Novel program for automatic calculation of EPG variables.

The electrical penetration graph (EPG) technique is the most powerful tool for studying the feeding behavior of pierce-sucking insects. However, calculating EPG variables is often very time-consuming, and consequently, several software programs have been developed for the automatic calculation of EPG variables. Here we present a new user-friendly Excel Workbook that uses a standardized list of EPG variables and follows expert guidelines for calculating them. The program developed in Visual Basic for Applications (VBA) is a step up from the existing software and allows easy data analysis and in

Frequency of processed kaolin application to prevent Diaphorina citri infestation and dispersal in flushing citrus orchards.

BACKGROUND: Processed kaolin is a promising tactic that can be utilized for managing Diaphorina citri Kuwayama infestation. However, the frequency of kaolin application to protect citrus trees during flushing remains undetermined. The objective of this study was to ascertain the frequency of kaolin application (2%) required to reduce D. citri infestation and dispersal in flushing citrus orchards and to measure the spectral reflectance of kaolin-sprayed leaves. RESULTS: Trees sprayed with kaolin at 7- and 14-day intervals showed an 80% psyllid reduction compared to untreated trees. In addition

Efficacy of CC traps and seasonal activity of adult Bemisia argentifolii (Homoptera: Aleyrodidae) in Imperial and Palo Verde Valleys, California.

Adult whitefly Bemisia argentifolii Bellows & Perring trap (CC trap) catches were compared with suction type trap catches. CC trap catches were significantly correlated to the suction trap catches. Higher numbers of B. argentifolii adults were caught in CC traps oriented toward an untreated, B. argentifolii-infested, cotton field as compared with traps oriented toward Bermuda grass fields, farm roads, or fallow areas. CC trap catches at five heights above ground (from 0 to 120 cm) were significantly related to each other in choice and no-choice studies. CC trap catches were low in the Imperial

Survey of Parasitic Bacteria in Bat Bugs, Colorado.

Bat bugs (Cimex adjunctus Barber) (Hemiptera: Cimicidae) collected from big brown bats (Eptesicus fuscus Palisot de Beauvoir) in Colorado, United States were assessed for the presence of Bartonella, Brucella, and Yersinia spp. using molecular techniques. No evidence of Brucella or Yersinia infection was found in the 55 specimens collected; however, 4/55 (7.3%) of the specimens were positive for Bartonella DNA. Multi-locus characterization of Bartonella DNA shows that sequences in bat bugs are phylogenetically related to other Bartonella isolates and sequences from European bats.

Protocol for in vivo tracking of lipoproteins in developing oocytes of multiple species of Chagas disease vectors using fluorescent probes.

In insects, the circulating lipoproteins vitellogenin and lipophorin function as yolk protein precursors during oogenesis. Here, we present a protocol for tracing vitellogenin and lipophorin pathways during egg development in three species of Chagas disease vectors (Triatominae, Hemiptera: Reduviidae). We describe steps for purifying and fluorescent labeling of lipoproteins, their injection into vitellogenic females, and analysis using confocal microscopy. This approach enables the fate of lipoprotein particles and their lipid cargo in ovarian tissues to be tracked at the subcellular level. Fo…

Emergent Periodical Cicada Nymphs Use Skototaxis to Navigate to Trees.

AbstractAfter 13 or 17 years underground, periodical cicada nymphs come aboveground to molt into adulthood. Observers have long noted the strikingly directional orientation of nymphs toward trees on which they molt and have speculated on possible mechanisms to account for these movements. Through a combination of observational, experimental, and modeling studies, we demonstrate that cicada nymphs use skototaxis, or movement toward darkness, to reach their targets. Emergent nymphs walked directly toward isolated trees, regardless of initial compass direction or time of day, traveling only 15% f

New Host Plant Species of Grapevine Virus A Identified with Vector-Mediated Infections.

Grapevine virus A (GVA) is an economically important virus and a member of the genus Vitivirus (family Betaflexiviridae) that causes a range of symptoms with qualitative and quantitative effects on grape production. Wild and domesticated species of Vitis, including hybrids used as rootstocks, are considered important natural hosts of GVA. Mechanical transmission to some herbaceous plant species, graft transmission, and vector transmission from grape to grape by various mealybugs and soft scale insects have been reported. Under laboratory and greenhouse conditions, this study demonstrates the t

Chromosome-level genome assembly of the cottony cushion scale Icerya purchasi.

