Substance P counteracts neurotoxin damage on norepinephrine neurons in rat brain during ontogeny.

Systemic treatment of newborn rats with the catecholamine neurotoxin 6-hydroxydopamine alters the postnatal development of the central norepinephrine neurons. The changes are permanent and consist of denervation of distant nerve terminal projections (for example, cerebral cortex) and hyperinnervation of terminal areas close to the cell bodies (for example, cerebellum). Intracisternal injection of substance P counteracted both of these alterations. The results indicate that substance P may prevent degeneration of damaged norepinephrine neurons during ontogeny or may have a regrowth stimulatory

Measurement of catecholamines in biological fluids by high-performance liquid chromatography: a comparison of fluorimetric with electrochemical detection.

An improved method for the determination of catecholamines in biological fluids, by reversed-phase high-performance liquid chromatography (HPLC) with fluorimetric detection is presented. The pH titration previously employed in the alumina extraction was abandoned in favour of the use of a molar excess of PH 8.5 Tris--HCl buffer. A novel lyophilisation step serves to concentrate the catechols and by reconstituting in mobile phase, chromatography disturbances are minimised. The addition of 2 mM octanesulphonic acid to citrate--phosphate mobile phase at pH 6.0 gave optimal resolution and sensitiv

Localization of the brain regions concerned in the inhibition of shivering by trauma.

The effect on the inhibition of shivering by limb ischaemia of small 6-hydroxydopamine (6-OHDA) lesions in the hypothalamus has been studied in rats. The injection of 6-OHDA into the posterior part of the hypothalamus in the neighbourhood of the N. dorsomedialis or into the ventral ascending catecholaminergic bundle caudal to that nucleus prevented the depression of the ambient temperature threshold for the onset of shivering which usually occurs during hind-limb ischaemia and lowered the slope of the regression line relating the intensity of shivering to ambient temperature. In rats not treat

Central and peripheral noradrenaline in the two kidney model of renovascular hypertension in the rat.

1 Plasma noradrenaline concentrations were similar in rats following unilateral renal arterial constriction (two kidney Goldblatt model) and in sham-operated control rats. 2 The development of hypertension was not affected by pretreatment with intracisternal injections of 6-hydroxydopamine. 3 These data suggest that sympathetic mechanisms do not contribute to the development of hypertension in this model.

Partial selectivity of 6-OHDA induced neuronal degeneration after intraissular injection in the brain with special reference to the nigro-striatal dopamine system.

The selectivity of 6-OHDA induced neuronal degeneration after intra-tissular injection in the brain is discussed considering the morphological and biochemical events occurring in the substantia nigra and in the striatum--after intranigral or intrastriatal application of the drug. The data suggest that a critical intratissular concentration of 6-OHDA is required to induce an elective neuronal degeneration. The importance of the non-specific damage can not be neglected when high doses of the drug are administered in the brain tissue.

Catha edulis extract and its active principle cathinone induce ipsilateral rotation in unilaterally lesioned rats.

Catha edulis extract, a natural psychostimulant, and its active component, S-(-)-cathinone, were evaluated for their rotational effects in unilaterally lesioned rats. In our earlier study, we demonstrated that commercially available S-(-)-cathinone, the active principle of C. edulis extract, reverses haloperidol-induced catalepsy. In the current study, we evaluated the effect of C. edulis extract or its active principle, cathinone, in 6-hydroxydopamine unilaterally lesioned rats. Evaluation of the rotational behaviour induced by this natural psychostimulant was made upon acute and repeated ora

Sympathetic innervation in the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine primate model of Parkinson's disease.

Cardiac sympathetic denervation occurs commonly in Parkinson's disease. This study explored whether analogous denervation occurs in primates with Parkinsonism from systemic administration of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). 6-[18F]Fluorodopamine positron emission tomographic scanning and plasma levels of catecholamines and their deaminated metabolites were used to assess sympathetic and adrenomedullary function in rhesus monkeys, in the untreated state (n = 3), 2 weeks after a series of four MPTP injections, before establishment of Parkinsonism (acute phase, n = 1); a month

Antiparkinsonian potential of interaction of LEK-8829 with bromocriptine.

