Developing a universal multi-epitope protein vaccine candidate for enhanced borna virus pandemic preparedness.

INTRODUCTION: Borna disease virus 1 (BoDV-1) is an emerging zoonotic RNA virus that can cause severe acute encephalitis with high mortality. Currently, there are no effective countermeasures, and the potential risk of a future outbreak requires urgent attention. To address this challenge, the complete genome sequence of BoDV-1 was utilized, and immunoinformatics was applied to identify antigenic peptides suitable for vaccine development. METHODS: Immunoinformatics and antigenicity-focused protein screening were employed to predict B-cell linear epitopes, B-cell conformational epitopes, and cy…

Design and computational evaluation of a cross-protective multi-epitope vaccine candidate against Bartonella henselae and Bartonella clarridgeiae pathogens causing the zoonotic cat scratch disease in humans.

Cat scratch disease (CSD), primarily caused by Bartonella henselae and Bartonella clarridgeiae, presents a global zoonotic concern, particularly in immunocompromised individuals. Conventional antibiotics offer limited protection, necessitating novel preventive strategies. Also, CSD is still one of the most prevalent infections brought on by Bartonella genus. The current study aimed at developing a multi-epitope peptide vaccine by targeting conserved antigenic proteins (Pap31, Omp43, and Omp89) from two Bartonella species utilizing immunoinformatics techniques. Comprehensive immunoinformatics a…

Design, development, and assessment of a novel multi-peptide vaccine targeting PspC, PsaA, and PhtD proteins of Streptococcus pneumoniae.

Pneumococcus is the top cause of diseases such as pneumonia/meningitis, and of secondary infections after viral respiratory diseases like COVID-19/flu. Pneumococcal protein-based vaccines consisting of proteins with various functions in virulence might provide a qualified alternative for present vaccines. In this project, PspC, PsaA, and PhtD proteins were considered to anticipate B/T-cell epitopes using immunoinformatics to develop 4 multi-peptide constructs (C, A, and D individual constructs, and a fusion construct CAD). We tested whether vaccination with CAD is able to elicit more efficient…

Agent-based modeling identifies multiple paths for inflammation resolution.

SUMMARY: Sterile inflammation emerges from spatially coordinated interactions between immune recruitment, polarization, signal diffusion, and tissue turnover. Capturing this complexity requires modeling frameworks that integrate stochasticity, spatial heterogeneity, and functional redundancy across immune cell types. We present a spatially explicit agent-based model of sterile inflammation based on a functional mono-agent abstraction, in which immune cells share core capabilities, e.g. polarization, chemotaxis, signal production, and phagocytosis, while differing through parameterization rathe

Systems immunology of transcriptional responses to viral infection identifies conserved antiviral pathways across macaques and humans.

Viral pandemics and epidemics pose a significant global threat. While macaque models of viral disease are routinely used, it remains unclear how conserved antiviral responses are between macaques and humans. Therefore, we conducted a cross-species analysis of transcriptomic data from over 6,088 blood samples from macaques and humans infected with one of 31 viruses. Our findings demonstrate that irrespective of primate or viral species, there are conserved antiviral responses that are consistent across infection phase (acute, chronic, or latent) and viral genome type (DNA or RNA viruses). Engineering a multi epitope vaccine against SARS-CoV-2 by exploiting its non structural and structural proteins.

SARS-CoV-2, the causative agent behind the ongoing pandemic exhibits an enhanced potential for infection when compared to its related family members- the SARS-CoV and MERS-CoV; which have caused similar disease outbreaks in the past. The severity of the global health burden, increasing mortality rate and the emergent economic crisis urgently demands the development of next generation vaccines. Amongst such emergent next generation vaccines are the multi-epitope subunit vaccines, which hold promise in combating deadly pathogens. In this study we have exploited immunoinformatics applications to …

In silico approach to Helicobacter pylori vaccine design.