The cottony cushion scale, Icerya purchasi, a polyphagous pest, poses a significant threat to the global citrus industry. The hermaphroditic self-fertilization observed in I. purchasi is an exceptionally rare reproductive mode among insects. In this study, we successfully assembled a chromosome-level genome sequence for I. purchasi using PacBio long-reads and the Hi-C technique, resulting in a total size of 1,103.38 Mb and a contig N50 of 12.81 Mb. The genome comprises 14,046 predicted protein-coding genes, with 462,722,633 bp occurrence of repetitive sequences. BUSCO analysis revealed a compl

Phototoxicity of Ultraviolet-A against the Whitefly Bemisia tabaci and Its Compatibility with an Entomopathogenic Fungus and Whitefly Parasitoid.

Ultraviolet (UV) radiation significantly affects insect life and, as a result, has been widely used to control different invertebrate pests. The current results demonstrate that when Bemisia tabaci first instar nymphs are exposed to UV-A light for 12, 24, 48, and 72 h, their developmental and biological parameters are negatively affected by UV-A exposure; the effect increased with an increase in exposure time. We hypothesized that UV-A light is compatible with other biological control agents. Results showed that when the entomopathogenic fungus Cordyceps fumosorosea was applied to third instar

Peptidomics Approaches for the Identification of Bioactive Molecules from Diaphorina citri.

Huanglongbing (HLB), a deadly citrus disease, is primarily associated with Candidatus Liberibacter asiaticus (CLas) and spread by the hemipteran insect Diaphorina citri. Control strategies to combat HLB are urgently needed. In this work, we developed and compared workflows for the extraction of the D. citri peptidome, a dynamic set of polypeptides produced by proteolysis and other cellular processes. High-resolution mass spectrometry revealed bias among methods reflecting the physiochemical properties of the peptides: while TCA/acetone-based methods resulted in enrichment of C-terminally amida…

Comparative genomics and metabolic model analysis of primary and secondary endosymbionts present in different species of whitefly.

Candidatus Portiera aleyrodidarum (hereafter Portiera) is the primary endosymbiont of whiteflies (Hemiptera: Aleyrodidae), residing in specialized bacteriocytes that also co-localize diverse secondary endosymbionts. This study aimed to carry out in-depth genomic analysis of different strains of Portiera and secondary endosymbionts namely Arsenophonus, Cardinium, Hamiltonella, Rickettsia, and Wolbachia obtained from different whitefly species (data obtained from NCBI). Interestingly, hierarchical clustering of the amino acid identity data resulted in a dendrogram that resolved the different str…

Together forever: patterns and strategies of vertical symbiont transmission in planthoppers.

Insects host a wide variety of microbial symbionts, including bacteria and fungi, which contribute to their survival by supplementing essential nutrients and aiding in adaptation to diverse ecological niches. The mode of symbiont transmission between generations, particularly vertical transmission, plays a central role in maintaining these mutualistic relationships. In this review, we focus on the transmission mechanisms of symbionts in planthoppers (Fulgoromorpha), a group of sap-feeding insects that rely heavily on obligate, nutritional symbionts for amino acid biosynthesis. These symbionts

Harnessing Lecanicillium attenuatum: A novel strategy for combatting Nilaparvata lugens in rice fields.

Nilaparvata lugens is a notorious rice pest causing significant annual yield and economic losses. The use of entomopathogenic fungi offers a promising and eco-friendly approach to sustainable pest management programs. However, research in this area is currently limited to a few specific types of insects and other arthropods. This study aimed to analyze the biocontrol potential of Lecanicillium attenuatum against N. lugens. Bioassays showed that L. attenuatum 3166 induced >80% mortality in N. lugens following 7 d exposure. Greenhouse and field investigations demonstrated that L. attenuatum 3166

On the morphological variability of Limotettix (Limotettix) striola (Falln, 1806) (Homoptera: Auchenorrhyncha: Cicadellidae: Deltocephalinae: Limotettigini) with notes on the taxonomic status and distribution of L. striola ferganensis Dubovskiy, 1966.

Investigation of the male calling signals of Limotettix (Limotettix) striola (Falln, 1806) in populations from central and southern European Russia, Northern Caucasus, and West Tien Shan, Kyrgyzstan demonstrated that they belong to the same species. Males of L. striola from West Tien Shan have a somewhat different penis shape and belong to a separate subspecies, L. striola ferganensis Dubovskiy, 1966. L. striola ferganensis from West Tien Shan differs from L. ferganensis sensu Hamilton (1983, 1994) from the Nearctic in the shape of penis in dorsal view. Apparently, L. ferganensis sensu Hamilto
