Aberrant follicle development and anovulation in polycystic ovary syndrome.

Endocrine factors appear to play an important part in arrest of antral follicle maturation in polycystic ovary syndrome (PCOS) but are unlikely to have an impact on early, preantral follicle development, which is clearly abnormal in PCOS. Disordered early folliculogenesis in PCOS is characterised by a higher proportion of follicles entering the growing phase and more prolonged survival of small follicles than in normal ovarian tissue. The factors responsible for aberrant preantral follicle development remain to be determined but IGFs, growth factors of the TGFbeta family and androgens may all

Differences of hexarelin-induced prolactin and cortisol responses between prepubertal and early pubertal short children and lack of correlation with gonadotropin-releasing hormone-induced gonadotropin response.

Hexarelin (HEX), a synthetic hexapeptide with strong GH-stimulating activity, is known to induce the release of prolactin (PRL) and cortisol (F). The responses of GH and F vary according to age and pubertal development, correlating with serum levels of sex steroids, while the release of PRL does not. We evaluated GH, PRL and F responses to HEX (2 microg/kg i.v.) in 19 children with short stature, 12 prepubertal (Tanner stage I) and 7 early pubertal (stage II), and their correlation with those of FSH and LH to GnRH and with the serum levels of testosterone (T) or estradiol (E2). At baseline, th

Hexarelin, a synthetic growth hormone releasing peptide, stimulates prolactin secretion in acromegalic but not in hyperprolactinaemic patients.

OBJECTIVE: In man, new synthetic peptides such as hexarelin have been shown to have a potent and dose dependent GH releasing activity. Furthermore, a significant PRL releasing activity has also been demonstrated, but this has been investigated in less detail. We have therefore evaluated the effect of hexarelin on PRL and GH secretion in patients with active acromegaly or pathological hyperprolactinaemia. DESIGN: Hexarelin (2 micrograms/kg i.v.), a modified derivative of GHRP-6 of the following structure: His-2-Me-D-Trp-Ala-Trp-D-Phe-Lys-NH2, or placebo, was administered in random order on two

Tumor metastasis to brain: role of endothelial cells, neurotrophins, and paracrine growth factors.

An important clinical endpoint in patients with cancer is formation of metastases in the brain. Understanding this phenomenon is important in several types of malignancies, including melanoma, lung and breast cancers. Metastatic tumor cells use specific adhesion molecules to home to brain, and there they must attach to microvessel endothelial cells and respond to brain endothelial cell-derived motility factors and brain invasion factors to invade the CNS. Neurotrophins are important invasion factors in this process, and the ability to invade into the brain may well depend on metastatic cell re

Effects of growth factors on the differentiation of neural crest cells and neural crest cell-derivatives.

During neurulation, neural crest cells migrate to many regions of the body to give rise to a wide variety of cell types. Many premigratory neural crest cells are pluripotent, their potency for differentiation being gradually restricted as they migrate along definite pathways and interact with factors present in the microenvironment. Effects of growth factors on these cells have been discussed in the present review. Mediation of growth factors in differentiation varies with the cell type. Growth factors exert a direct influence on the differentiation of neural and other related neural crest-der

A growth factor in bovine and human testes structurally related to basic fibroblast growth factor.

Homogenates of human testes, epididymides and prostate, and calf testes and epididymides are mitogenic for cultured human foreskin fibroblasts. The growth factors appear similar in that they are inactivated by boiling and acid, but not by treatment with reducing agent. The growth factor in human and bovine testes was partially purified from tissue homogenates, prepared in high ionic strength buffer (pH 7.6) containing protease inhibitors, by ammonium sulfate precipitation and two cycles of heparin-Sepharose chromatography. The growth factor in calf testes was also partially purified from tissu

Isolation and long-term serial cultivation of endothelial cells from the microvessels of the adult human dermis.

A method to isolate and maintain microvascular endothelial cells from the cutaneous vessels of adult human skin in long-term culture has been developed. Endothelial cells lining the microvessels of the papillary dermis are released from surrounding tissue during a brief trypsin incubation (0.3% trypsin, 1% EDTA). Cells are plated onto a fibronectin substrate and maintained in Leibovitz (L15) culture medium containing pooled human serum (50%) and antibiotics. Proliferation is dependent upon the presence of several additional growth factors, cholera enterotoxin (1 X 10(-9) M), isobutyl methylxan

Experimental (Re)structuring: The Clinical Trial as Turning Point Among Medical Research Participants.

Amid the growth of addiction medicine randomized controlled trials (RCTs), scholars have begun examining participants' study experiences, highlighting facilitators and barriers to enrollment. However, this work can overlook the interplay between trial participation and social-structural dimensions among people with substance use disorders linked to the social nature of use, socioeconomic marginalization, and time demands of substance procurement and use. To effectively conduct RCTs with this unique population, it is necessary to examine the broader social context of study participation. We con

Isolation, structure determination and antibacterial activities of succinamide conjugate diacid from Acinetobacter sp. BJ-L.

Strain BJ-L was isolated from the a water sample taken from Xiao Yue River in Beijing and identified as Acinetobacter sp. BJ-L based on the study of its morphology, physiology, biochemistry and 16S rRNA gene sequence. A new antimicrobial substance was produced after the strain was incubated in potato extract medium at 15°C for 72 h. The antimicrobial substance was sequentially purified by reduced pressure condensation, EtOAc extract, and silica gel column chromatography. The structure of the antimicrobial substance was elucidated as succinamide conjugate diacid (SCD) by spectroscopic data inte

[Environmental study on toxicity of water resulting from pesticide and biocide pollution].

