Reversible and irreversible inhibition of CYP3A enzymes by tamoxifen and metabolites.

1. Preliminary studies have identified cytochrome P450 (CYP) 3A4 and CYP1B1 as the human CYPs inhibited by tamoxifen. To quantify the inhibitory potency of tamoxifen and its major metabolites, the metabolism of three substrates of CYP3A, midazolam, diltiazem and testosterone, and 7-ethoxyresorufin as a substrate of CYP1B1 were examined in catalytic assays carried out using human liver microsomes and cDNA-expression systems. 2. Tamoxifen, N-desmethyltamoxifen, 4-hydroxytamoxifen and 3-hydroxytamoxifen reversibly inhibited midazolam 1'-hydroxylation, diltiazem N-demethylation and testosterone 6b

Pleiotropic functions of CpdB in Bacillus anthracis.

Bacillus anthracis is the causative agent of anthrax disease. Our prior research indicated that cyclic (c)-di-AMP accumulation attenuated bacterial virulence in mice. However, the function of c-di-AMP secretion and its regulatory mechanism in anthrax have not been revealed. We characterized the role of the ecto-nucleotidase CpdB in B. anthracis. CpdB exhibits phosphodiesterase activity towards c-di-AMP and 2'3'-cGMP-AMP, and nucleotidase activity towards several mononucleotides, including the preferred substrates c-di-AMP and pApA. Our results demonstrated that inactivation of cpdB altered pur

Analysis of different fermentation processes of jujube-hawthorn fermented beverage based on untargeted metabolomics and GC-MS.

In this study, we compared two processes: "Lactobacillus single fermentation 36 h→ inactivation→yeast re-fermentation (InLA)" and "Lactobacillus-yeast simultaneous co-fermentation (LSA)," using jujube and hawthorn composite juice as the substrate. We applied non-targeted liquid chromatography tandem mass spectrometry (LC-MS/MS), gas chromatography mass spectrometry (GC-MS), and kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis to evaluate how sterilization affects quality. The results showed that in LSA, the total acid continued to increase, and levels of total phenolics, tota

Impact of Heterotropic Allosteric Modulation on the Time-Dependent Inhibition of Cytochrome P450 3A4.

The current study was designed to investigate the influence of allosteric effectors on the metabolism of the prototypical cytochrome P450 (CYP) 3A4 substrate midazolam (MDZ), and on the determination in vitro time-dependent inhibition (TDI) of CYP3A4 using human liver microsomes (HLM). As the concentration of midazolam increased to 250 µM in HLMs, homotropic cooperativity resulted in a decrease in the 1'-hydroxymidazolam to 4-hydroxymidazolam ratio to a maximum of 1.1. The presence of varying concentrations of testosterone, progesterone (PGS), or carbamazepine (CBZ) in HLMs with MDZ could reca

Deficiency of the Tangier Disease Gene Abca1 Is Associated With Microglial Defects in Mice.

Microglia, the resident macrophages of the central nervous system, are a diverse population that develop during embryonic and postnatal stages in the mouse. Several signalling pathways are involved in their specification and maturation, but other types of cues might be involved, including lipid metabolism. Here, we evaluated the effect of the inactivation of two main cholesterol transporters in mice, ABCA1 and SR-B1, on microglial development. Using public datasets, we showed that both transporters are expressed in microglia and are differentially regulated in neurodegeneration models. Inactiv

Metabolic studies on the possible mode of action of isoniazid tumorigenicity.

Data on tumorigenicity and mutagenicity of INH show that INH is tumorigenic in mice but not in rats. The metabolic studies on the two species denote that rats are rapid inactivators whereas mice are slow inactivators of INH. Rats are also resistant to the immediate inhibitory effect of INH on DNA biosynthesis. Using Ames test it was observed that INH is mutagenic to salmonella typhimurium strains TA 100 and 1535 and this effect is abolished in presence off 59 mixture. In vivo and in vitro studies on INH interaction with macromolecules reveal that there is a greater interaction with RNA than wi

Effect of carbenoxolone on prostaglandin synthesizing and metabolizing enzymes and correlation with gastric mucosal carbenoxolone concentrations.

