Ultra-high performance liquid chromatography with tandem mass spectrometry (UPLC-MS/MS) for simultaneous estimation of residual glyphosate and its metabolite (amino methyl phosphonic acid - AMPA) in various vaccines.

There is growing interest of monitoring of glyphosate (GLYP) and its active metabolite, aminomethylphosphonic acid (AMPA) in pharmaceuticals globally. Vaccines represents an important class of pharmaceuticals for human and veterinary use. In this work, a robust, sensitive and direct ultra-high-performance liquid chromatography coupled with mass spectrometry (UPLC-MS/MS) based method was developed and validated. This method enables simultaneous detection and quantification of GLYP and AMPA using a simple liquid-liquid extraction technique in complex vaccines. In absence of these residuals, the

Fluorescence spectroscopy to study dissolved organic matter interactions with agrochemicals applied in Swiss vineyards.

UV/Vis fluorescence spectroscopy was used to study the possible interactions of dissolved organic matter (DOM) with the herbicide glyphosate and copper-based fungicide used in vineyards. The study focused on the role of DOM in the transport of these micropollutants from parcels to surface waters (river, lake). Soil solution and river water samples were collected in the Lavaux vineyard area, western Switzerland. Their fluorescence excitation emission matrices (EEM) were decomposed using parallel factor (PARAFAC) analysis, and compared to their content in glyphosate and copper. PARAFAC analysis

Application of glyphosate or paraquat before the emergence of maize plants (Zea mays L.) had no detectable effect on the rhizosphere or endophyte bacterial community structure under the tested field conditions.

Herbicides are applied in agricultural practices to manage weeds. How these herbicides applied before maize emergence (Zea mays L.) might affect the rhizosphere and endophytic bacterial community is still largely unknown. In this study, plots were treated with 400 g paraquat ha-1 (Gramoxone®, paraquat ion 20% w/w), 1,089 g glyphosate ha-1 (Faena®, potassium salt of glyphosate 446 g/L) or weeds were hand removed before maize emerged and the bacterial community was determined in the bulk soil at the start of the experiment, in the bulk soil, rhizosphere, roots and stem at flowering (after 101 da

Alterations in cell viability, reactive oxygen species production, and modulation of gene expression involved in mitogen-activated protein kinase/extracellular regulating kinase signaling pathway by glyphosate and its commercial formulation in hepatocellular carcinoma cells.

Glyphosate (N-phosphonomethyl glycine) is a non-selective, organophosphate herbicide widely used in agriculture and forestry. We investigated the possible toxic effects of the glyphosate active compound and its commercial formulation (Roundup Star®) in the human hepatocellular carcinoma (HepG2) cell line, including their effects on the cytotoxicity, cell proliferation, reactive oxygen species (ROS) levels, and expression of oxidative stress-related genes such as HO-1, Hsp70 Nrf2, L-FABP, and Keap1. MTT and NRU tests indicated that the IC50 values of Roundup Star® were 219 and 140 μM, respectiv

The effect of commercial herbicide exposure on the life history and insecticide resistance phenotypes of the major malaria vector Anopheles arabiensis (Diptera: culicidae).

Herbicides, such as atrazine and glyphosate, are common agrochemicals known to pollute surface ground water. As such, aquatic invertebrates associated with agricultural activities can be exposed to varying doses of these xenobiotics. Anopheles arabiensis, a major malaria vector species in southern Africa, is often closely associated with agricultural activities. This study aimed to examine the effects of larval atrazine or glyphosate exposure on larval and adult life history traits on two laboratory strains of An. arabiensis; one insecticide susceptible (SENN), the other selected for resistanc

The herbicides glyphosate and glufosinate and the cyanotoxin β-N-methylamino-l-alanine induce long-term motor disorders following postnatal exposure: the importance of prior asymptomatic maternal inflammatory sensitization.

BACKGROUND: Prenatal maternal immune activation (MIA) and/or perinatal exposure to various xenobiotics have been identified as risk factors for neurological disorders, including neurodegenerative diseases. Epidemiological data suggest an association between early multi-exposures to various insults and neuropathologies. The "multiple-hit hypothesis" assumes that prenatal inflammation makes the brain more susceptible to subsequent exposure to several kinds of neurotoxins. To explore this hypothesis and its pathological consequences, a behavioral longitudinal procedure was performed after prenata

Titanium dioxide-coated core-shell silica microspheres-based solid-phase extraction combined with sheathless capillary electrophoresis-mass spectrometry for analysis of glyphosate, glufosinate and their metabolites in baby foods.

Because of their extremely low amount in complex samples, it is quite challenging to accurate determine residues of phosphorus-containing amino-acid-like herbicides (PAAHs) in food products. Here we develop novel core-shell mesoporous silica (CSMS) microspheres coated by titanium dioxide (CSMS@TiO2) for extraction and enrichment of PAAHs in baby foods. After the dispersive solid phase extraction (d-SPE), sheathless capillary electrophoresis-mass spectrometry (sheathless CE-MS) is utilized to achieve efficient separation and sensitive detection. The synthesized CSMS@TiO2 composites are characte

Photocurrent-Polarity-Switch-Enabled Signal "on-off-on" Mode Leveraging the Z-Scheme BiOI NFs/Au NPs/TiO2 NPs Heterojunction and Magnetic Nanozyme Inhibition for Glyphosate Detection in Agricultural Matrices.

Critical to sensor performance in real agricultural matrices are high interference resistance and prevention of false-positive/negative signals. To address this, a Z-scheme BiOI nanoflakes (NFs)/Au NPs/TiO2 NPs heterojunction was designed with photocurrent-polarity-switching capability, an inherent feature that could minimize interference from oxidative and reductive species. Subsequently integrated Fe3O4 NPs with catalase-mimicking activity suppressed the photocurrent response to baseline levels, substantially reducing the background noise. In the presence of glyphosate (Gly), the quenching e

Ultratrace-level determination of glyphosate, aminomethylphosphonic acid and glufosinate in natural waters by solid-phase extraction followed by liquid chromatography-tandem mass spectrometry: performance tuning of derivatization, enrichment and detection.

