a commensal bacterium in the oral cavity and is closely related to several oral infectious diseases. In addition to oral colonization, F. nucleatum is enriched in colorectal cancer (CRC) and is closely associated with CRC progression, while the mechanism of F. nucleatum adhesion to cancer cells has not been fully explained. Sialylation is of high levels in CRC and host cell surface sialoglycans are considered as potential targets for microorganism adhesion. However, the effect of excessive sialylation in CRC on the adhesion of gut microorganisms remains unknown. Here

Noroviruses causes acute gastroenteritis (AGE) worldwide, with children living in low-income areas aged ≤5 years being the most vulnerable. Histo-blood group antigens (HBGA) are recognized as attachment factors for human noroviruses and susceptibility of a given genotype is frequently dependent on the secretor phenotype mediated by FUT2 genotype. This retrospective study involved 734 children aged ≤5 years with AGE (cases group; 66 %, 485/734) or non-AGE (control group; 34 %, 249/734), from the Amazon rainforest (Brazil, Venezuela, and Guyana) and in our previous studies, 87 % of children were

The FUT2 loss-of-function mutations are highly prevalent and are associated with inflammatory bowel disease (IBD). To investigate the impact of FUT2 loss-of-function mutation on the gut microbiota in patients with IBD, 81 endoscopically confirmed IBD patients were genotyped and divided into 3 groups: homozygous for functional FUT2 genes (SeSe), with one copy of non-functional FUT2 gene (Sese), or homozygous for non-functional FUT2 genes (sese). Escherichia, which attaches to fucosylated glycoconjugates, was the only abundant genus exhibiting decreased abundance in sese patients. Compared with

BACKGROUND: If 'passenger lymphocyte syndrome' (PLS) is a well-recognized complication in ABO-mismatched solid organ transplantation, the coexistence of this reaction with recipient's alloimmunization against multiple antigens expressed on the residual red blood cells in the graft is less common and unpredictable.
METHODS: The receiver of an ABO minor-mismatch liver graft from a cadaveric donor developed haemolytic anaemia within 2 weeks after transplantation. The organ donor was of blood group O D+C+c+E+e+ K+k+ Le(a+b-) and the recipient, A1 D-C-c+E-e+ K-k+ Le(a-b-). The donor and recipient

OBJECTIVE: Fucosyltransferase locus 2 (FUT2) controls the presence or absence of blood group substances (A, B, H) in the saliva and other body secretions. Secretor/non-secretor phenotypes are associated with some metabolic and infectious diseases. ABO and FUT2 contribute to build up oligosaccharide structures of the cell surface that are important for blastocyst adhesion and resistance to microbial invasion. We investigated a possible selection on ABH secretor phenotypes during intrauterine life.
STUDY DESIGN: Three hundred and fifty-six consecutive healthy puerperae and their newborn infants

In the course of bioassay-guided study on the EtOAc extract of a culture broth of the marine-derived fungus Cosmospora sp. SF-5060, aquastatin A (1) was isolated as a protein tyrosine phosphatase 1B (PTP1B) inhibitory component produced by the fungus. The compound was isolated by various chromatographic methods, and the structure was determined mainly by analysis of NMR spectroscopic data. Compound 1 exhibited potent inhibitory activity against PTP1B with IC(50) value of 0.19muM, and the kinetic analyses of PTP1B inhibition by compound 1 suggested that the compound is inhibiting PTP1B activity

The activities of glycoprotein sialyl-, galactosyl- and N-acetylglucosaminyl-transferase in serum were examined in 14 alcoholic patients with a median BAC of 65 mmol/l and in 14 healthy age- and sex-matched controls. The median alcohol intake of the patients was 250 g/day for 1-3 weeks. All had elevated levels of carbohydrate-deficient transferrin in serum and normal or near normal liver function tests. The activities of galactosyl- and N-acetylglucosaminyl-transferase at subsaturated substrate concentration were significantly reduced in the patients. Vmax for all three enzymes was 35-55% lowe

