Transmissible cancers are malignant cell lineages that spread clonally between individuals. Several such cancers, termed bivalve transmissible neoplasia (BTN), induce leukemia-like disease in marine bivalves. This is the case of BTN lineages affecting the common cockle, Cerastoderma edule, which inhabits the Atlantic coasts of Europe and northwest Africa. To investigate the evolution of cockle BTN, we collected 6,854 cockles, diagnosed 390 BTN tumors, generated a reference genome and assessed genomic variation across 61 tumors. Our analyses confirmed the existence of two BTN lineages with hemo

A tumor was found for the first time in a mussel Mytilus trossulus from a heavily polluted area of Nakhodka Bay, Sea of Japan. Tumor cells were found in the connective tissue of different organs and also in gill vessels and hemal sinuses of the visceral mass. They were both attached and diffuse. The tumor was at an advanced stage, replacing the normal connective tissue cells, and formed nodes. The tumor cells were polymorphic, with a high nucleocytoplasmic ratio, and had a prominent nucleolus. The size of their nuclei was three to five times that of the nuclei of agranular hemocytes. The mitot

Drosophila has highly efficient defenses against infection. These include both cellular immune responses, such as the phagocytosis of invading microorganisms, and humoral immune responses, such as the secretion of antimicrobial peptides into the hemolymph [1] [2]. These defense systems are thought to interact, but the nature and extent of these interactions is not known. Here we describe a method for inhibiting phagocytosis in Drosophila blood cells (hemocytes) by injecting polystyrene beads into the body cavity. This treatment does not in itself make a fly susceptible to Escherichia coli infe

UNLABELLED: A dynamic homeostasis is maintained between the host and native bacteria of the gastrointestinal tract in animals, but migration of bacteria from the gut to other organs can lead to disease or death. Enterococcus faecalis is a commensal of the gastrointestinal tract; however, Enterococcus spp. are increasingly frequent causes of nosocomial infections with a high mortality rate. We investigated the commensal-to-pathogen switch undergone by E. faecalis OG1RF in the lepidopteran model host Manduca sexta associated with its location in the host. E. faecalis persists in the harsh midgut

Heteroaggregates (HAs) formed by nanoplastics (NPs) and microalgae occur ubiquitously in natural aquatic systems. However, their influence on the toxicokinetics and biological effects of NPs in marine mussels remains largely unknown. Here, the green mussels (Perna viridis) were exposed to NPs and HAs at their environmentally relevant concentrations for 21 d, followed by a 7-d depuration phase. The effects on toxicokinetics, immunological responses, and microbiota of digestive gland were evaluated. The results showed that HAs increased the uptake rate constant in digestive gland by 5.5-fold and

The prevalence of pharmaceutical residues like diltiazem in environments raises concerns over their potential threat to non-target organisms. While the immune system poses as a potential target, little is known about the immunotoxicity of diltiazem to aquatic species such as bivalve molluscs. In this study, the binding affinity of diltiazem to the calcium channels of several aquatic species was evaluated by molecular docking. Taking blood clam as a representative, the impacts of diltiazem on Ca2+-homeostasis and immune parameters were also assessed. Our results illustrated diltiazem exhibit a

Echinoderms are important marine organisms that live in a wide range from the intertidal zone to the abyssal zone. Members of this phylum are prone to dramatic population fluctuations that may trigger dramatic shifts in ecosystem structure. Despite the extremely complex nature of the marine environment, the immune systems of echinoderms induce a complex innate immune response to prokaryotic and eukaryotic pathogens. Previous studies showed that many echinoderm disease outbreaks were associated with specific bacteria, whereas recent scientific investigations using newly developed technologies r

Spindle defect and chromosome misalignment occuring in oocyte meiosis induce nondisjunction. Nondisjunction causes Down syndrome, also known as trisomy 21. Folic acid (FA) is an essential nutrient composition for fetal growth and development. It has been reported that FA nutritional status is associated with the risk of Down syndrome. However, to our knowledge, little is known about the effect of FA deficiency on abnormal oocytes (spindle defects, chromosome misalignments and immature oocyte) in vivo. In the present study, we investigate the effects of FA deficiency on oocyte meiosis in female

Coxiella burnetii is a Gram-negative intracellular bacterium and is the causative agent of the zoonotic disease Q fever. Several rodent and non-human primate models of virulent phase I C. burnetii [Nine Mile (NM)I] have been developed, and have been used to determine the efficacy of antibiotics and vaccine candidates. However, there are several advantages to using insect models to study host-microbe interactions, such as reduced animal use, lowered cost and ease of manipulation in high containment. In addition, many laboratories use the avirulent phase II C. burnetii clone (NMII) to study cell