A sensitive and robust analytical method for the quantification of glyphosate, aminomethylphosphonic acid (AMPA) and glufosinate in natural water has been developed on the basis of a derivatization with 9-fluorenylmethylchloroformate (FMOC-Cl), solid-phase extraction (SPE) and liquid chromatography followed by electrospray tandem mass spectrometry (LC-ESI-MS/MS). In order to maximize sensitivity, the derivatization was optimized regarding organic solvent content, amount of FMOC-Cl and reaction time. At an acetonitrile content of 10% a derivatization yield of 100% was reached within two hours i

Soil microbial activity is affected by Roundup WeatherMax and pesticides applied to cotton (Gossypium hirsutum).

Adoption of glyphosate-based weed control systems has led to increased use of the herbicide with continued use of additional pesticides. Combinations of pesticides may affect soil microbial activity differently than pesticides applied alone. Research was conducted to evaluate the influence of glyphosate-based cotton pest management systems on soil microbial activity. Soil was treated with commercial formulations of trifluralin, aldicarb, and mefenoxam + pentachloronitrobenzene (PCNB) with or without glyphosate (applied as Roundup WeatherMax). The soil microbial activity was measured by quantif

Removal and fate of pesticides in a farm constructed wetland for agricultural drainage water treatment under Mediterranean conditions (Italy).

A non-waterproofed surface flow constructed wetland (SFCW), treating agricultural drainage water in Northern Italy, was investigated to gain information on the potential ability for effective pesticide abatement. A mixture of insecticide imidacloprid, fungicide dimethomorph, and herbicide glyphosate was applied, by simulating a single rain event, into 470-m-long water course of the SFCW meanders. The pesticides were monitored in the wetland water and soil for about 2 months after treatment. Even though the distribution of pesticides in the wetland was not uniform, for each of them, a mean diss

Challenges in direct determination of Glyphosate and aminomethylphosphonic acid in water by liquid chromatography-tandem mass spectrometry at modified reverse phase columns: Effect of matrix interferences in fresh and hard water systems.

This study validated two underivatized methods (M1 and M2) according to the Eurachem guidelines to analyze the herbicide Glyphosate and its major metabolite aminomethylphosphonic acid simultaneously by liquid chromatography-tandem mass spectrometry in both fresh and hard waters. Samples were analyzed directly after filtration through 0.22 μm syringe filters in M1, while samples were acidified with acetic acid before filtration in M2. Spike recoveries were greater than 80% for Glyphosate and aminomethylphosphonic acid in both methods. The limit of quantitation was 0.5 μg/L for M1, and 0.1 μg/L

Integrated metabolomics and transcriptomics reveal that Lolium rigidum ABCC3.2 contributes to multiple herbicide resistance.

The widespread application of glyphosate has resulted in the evolution of glyphosate resistance in Lolium rigidum. This study investigated the resistance mechanism in an Australian population of L. rigidum, designated WALR60. Transcriptome sequencing coupled with quantitative real-time PCR (qRT-PCR) analyses identified significant upregulation of ABCC3.2, ABCB11, and ABCC13 in the WALR60 population compared to susceptible controls. Structural analysis revealed that LrABCC3.2 lacks the N-terminal TMD0 domain present in its homolog LrABCC3.1, representing a core ABC transporter. LrABCC3.2-transf

The role of Fe(III) on phosphate released during the photo-decomposition of organic phosphorus in deionized and natural waters.

The photo-decomposition of organic phosphorus is an important route for the phosphorus cycle by which phosphate is regenerated in the aquatic environment. In this study, the role of Fe3+ as a natural photosensitizer toward the decomposition of organic phosphorus to release phosphate was examined in deionized and natural waters under UV and sunlight irradiation using glyphosate as the organic phosphorus model. The results showed that the concentration of glyphosate decreased with irradiation time in the Fe3+/UV and Fe3+/sunlight systems and TOC gradually decreased, which confirmed that glyphosa

Influences of glyphosate residues and different concentrate feed proportions in dairy cow rations during early gestation on performance, blood parameters, functional properties and DNA damage of blood cells in cows and their offspring.

Maternal exposure to various stimuli can influence pre- and postnatal development of the offspring. This potential has been discussed for glyphosate (GLY), active substance in some non-selective herbicides. Accordingly, present study investigated putative effects of GLY residues in rations on cows and their offspring. Dams received either GLY-contaminated (GLY groups) or control (CON groups) rations combined with low (LC groups) or high (HC groups) concentrate feed proportions (CFP) for 16 weeks during mid- and late lactation and early gestation (59±4 days at beginning of GLY exposure; mean±SE

Antioxidant Vitamins Attenuate Glyphosate-Induced Development of Type-2 Diabetes Through the Activation of Glycogen Synthase Kinase-3 β and Forkhead Box Protein O-1 in the Liver of Adult Male Rats.

INTRODUCTION: Glyphosate is a well-known broad-spectrum desiccant and herbicide. It is an active component used widely in popular weed control products like Roundup (BigHaat Agro Pvt Ltd, Bangalore, Karnataka, India), Rodeo (Corteva, Inc., Indianapolis, Indiana, United States), and PondMaster (PBI-Gordon Corporation, Shawnee, Kansas, United States). However, due to sustained presence, they tend to get deposited in the environmental resources and leach into the living system. It has been shown to develop various cancers and diabetes. However, its impact on GSK-3β (glycogen synthase kinase-3 bet