The ergoline derivative, LEK-8829 (9,10-didehydro-N-methyl-(2-propynyl)-6-methyl-8-aminomethylerg oline), has been proposed as a potential atypical antipsychotic drug with antagonistic actions at dopamine D2 and serotonin 5-HT2 and 5-HT1A receptors (Krisch et al., 1994, 1996). LEK-8829 also induces contralateral turning in rats with 6-hydroxydopamine-induced unilateral lesion of dopamine nigrostriatal neurons. Turning is blocked by SCH-23390 (R(+)-chloro-8-hydroxy-3-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzaze pine), a dopamine D1 receptor antagonist. It has been suggested that LEK-8829 cou

Protective effects of silk fibroin against 6-OHDA in SH-SY5Y human neuroblastoma cells and comparative study with its release from gelatin films.

In this study, we analyzed the neuroprotective action of silk fibroin (SF) regenerated with calcium chloride (CaCl2), distinguishing the effects of CaCl2 and SF, and subsequently fabricating a neuroprotective hybrid material based on SF gelatin film. Cytotoxicity induced by 6-hydroxydopamine (6-OHDA) on the human neuroblastoma SH-SY5Y cell line showed that SF had a significant shielding power against 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in SH-SY5Y neuroblastoma cells, as assessed by the CCK-8 assay, cell imaging and cell cycle using flow cytometer. Specifically, the concurrent trea

Rewarding properties of the stereoisomers of 4-methylaminorex: involvement of the dopamine system.

4-Methylaminorex is a potential psychostimulant drug of abuse that exists as four stereoisomers: cis-4R,5S, cis-4S,5R, trans-4S,5S, and trans-4R,5R. The racemic mixture of the cis-isomers has been encountered in illicit samples, but previous animal studies suggest that also the trans-isomers could have similar stimulant-like properties. We tested whether the stereoisomers possess rewarding properties and compared their potency using the conditioned place preference method in rats. Furthermore, the involvement of the brain dopaminergic system in the 4-methylaminorex reward was tested with the d

Centrally acting non-narcotic antitussives prevent hyperactivity in mice: Involvement of GIRK channels.

We have previously reported that centrally acting non-narcotic antitussives inhibited G protein-coupled inwardly rectifying potassium (GIRK) channel-activated currents, and that the antitussives had multiple pharmacological actions on various models of intractable brain diseases in rodents. In this study, the question of whether these antitussives inhibit drug-induced hyperactivity in mice was investigated. Antitussives, such as cloperastine and tipepidine, at cough suppressant doses, inhibited an increase in ambulation of mice neonatally treated with 6-hydroxydopamine. In addition, all antitu

Behavioral and neurochemical dysfunction in the circling (ci) rat: a novel genetic animal model of a movement disorder.

One of the crucial breakthroughs in research on parkinsonism was the observation of circling behaviour in rodents after unilateral intranigral injection of 6-hydroxydopamine. This Ungerstedt model remains one of the basic animal models of Parkinson's disease. We report here the first mutant rat strain with abnormal circling behaviour and several other features reminiscent of the Ungerstedt Parkinson model. The neurological disorder in the novel mutant rat strain is determined monogenetically by a recessive autosomal gene termed circling (ci). Mutant rats of both genders exhibit an intense asym

Complex motor disturbances in a sequential double lesion rat model of striatonigral degeneration (multiple system atrophy).

This study characterizes paw reaching, stepping and balance abnormalities in a double lesion rat model of striatonigral degeneration, the core pathology underlying levodopa unresponsive parkinsonism associated with multiple system atrophy. Extensive unilateral nigral or striatal lesions induced by 6-hydroxydopamine or quinolinic acid, respectively, produced a similarly marked contralateral paw reaching deficit without further deterioration following a secondary (complementary) lesion of ipsilateral striatum or substantia nigra. Contralateral stepping rates were reduced by unilateral 6-hydroxyd

The effect of selective lesions of brain noradrenergic systems on the development of barbiturate tolerance in rats.