Lack of shrimp cell lines has hindered the study of pollutants which adversely affects shrimp health and its export value. In this context a primary haemocyte culture developed from Penaeus monodon was employed for assessing the cytotoxicity and genotoxicity of two heavy metal compounds, cadmium chloride and mercuric chloride and two organophosphate insecticides, malathion and monocrotophos. Using MTT assay 12 h IC(50) values calculated were 31.09 ± 16.27 μM and 5.52 ± 1.16 μM for cadmium chloride and mercuric chloride and 59.94 ± 52.30 mg l(-1) and 186.76 ± 77.00 mg l(-1) for malathion and mo

The roles of the amebocyte in providing hemostasis and controlling infection, and its reaction to endotoxin, suggest that the response of platelets and the blood coagulation system in various mammals to gram-negative infection or endotoxin is an evolutionary remnant of this ancient mechanism. In humans, this mechanism occasionally subverts its presumed protective function by overresponding in a manner that results in pathophysiologic thrombosis or hemorrhage. (In this regard, it is interesting that human platelets are much more resistant to the effects of bacterial endotoxins than are other sp

This study investigates the prolonged effect of immune disease resistance in Litopenaeus vannamei through the administration of tyramine (TA) formulated with polyethylene glycol (PEG). Facing the challenges of intensive farming, environmental stress, and global climate changes, innovative approaches to improve shrimp health are essential. The research focuses on the role of biogenic amines in stress response and immune regulation, demonstrating that TA, especially when combined with PEG, significantly prolongs immunity and resistance against Vibrio alginolyticus. The experimental design includ

Phagocytosis by hemocytes, Drosophila macrophages, is essential for resistance to Streptococcus pneumoniae in adult flies. Activated macrophages require an increased supply of energy and we show here that a systemic metabolic switch, involving the release of glucose from glycogen, is required for effective resistance to S. pneumoniae. This metabolic switch is mediated by extracellular adenosine, as evidenced by the fact that blocking adenosine signaling in the adoR mutant suppresses the systemic metabolic switch and decreases resistance to infection, while enhancing adenosine effects by loweri

Outbreaks of infectious diseases have resulted in high mortality and huge economic losses in penaeid shrimp culture. Interest in understanding shrimp immunity has increased because of its importance in disease control. Here we report cDNA cloning of a Toll receptor from the white shrimp Litopenaeus vannamei. L. vannamei Toll (lToll) is 926 residues, with a putative signal peptide of 19 residues. The protein contains distinct structural/functional motifs of the Toll like receptor (TLR) family, including an extracellular domain containing 16 leucine-rich repeats (LRRs) flanked by cysteine-rich m

Immunostimulation through dietary interventions opened new avenues in developing disease control and prevention tools for shrimp aquaculture. We have previously shown that feeding with freeze-dried Lactiplantibacillus plantarum (LAB) increased disease resistance of Penaeus vannamei against both Vibrio parahaemolyticus and white spot syndrome virus (WSSV) based on bulk RNA sequencing of shrimp gills. This tissue participates in ion transport and serves as a first line of defense against environmental stressors and pathogenic infections. However, characterization of their cell composition and fu

While nanoplastics (NPs, <1 µm) concentration is anticipated to increase in the marine environment, the analytical tools for NP quantification and data on their impacts are still scarce. This work aimed to expand knowledge regarding bioaccumulation and effects of NPs on the sentinel organism Mytilus galloprovincialis. For this, mussels were dietarily exposed for 7 days to model palladium-doped-polystyrene NPs (Pd-PS NPs) of 142.6 ± 1.0 nm at 1 × 109 Pd-PS NPs/mL. ICP-MS analysis on water samples showed that after one day of exposure no Pd was detected suggesting that mussels cleared the NPs. A

The spoilage bacterium Aeromonas veronii poses increasing risks to the safety of edible bivalves, yet the regulatory mechanisms of its key spoilage factors remain largely unknown. In this study, a foodborne pathogen A. veronii GL2 was isolated from edible Hyriopsis cumingii and its genomic sequencing was conducted, in which the mRNA level of metalloprotease-encoded gene htpX was significantly upregulated under thermal processing. A htpX-defected mutant (ΔhtpX) was then constructed to investigate its role in virulence and spoilage potentials. Although htpX deficiency did not affect growth or pr

BACKGROUND: Octopamine, the functional equivalent of noradrenaline in invertebrates, is a key neuromodulator that also influences immune functions. However, the receptor‑specific roles of octopamine in regulating innate immunity in Drosophila melanogaster remain incompletely understood.
METHODS: Using loss‑of‑function mutants for the two major octopamine receptors Octβ1R and Octβ2R, combined with systemic bacterial infection models, bacterial clearance assays, in vivo and in vitro phagocytosis assays, and quantification of antimicrobial peptide (AMP) gene expression, we dissected the contribu