A chemical product has a level of danger or security which depends on the concentration and on the exposure time. The toxicity of the water is established by the answer (survival) of an animal organism in contact with a specific chemical product in short time. There are several reliable methods in the world; in any case the most significant aspect is given by the mortal concentration of the substance (mg/L) in connection with the organism dimensions. Among the methods we can point out:--% of mortality;--N.O.E.L. (No Observed Effect Level);--LC50. We must know the following data too:--responsab

Intestinal adaptation after massive intestinal resection.

Patients with short bowel syndrome require long term parenteral nutrition support. However, after massive intestinal resection the intestine undergoes adaptation and nutritional autonomy may be obtained. Given that the complications of parenteral nutrition may be life threatening or result in treatment failure and the need for intestinal transplantation, a more attractive option is to wean patients off nutrition support by optimising the adaptive process. The article examines the evidence that after extensive small bowel resection adaptation occurs in humans and focuses on the factors that inf

Epithelial-mesenchymal interaction in gastric mucosal restoration.

In this review article we discuss the role of growth factors in gastric ulcer healing using an in vitro wound repair model with gastric epithelial and mesenchymal cells. Several growth factors accelerate gastric epithelial and mesenchymal wound healing in vitro with acceleration of cell migration and proliferation. Epidermal growth factor, transforming growth factor-alpha (TGFalpha), hepatocyte growth factor, and insulin accelerate predominantly gastric epithelial wound healing; and TGFbeta and basic fibroblast growth factor predominantly accelerate gastric mesenchymal wound healing. Platelet-

Molecular mechanisms of parathyroid hyperplasia and neoplasia.

In the past decade we have witnessed an explosion in our knowledge of the molecular bases of hyperparathyroidism. In spite of the generally benign phenotype of parathyroid tumors, several of the well-recognized mechanisms in tumor biology are applicable to parathyroid tumorigenesis. The concept of evolution of nonneoplastic endocrine hyperplasia into a true tumor is as operative for the parathyroid tissue as for any other neoplasia. The molecular mechanisms underlying parathyroid tumor development, either on the ground of tissue hyperplasia or as neoplasia "ab initio', are starting to be recog

[Growth and aging factors in physiological senility and Alzheimer's disease].

INTRODUCTION: Both the normal and pathological development and involution of the CNS are regulated by an important set of both, 'growth factors' (inducing growth, differentiation, survival, adaptation or plasticity in neurons) and 'ageing factors' (inducing involution, atrophy, necrosis or apoptosis). In neurodegenerative diseases, significant deficits of growth factors and important presence of ageing factors are being detected, mainly in Alzheimer's disease (AD). DEVELOPMENT: In this review, the most important growth factors (NGF, BDNF, neuromodulators, glial factors, etc.) and ageing facto

Growth factors for human pleural mesothelial cells in soluble products from formed clots.

Fibrin deposition within the pleural space may influence repair following pleural injury. Although the mesothelial surface can organize fibrin, the contribution of pleural mesothelial cells to pleural repair is unknown. During coagulation thrombin cleaves Fibrinopeptide A (FPA, A alpha 1-16) and fibrinopeptide B (FPB) from the A alpha and B beta chains of fibrinogen to generate fibrin monomer. Since these peptides are mitogenic for human fibroblasts, we considered that they might stimulate replication of human pleural mesothelial cells (HPMC). Application of fluid expressed from fibrin clots s

The potential roles of intraovarian peptides in normal and abnormal mechanisms of reproductive physiology.

Normal and abnormal follicular growth and steroidogenesis depend on gonadotropins as well as intraovarian peptide and polypeptide growth factors, which may mediate or potentiate gonadotropin action. Epidermal growth factor is mitogenic to ovarian granulosa and is a potent inhibitor of granulosa aromatase. It may be involved in the apparent arrest of follicular development commonly seen in women with polycystic ovarian syndrome as well as in the blunted response to gonadotropins seen in this syndrome. Insulin-like growth factors are also mitogenic to ovarian granulosa, but in contrast to epider

Chemical synthesis, growth-promoting activity, and metabolism of all-trans retinyl beta-glucose in the rat.

Reaction of all-trans retinol with alpha-D-glucopyranosyl bromide tetrabenzoate in the presence of silver carbonate gave all-trans retinyl beta-glucose in good yield. Depending on the mode of administration, retinyl beta-glucose, which is soluble in water, showed 67-100% of the growth-promoting activity of retinyl acetate in vitamin A-deficient rats. In metabolic studies on vitamin A-deficient rats, retinyl beta-glucose was rapidly hydrolyzed to retinol. The possible therapeutic uses of retinyl glucose are discussed.

DNA synthesis in exocrine and endocrine pancreas after partial hepatectomy in Syrian golden hamsters.

3H-thymidine autoradiography showed an enhanced DNA synthesis in acinar and islet cells of pancreas after partial hepatectomy in syrian golden hamsters. A significant nuclear labeling index of acinar cells was observed between 48 and 84 h and reached control levels by 120 h. An increased labeling index of islet cells was also observed, however, this increase was not statistically significant. These results indicate growth factor(s) produced after partial hepatectomy is capable of inducing DNA synthesis in pancreas.

Generalized acanthosis nigricans.

A patient suffering from an advanced and inoperable gastric carcinoma developed an unusual paraneoplastic syndrome, consisting of intensive pruritus, acanthosis nigricans, sign of Leser-Trélat (seborrhoic keratoses and freckles), warty and papillomatous excrescences, palmoplantar hyperkeratosis and florid papillomatous and granular lesions of the mouth mucosa. The whole skin of the patient was involved. The authors believe this disorder to be generalized acanthosis nigricans. As a cause they suggest the possibility of a growth-stimulating factor, produced by the tumor cells. The relationship o