Methyl beta-D-galactofuranoside was readily obtained by tin(IV) chloride-catalyzed glycosylation of penta-O-benzoyl-alpha,beta-D-galactofuranose, followed by debenzoylation with sodium methoxide. Glycosylation of 1 with 2,3,5-tri-O-benzoyl-D-galactono-1,4-lactone or with the 6-O-trityl-lactone derivative 5 gave the benzoylated beta-D-galactofuranosyl-(1----6)-D-galactono-1,4-lactone 6 in excellent yield. The structure of disaccharide 6 was confirmed by borohydride reduction to the glycosyl-alditol 7. A byproduct of the condensation reaction of 1 with 4 or 5 was identified as the benzoylated (1

Siglec-15 regulates osteoclast differentiation. The cellular pathways contributing to the expression of Siglec-15 ligand on osteoclast precursors are incompletely understood. We used genome-wide knockout screening to identify genes contributing to the expression of Siglec-15 ligands in the RAW264.7 cell line based on Siglec-15 binding. These genes include sialyltransferase St3gal4 and other enzymes involved in sialic acid biosynthesis and N-glycan processing, indicating that α2-3-linked sialic acid on N-glycans is the glycan epitope (glycotope) recognized by Siglec-15. LRP1 is identified as a

The interactions between viral ligands and cellular receptors, along with the subsequent entry of Human cytomegalovirus (HCMV) into host cells, represent an important focus of research in virus-host relationships. This study investigated the function and mechanistic role of α2,6-sialylation of Integrin β3 during the early stages of infection in fibroblasts. Analysis of the human Integrin β3 glycomics database revealed that the surface glycans of Integrin β3 are extensively modified with terminal sialic acid residues and occur at relatively high abundance. To evaluate their functional relevance

Bone infection represents a pathological intersection of immunity and skeletal remodeling, where inflammatory responses drive excessive bone loss. Neutrophils dominate the early immune infiltration during infection, yet how neutrophil fate within infected bone marrow influences disease progression remains poorly understood. Here, we identify neutrophil-derived apoptotic bodies (Neut-ABs) as key mediators of bone protection during infectious osteomyelitis. Bone infection triggers extensive neutrophil apoptosis, resulting in the accumulation of Neut-ABs. Integrated transcriptomic and proteomic p

Human genetic diversity plays a key role in modulating susceptibility to infectious diseases, reflecting a long history of host-pathogen co-evolution. While clinical outcomes were once attributed primarily to pathogen virulence, evidence today underscores the impact of host genes in infection susceptibility. Noroviruses and rotaviruses, leading causes of acute gastroenteritis, illustrate this dynamic interplay. Resistance to gastroenteritis infection in a subset of individuals is conferred by loss-of-function mutations in FUT2, which encodes α(1,2)-fucosyltransferase responsible for histo-bloo

BACKGROUND: Primary sclerosing cholangitis (PSC) is a chronic liver disease strongly linked to inflammatory bowel disease (IBD), yet its causal genetic drivers and the mechanisms underlying this comorbidity remain poorly understood. This study aimed to identify causal genes for PSC and elucidate the role of the gut-liver axis in its pathogenesis.
METHODS: We performed a multi-omics study integrating transcriptome-wide association studies (TWAS), summary Mendelian randomization (SMR), and colocalization using PSC GWAS data (2871 cases; 12 019 controls) and tissue-specific eQTL data. Bidirectio

The Bombay blood group is an exceptionally rare phenotype characterized by the absence of H antigen on red blood cells due to mutations in the FUT1 gene and absence of H substance in secretions from concurrent FUT2 deficiency. These individuals produce potent anti-H antibodies in addition to anti-A and anti-B antibodies, posing unique challenges in transfusion and solid organ transplantation. We report a successful ABO-H-incompatible renal transplantation in a patient with genetically confirmed Bombay phenotype. Hereditary multiple exostoses (HME) is an autosomal dominant skeletal disorder primarily linked with mutations in Exostosin-1 (EXT1) and Exostosin-2 (EXT2) genes. However, not all HME cases can be explained by these mutations, and its pathogenic mechanisms are not fully understood. Herein, utilizing whole-exome sequencing and genetic screening with a family trio design, we identify two novel rare mutations co-segregating with HME in a Chinese family, including a nonsense mutation (c.204G>A, p.Trp68*) in EXT1 and a missense mutation (c.893T>G, p.Phe298Cys) in FUT7. Functional assays reveal that