Carbenoxolone in a dose dependent manner inhibits the activity of the prostaglandin (PG) metabolizing enzymes 15-hydroxy-PG-dehydrogenase and delta 13-PG-reductase in vitro, while this drug in the same dose range does not influence gastric mucosal PG synthesis by a microsomal cell fraction. Using a radioimmunoassay for carbenoxolone determination, we could show that during absorption high levels of the drug are reached within the gastric mucosa of human volunteers and gastric ulcer patients. From the tissue levels reached it seems possible that carbenoxolone inhibits PG inactivating enzymes of

Effects of inactivated Lactobacillus acidophilus on growth performance, blood parameters, fecal fermentation and microbiota in neonatal Holstein calves.

Inactivated probiotics can exhibit beneficial properties similar to live probiotics while offering greater stability and safety. However, the understanding of how inactivated probiotics affect the growth, health, and gut microbiota of neonatal calves is still limited. This study investigated the effect of inactivated Lactobacillus acidophilus (ILA, ≥1 × 108 CFU equivalents/mL) on growth performance, blood parameters, and fecal fermentation and microbiota of neonatal dairy calves. Forty female Holstein calves (37.92 ± 2.04 kg; 2.45 ± 0.96 d) were randomly assigned to a basal diet supplemented w

Real-Time Monitoring of Self-Repair in Single Bacteria after Photodamage by Synchronous Bioluminescence-Autofluorescence Imaging.

Understanding how individual bacteria recover from cellular damage is essential for clarifying the basic principles of self-repair. However, most existing single-cell approaches often rely on extrinsic reporters or invasive methods, which perturb native metabolism and preclude longitudinal kinetic tracking. Here, we introduce a label-free methodology, termed single-bacteria bioluminescence recovery after photobleaching (BRAP), for real-time monitoring of metabolic self-repair in individual luminescent bacteria following defined photodamage. By using intrinsic bacterial bioluminescence (BL) as

Multiple pathways towards reduced membrane potential and concomitant reduction in aminoglycoside susceptibility in Staphylococcus aureus.

Staphylococcus aureus is responsible for life-threatening and difficult-to-treat infections worldwide and antimicrobial resistance is an increasing concern. Whilst acquired resistance has been widely studied, little is known of the contributions from chromosomal determinants that upon inactivation may reduce the susceptibility of S. aureus to antibiotics. The aim of this study was to identify genetic determinants that upon inactivation reduce aminoglycoside susceptibility in S. aureus. The Nebraska Transposon Mutant Library of 1920 single-gene inactivations in S. aureus strain JE2 was screened

Structure of human aldehyde oxidase under tris(2-carboxyethyl)phosphine reducing conditions.

The importance of human aldehyde oxidase (hAOX1) has increased in recent decades due to its involvement in drug metabolism. Inhibition studies involving hAOX1 are extensive and a common reducing agent, dithiothreitol (DTT), was recently found to inactivate the enzyme. However, in previous crystallographic studies of hAOX1, DTT was found to be essential for crystallization. To surpass this concern, another reducing agent was used in crystallization trials. Using tris(2-carboxyethyl)phosphine (TCEP), a sulfur-free reducing agent, it was possible to obtain well ordered crystals of wild-type hAOX1

Critical role of an endogenous gastric peroxidase in controlling oxidative damage in H. pylori-mediated and nonmediated gastric ulcer.

The objective of the present study is to delineate the mechanism of oxidative damage in human gastric ulcerated mucosa despite the presence of some antioxidant enzymes. We report for the first time the critical role of an endogenous peroxidase, a major H(2)O(2) metabolizing enzyme, in controlling oxidative damage in gastric mucosa. Human gastric mucosa contains a highly active peroxidase in addition to the myeloperoxidase contributed by neutrophil. In both non-Helicobacter pylori (H. pylori)- and H. pylori-mediated gastric ulcer, when myeloperoxidase level increases due to neutrophil accumulat

Dietary Modulation of Osteoporosis via Probiotics and the Gut-Bone Axis: From Mechanisms to Food Applications.

Osteoporosis, a metabolic bone disorder affecting nearly one-third of women and one-fifth of men globally, poses a major public health burden. Current pharmacological treatments face limitations, driving interest in safer nutritional interventions. The gut-bone axis, through which gut microbiota modulate bone homeostasis, offers a promising target. This review synthesizes recent evidence on probiotics, including both viable and heat-inactivated strains of Lactobacillus and Bifidobacterium, as modulators of osteoporosis. Probiotics exert integrated protective effects by reinforcing intestinal b

Fibroblast growth factor-23 and phosphorus metabolism.