A method for chronic infusion of barbiturates into the central nervous system of rats was developed and used to study the importance of noradrenergic systems in the development of barbiturate tolerance. Destruction of noradrenergic neurons by intraventricular administration of 6-hydroxydopamine or by specific lesions of the dorsal or ventral noradrenergic bundles prevented the development of barbiturate tolerance without altering the animal's response to the acute administration of barbiturate.

Engineering Bacopa monnieri for improved bacoside content and its neurological evaluation.

Bacosides are triterpenoidal saponins with numerous pharmacological benefits. One of the significant drawbacks is the low availability of these bacosides. The bacoside pathway is not well elucidated, and there is no prior report of a metabolic engineering approach in this plant. In this study, we have over-expressed the active isoform of Bacopa monnieri squalene synthase (BmSQS1-OE) and silenced the B. monnieri G10H (BmG10H-1-KD), the competitive metabolic pathway, to divert the flux towards triterpene biosynthesis. Absolute quantification of bacosides in these BmSQS1(OE)-BmG10H1(KD) lines has

Arterial baroreflex dysfunction fails to mimic Parkinson's disease in rats.

Patients with Parkinson's disease (PD) often have attenuated baroreflex function, which may occur before the onset of PD-associated movement disorders. The aim of the present study was to test whether impaired arterial baroreflex (ABR) function could contribute to the pathogenesis of PD. 6-Hydroxydopamine (8 mug in 4 mul) was microinjected into the left substantia nigra of rats to establish unilateral PD models, and bilateral PD models were established in rats by administration of rotenone by osmotic minipump for four weeks, at a dose of 2.5 mg . kg(-1) . day(-1). An ABR dysfunction model was

Can the brain be protected through exercise? Lessons from an animal model of parkinsonism.

Evidence suggests that following injury the brain has the capacity for self-repair and that this can be promoted through a variety of experiences including motor activity. In their article, Döbrössy and Dunnett have provided further evidence that this is the case in an animal model in which an excitotoxin is applied to the neostriatum. Under standard conditions, such a toxin would cause considerable damage to the GABAergic cells of this region and produce behavioral deficits. This model has been used to explore certain aspects of Huntington's disease, which also involves the loss of these neur

In vitro evidence for increased facilitation of striatal acetylcholine release via pre- and postsynaptic NMDA receptors in hemiparkinsonian rats.

The NMDA-evoked acetylcholine release from striatal slices and synaptosomes was investigated in rats subjected to unilateral injection of 6-hydroxydopamine into the substantia nigra. In slices prepared from the striatum contralateral to the lesion, the NMDA-evoked endogenous acetylcholine release was not significant at 10 microM NMDA and maximal at 100 microM NMDA (124 +/- 19%). Conversely, in slices taken from the dopamine-depleted striatum, NMDA was effective even at 10 microM (41 +/- 4%), and at 100 microM (196 +/- 24%) efficacy was nearly doubled. In synaptosomes prepared from the contrala

Interaction between renin-angiotensin and sympathetic nervous systems in a rat model of pressure overload cardiac hypertrophy.

1 A raised cardiac workload activates neurohormones which will increase muscle mass and shift contractility to the right along the Frank-Starling curve. 2 This study examined the interaction between the SNS and RAS in contributing to vascular responsiveness following the development of cardiac hypertrophy due to aortic banding. 3 Sprague Dawley rats (180-200 g) were assigned to one of six groups; Normal, Sham-operated, Aortic Banded (AB), Aortic Banded treated with losartan (ABLOS), Aortic Banded treated with 6-hydroxydopamine (ABSYMP) and Aortic banded treated with both losartan and 6-hydroxy