Helicobacter pylori, a Gram-negative microorganism, infects over half of the human population worldwide. Its extensive genetic variability and capacity to bypass host immune defences continue to impede the creation of an effective vaccine. Nonetheless, contemporary progress in computational biology and immunoinformatics has introduced innovative methodologies that can substantially support and accelerate vaccine design against this pathogen. In the present work, an in silico strategy was employed to develop a multi-epitope vaccine targeting H. pylori. The FlaA, UreB and PSA D15 proteins were c…

Rational design of a multivalent vaccine targeting arthropod-borne viruses using reverse vaccinology strategies.

Viruses transmitted by arthropods, such as Dengue, Zika, and Chikungunya, represent substantial worldwide health threats, particularly in countries like India. The lack of approved vaccines and effective antiviral therapies calls for developing innovative strategies to tackle these arboviruses. In this study, we employed immunoinformatics methodologies, incorporating reverse vaccinology, to design a multivalent vaccine targeting the predominant arboviruses. Epitopes of B and T cells were recognized within the non-structural proteins of Dengue, Zika, and Chikungunya viruses. The predicted epito…

T-cell epitope-based vaccine prediction against Aspergillus fumigatus: a harmful causative agent of aspergillosis.

BACKGROUND: Among the most common causes of invasive aspergillosis and acute bronchopulmonary aspergillosis is Aspergillus fumigatus. Transmission with A. fumigatus produces aggressive aspergillosis in allogeneic haematopoietic stem cell transplant recipients, HIV patients, and cancer patients. Asthmatics and cystic fibrosis patients are allergic to A. fumigatus. MHC class-II binding epitopes can initiate immunogenic responses in patients. In this study, we deployed immunoinformatic study to reveal epitopes from fungal proteins. RESULTS: In modern research, we found multiple epitopes ITLKLLHR

Epitope prediction and immunogenicity assessment of Leishmania donovani TryS protein as a potential vaccine candidate.

Leishmaniasis, a vector-borne tropical disease, is caused by the bite of Leishmania-infected sand flies. Currently, chemotherapy is the only available option because of the absence of any approved vaccine. Vaccines are considered decisive for the complete eradication of the disease. In this study, we have designed a multiepitope-based vaccine and evaluated its potential immunogenicity using immunoinformatic tools. We selected Leishmania donovani trypanothione synthetase (LdTryS) for the development and analysis of potential immunogenic epitopes. A total of 4 CTL, 3 HTL and 2 LBL epitopes were

Recombinant and epitope-based vaccines on the road to the market and implications for vaccine design and production.

Novel vaccination approaches based on rational design of B- and T-cell epitopes - epitope-based vaccines - are making progress in the clinical trial pipeline. The epitope-focused recombinant protein-based malaria vaccine (termed RTS,S) is a next-generation approach that successfully reached phase-III trials, and will potentially become the first commercial vaccine against a human parasitic disease. Progress made on methods such as recombinant DNA technology, advanced cell-culture techniques, immunoinformatics and rational design of immunogens are driving the development of these novel concepts…

Synthetic Peptides Elicit Strong Cellular Immunity in Visceral Leishmaniasis Natural Reservoir and Contribute to Long-Lasting Polyfunctional T-Cells in BALB/c Mice.

Reverse vaccinology or immunoinformatics is a computational methodology which integrates data from in silico epitope prediction, associated to other important information as, for example, the predicted subcellular location of the proteins used in the design of the context of vaccine development. This approach has the potential to search for new targets for vaccine development in the predicted proteome of pathogenic organisms. To date, there is no effective vaccine employed in vaccination campaigns against visceral leishmaniasis (VL). For the first time, herein, an in silico, in vitro, and in v…

Sustained antibody response to a linear epitope of Nipah virus fusion protein in human survivor serum samples.

Nipah virus (NiV), a pathogen with pandemic potential, lacks approved treatments or vaccines, highlighting the urgent need for research on immune-targeted antigenic determinants. A significant gap persists in NiV research, as studies on the fusion (F) protein critical for viral entry as well as the B cells epitopes, have primarily focused on computational prediction rather than experimental validation of immunogenic epitopes obtained through immunoinformatics approach. This study focused on the conserved F protein across NiV human isolates and employed an immunoinformatics approach to predict …