The understanding of phosphorus metabolism has expanded considerably over the last decade. Recent studies have identified a novel bone-kidney endocrine axis that maintains phosphate homeostasis. When phosphate is in excess, FGF-23 is secreted from bone and acts on the kidney to promote phosphate excretion into urine and to suppress vitamin D synthesis, thereby inducing negative phosphate balance. This review summarizes the role of the FGF-23 axis on phosphorus metabolism, and presents the clinical entities that arise from activation or inactivation of the FGF-23 axis.

[Gas exchange and metabolic functions of the lungs in endotoxic shock].

The experiments were carried out in 27 dogs with endotoxic shock under conditions of the artificial ventilation of the lungs and barbital anesthesia. It was established that under the above conditions the metabolic functions of the lungs were the first to be impaired. The artificial ventilation of the lungs fails to prevent the development of the impairment of the surfactant-synthesizing function of the lungs as well as the serotonin-inactivating function of the lungs. Two stages of the development of the shock lung syndrome were distinguished: the first one is the disturbance of the metabolic

Glyoxylate detoxification is an essential function of malate synthase required for carbon assimilation in Mycobacterium tuberculosis.

The glyoxylate shunt is a metabolic pathway of bacteria, fungi, and plants used to assimilate even-chain fatty acids (FAs) and has been implicated in persistence of Mycobacterium tuberculosis (Mtb). Recent work, however, showed that the first enzyme of the glyoxylate shunt, isocitrate lyase (ICL), may mediate survival of Mtb during the acute and chronic phases of infection in mice through physiologic functions apart from fatty acid metabolism. Here, we report that malate synthase (MS), the second enzyme of the glyoxylate shunt, is essential for in vitro growth and survival of Mtb on even-chain

Inhibition of hepatic drug metabolism by (+)-cyanidanol-3 (Catergen) in chronic alcoholic liver disease.

There are some controversies regarding the effect of (+)-cyanidanol-3 on hepatic drug metabolism. In the present study the effect of (+)-cyanidanol-3 was investigated on the microsomal and mitochondrial enzyme functions (hydroxylation, glucuronide production, D-glucaric acid excretion and acetylation) in alcoholic liver disease. Eight patients with biopsy-proven chronic alcoholic hepatitis were investigated before and after a 10-day, as well as after a further 30-day, treatment with (+)-cyanidanol-3 (Catergen), 3000 mg/day orally. Besides biochemical liver function tests (serum bilirubin level

Cyanide in human disease: a review of clinical and laboratory evidence.

Experimental cyanide exposure in animals causes demyelination and circumstantial clinical and laboratory evidence suggest that there are human parallels. In Leber's hereditary optic atrophy there appears to be a defect in the conversion of cyanide to thiocyanate because of deficient rhodanese activity. For transmitters of the disease smoking carries the risk of blindness and in the most severely affected patients, there is diffuse neurological disease. It is possible that other hereditary optic atrophies (dominant and recessive) may also reflect inborn errors of cyanide metabolism. In the retr

Reengineering Escherichia coli for Succinate Production in Mineral Salts Medium.

The fermentative metabolism of glucose was redirected to succinate as the primary product without mutating any genes encoding the native mixed-acid fermentation pathway or redox reactions. Two changes in peripheral pathways were together found to increase succinate yield fivefold: (i) increased expression of phosphoenolpyruvate carboxykinase and (ii) inactivation of the glucose phosphoenolpyruvate-dependent phosphotransferase system. These two changes increased net ATP production, increased the pool of phosphoenolpyruvate available for carboxylation, and increased succinate production. Modest

The role of non-coding RNA in ferroptosis of liver cancer and its impact on lipid peroxidation.

Ferroptosis is an iron-dependent programmed death caused by the imbalance of lipid peroxides in cells. Unlike apoptosis, autophagy and necrosis, ferroptosis is mainly induced by the small molecule compound erastin. The main characteristics of ferroptosis were glutathione (GSH) depletion, inactivation of glutathione peroxidase 4 (GPX4) and reactive oxygen species (ROS) promoting lipid peroxidation. Eventually, the imbalance of lipid peroxidation regulation in cells leads to ferroptosis. The lipid metabolic pathway ultimately contributes to ferroptosis through the production of lipid peroxides.
