A ribonuclease-targeting chimera (RiboTAC) is a heterobifunctional compound that binds to an RNA target and recruits a ribonuclease to cleave the bound RNA. This study investigates the impact of linker length and composition on RiboTAC potency in leukemia and breast cancer cellular models. Increasing linker length from two polyethylene glycol units to eight increased RiboTAC potency, while longer linker lengths decreased potency. The optimized RiboTAC reduced the abundance of oncogenic LGALS1 transcripts in THP-1 leukemia cells (∼95%) and in MDA-MB-231 triple negative breast cancer cells (∼70%

There is always a need for new approaches for the control of virus burdens caused by seasonal outbreaks, the emergence of novel viruses with pandemic potential and the development of resistance to current antiviral drugs. The outbreak of the 2019 novel coronavirus-disease COVID-19 represented a pandemic threat and declared a public health emergency of international concern. Herein, the role of glycans for the development of new drugs or vaccines, as a host-targeted approach, is discussed where may provide a front-line prophylactic or threats to protect against the current and any future respir

Chikungunya fever (CHIKF) is an arboviral disease that has caused an epidemic burst of chronic inflammatory joint disease in Latin America in the last few years. Efforts are being spent in understanding the mechanisms by which it may cause such articular damage and in determining possible biomarkers of the disease. Galectins (GAL) are a family of animal lectins with an affinity for beta-galactosides. They have multiple functions including working as receptors in innate immunity and as a control for inflammatory responses in both innate and adaptive immunity. They regulate functions of immune c

Sialylation creates a negative charge on the cell surface that can interfere with blastocyst implantation. For example, α2,6-sialylation on terminal galactose, catalyzed by the sialyltransferase ST6GAL1, inhibits the binding of galectin-1, a β-galactoside-binding lectin. We recently reported the potential involvement of galectin-1 and -3 in the pathogenesis of tubal ectopic pregnancy; however, the precise role of galectins and their ligand glycoconjugates remain unclear. Here, we investigated the expression of the genes encoding α2,3- and α2,6-galactoside sialyltransferases (ST3GAL1-6 and ST6G

Pain is a common complication in individuals with HIV, yet the underlying pathogenic mechanisms remain poorly understood. Reactive astrocytes are selectively activated in the spinal dorsal horn of HIV patients experiencing pathological pain, implicating their potential role in pain pathogenesis. However, the specific pathogenic subtype of these reactive astrocytes has not been defined. Here, we identify a distinct subpopulation of pain-associated reactive astrocytes in the lumbar spinal cord of HIV-1 gp120 transgenic mice using single-nucleus RNA sequencing. This astrocyte subpopulation displa

Galectin-4 belongs to the galactoside-binding protein family and is a type of tandem repeat galectin. Despite previous studies indicating its importance in fish immunology, a comprehensive investigation is necessary to fully understand its role in immunomodulatory functions and cellular dynamics. Therefore, this study aimed to explore the immunomodulatory functions of galectin-4 with a particular focus on its antimicrobial and cellular proliferative properties. The open reading frame of PhGal4 spans 1092 base pairs and encodes a soluble protein of 363 amino acids with a theoretical isoelectric

BACKGROUND: Pancreatic cancer-associated diabetes mellitus (PCDM) is a paraneoplastic phenomenon characterized by worsening hyperglycaemia and weight loss. Galectin-3 and S100A9, mediators of PCDM, have pro-inflammatory functions and might thereby induce systemic inflammation and cachexia. We aimed to examine whether PCDM directly mediates cachexia.
METHODS: Consecutive pancreatic cancer (PC) patients with and without PCDM (n = 88 each) with complete information were included. Cachexia was defined as weight loss >5% within 6 months or weight loss >2% and body mass index <20 kg/m2 or sarcopeni

Fabry disease (FD) is an X-linked progressive multisystem disease due to mutations in the gene encoding the lysosomal enzyme α-galactosidase A (α-GalA). The deficiency in α-GalA activity leads to an intra-lysosomal accumulation of neutral glycosphingolipids, mainly globotriaosylceramide (Gb3), in various organs and systems. Enzyme replacement therapy is available and alternative therapeutic approaches are being explored. No diagnostic test, other than sequencing of the α-galactosidase A gene, is available, no biomarker has been proven useful to screen for and predict the disease, and underlyin

INTRODUCTION: Over the past four years, the COVID-19 pandemic has posed serious global health challenges. The severe form of disease and death resulted from the failure of immune regulatory mechanisms, closely highlighted by the dual proinflammatory cytokine and soluble immune checkpoint (sICP) storm. Identifying the individual factors impacting on disease severity, evolution and outcome, as well as any additional interconnections, have become of high scientific interest.
METHODS: In this study, we evaluated a novel panel composed of ten sICPs for the predictive values of COVID-19 disease sev

The pre-B cell receptor (pre-BCR) is an immature form of the BCR critical for early B lymphocyte development. It is composed of the membrane-bound immunoglobulin (Ig) heavy chain, surrogate light chain components, and the signaling subunits Igα and Igβ. We developed monovalent quantum dot (QD)-labeled probes specific for Igβ to study the behavior of pre-BCRs engaged in autonomous, ligand-independent signaling in live B cells. Single-particle tracking revealed that QD-labeled pre-BCRs engaged in transient, but frequent, homotypic interactions. Receptor motion was correlated at short separation

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent synovial inflammation, in which fibroblast-like synoviocytes (FLSs) play a central role by producing pro-inflammatory cytokines and matrix-degrading enzymes. Dimethyl fumarate (DMF) exhibits anti-inflammatory and antioxidant properties, partly through modulation of oxidative stress-related pathways. In this study, anti-CD40 antibody-modified poly(lactic-co-glycolic acid) (PLGA) nanoparticles loaded with DMF were designed to enable targeted delivery to CD40-expressing FLSs isolated from the synovial fluid of

The bone marrow (BM) microenvironment plays a key role in supporting B cell development. In acute lymphoblastic leukemia (B-ALL), the acquisition of oncogenic driver mutations blocks B cell differentiation at specific stages. When these pre-leukemic cells acquire secondary mutations, B-ALL develops. However, the role of the BM microenvironment in pre-leukemic cell fate remains unknown. Here, using a murine model of spontaneous B-ALL development, we show that disrupted pre-BCR signaling in pre-leukemic cells modifies their fate. Blocking expression of the pre-BCR ligand Galectin-1 by the microe

After viral infections, tissue-resident memory T cells (TRM cells) are generated and reactivated rapidly upon re-exposure to previously encountered viral pathogens, providing immediate immune effector functions to limit infection at the site of viral entry. Here we found that a subset of newly recruited CCR2+ monocytes differentiated into memory-stage CCR2-tdTomato+ cells and persisted in the lung for more than 4 months after infection with the influenza virus. Selective depletion of the memory-stage CCR2-tdTomato+ cells reduced significantly the formation of lung CD8+ TRM cells and compromise
