Role of ocular albinism type 1 (OA1) GPCR in Asian gypsy moth development and transcriptional expression of heat-shock protein genes.

The ocular albinism type 1 gene, named OA1, is a coding pigment cell-specific G protein-coupled receptor exclusively localized in intracellular organelles. However, the function of OA1 in insects remains generally unknown. In the present study, we explore for the first time the function of LdOA1 in the Asian gypsy moth, Lymantria dispar. To identify the function of LdOA1 gene in the development and growth of the Asian gypsy moth, the LdOA1 gene in third instar larvae was knocked down by RNAi. Compared with the controls, the knockdown of LdOA1 increased larval mortality but did not significantl

Orthosteric muscarinic receptor activation by the insect repellent IR3535 opens new prospects in insecticide-based vector control.

The insect repellent IR3535 is one of the important alternative in the fight against mosquito-borne disease such as malaria, dengue, chikungunya, yellow fever and Zika. Using a multidisciplinary approach, we propose the development of an innovative insecticide-based vector control strategy using an unexplored property of IR3535. We have demonstrated that in insect neurosecretory cells, very low concentration of IR3535 induces intracellular calcium rise through cellular mechanisms involving orthosteric/allosteric sites of the M1-muscarinic receptor subtype, G protein βγ subunits, background pot

A serine-rich salivary protein facilitates brown planthopper feeding and survival by mediating salivary sheath formation.

The piercing-sucking pest, brown planthopper (Nilaparvata lugens, BPH), poses a severe and persistent threat to rice in China and across Asia. During feeding, BPHs secrete salivary proteins to form salivary sheaths, which are crucial for successful and sustained phloem sap ingestion. These salivary sheath proteins are crucial for rice-BPH interactions and hold promise as targets for pest control strategies like RNAi. However, the specific functions of salivary proteins in sheath formation remain largely unclear. In this study, we identify NlSRP, a serine-rich salivary protein in BPH, as a nove

Integrated Venom Gland Transcriptomic and Venom Proteomic Analyses of the Digger Wasp Cerceris japonica.

Digger wasps are solitary apoid wasps that excavate ground nests and use venom to paralyze insect prey. Recent phylogenetic analyses suggest that digger wasps belong to an ancient lineage of aculeate Hymenoptera and share an evolutionary ancestry with other venomous hymenopterans, including bees and ants. Although the venoms of ants, bees, and social wasps have been extensively studied, those of digger wasps remain poorly characterized in terms of their composition, molecular diversity, and biological activity. In this study, we investigated the venom of the digger wasp Cerceris japonica using

A transgenic approach to control hemipteran insects by expressing insecticidal genes under phloem-specific promoters.

The first generation transgenic crops used strong constitutive promoters for transgene expression. However, tissue-specific expression is desirable for more precise targeting of transgenes. Moreover, piercing/sucking insects, which are generally resistant to insecticidal Bacillus thuringiensis (Bt) proteins, have emerged as a major pests since the introduction of transgenic crops expressing these toxins. Phloem-specific promoters isolated from Banana bunchy top virus (BBTV) were used for the expression of two insecticidal proteins, Hadronyche versuta (Blue Mountains funnel-web spider) neurotox

Transcriptional, translational, and physiological signatures of undernourished honey bees (Apis mellifera) suggest a role for hormonal factors in hypopharyngeal gland degradation.

Honey bee colonies function as a superorganism, where facultatively sterile female workers perform various tasks that support the hive. Nurse workers undergo numerous anatomical and physiological changes in preparation for brood rearing, including the growth of hypopharyngeal glands (HGs). These glands produce the major protein fraction of a protein- and lipid-rich jelly used to sustain developing larvae. Pollen intake is positively correlated with HG growth, but growth in the first three days is similar regardless of diet, suggesting that initial growth is a pre-determined process while later

Multifunctional properties of peptides derived from black cricket (Gryllus assimilis) and effects of in vitro digestion simulation on their bioactivities.

Insects are a rich source of proteins and are produced in systems that have lower environmental impact. As an alternative protein source, they can be consumed directly or used as an ingredient in other formulations. Recently, there has been growing interest in utilizing insect proteins as a substrate to obtain bioactive peptides as well as in investigating the maintenance of their biological properties under physiological conditions. This study aimed to evaluate the impact of simulated digestion on the bioactive properties of protein hydrolysates from black crickets (Gryllus assimilis). Follow…

Expression of Chemosensory Genes in Larval and Adult Cephalic Appendages in a Diving Beetle.

The impact of land-to-water transition on chemosensory genes has been explored in marine tetrapod vertebrates, with scarce data on aquatic insect lineages. Diving beetles (Dytiscidae) are predaceous freshwater insects with strictly aquatic larvae and amphibious adults. Using RNA-seq, we compared the expression of odorant receptors (ORs), gustatory receptors (GRs), ionotropic receptors (IRs), and odorant-binding proteins (OBPs) in the cephalic appendages of larval and adult Cybister lateralimarginalis. Overall, larvae expressed fewer chemosensory genes than adults but larva-specific genes displ

The Role of Lipids in Development of Allergic Responses.

Most allergic diseases are caused by activation of Th2 type immune responses resulting in the production of specific IgE against proteins found in normally harmless substances such as pollen, mites, epithelia or food. Allergenic substances are composed, in addition to proteins, of other compounds such as carbohydrates and lipids. Those lipids are able to promote the development of Th2-type responses associated with allergy. There are lipids found in pollen, milk or insect venom that are specifically recognized by CD1 restricted unconventional T lymphocytes, which can promote allergic reactions

Sequencing and immunochemical characterization of the American cockroach per a 3 (Cr-PI) isoallergenic variants.

Two additional members of the American cockroach (Periplaneta americana) Per a 3 (Cr-PI) allergen, C13 and C28, were isolated and sequenced. They encoded proteins of 470 and 393 amino acids with two and no potential N-glycosylation sites, respectively. The molecular weights for C13 and C28 cloned proteins are 56,200 and 46,7000, with PI values of 7.06 and 6.54. C13 and C28 display 95.4% identity with several overlapping predicted central antigenic determinants. Both allergens were also found to have a 95% sequence homology with previously cloned C20 and share similar antigenic determinants, as

Observations on morphological, physiological, and behavioural changes induced by protozoan infections in certain storage insect pests.

Entomopathogenic protozoans are, to a large extent, host specific, and through acute or chronic infections they negatively alter host reproductive fitness, metabolism, immune response, juvenile hormonal balance, and host development. Hence, these entomopathogens are well-suited to reduce populations of stored product insect pests. The first step towards achieving successful suppression, natural or applied, of storage insect pest populations is the detection for these entomopathogenic protozoans in their habitats. Therefore, a preliminary survey of naturally occurring protozoan infections in st

An Earliest Endosymbiont, Wolbachia massiliensis sp. nov., Strain PL13 from the Bed Bug (Cimex hemipterus), Type Strain of a New Supergroup T.

The symbiotic Wolbachia are the most sophisticated mutualistic bacterium among all insect-associated microbiota. Wolbachia-insect relationship fluctuates from the simple facultative/parasitic to an obligate nutritional-mutualistic association as it was the case of the bedbug-Wolbachia from Cimexlectularius. Understanding this association may help in the control of associated arthropods. Genomic data have proven to be reliable tools in resolving some aspects of these symbiotic associations. Although, Wolbachia appear to be fastidious or uncultivated bacteria which strongly limited their study.

Analysis of Peptide Ligand Specificity of Different Insect Adipokinetic Hormone Receptors.

Adipokinetic hormone (AKH) is a highly researched insect neuropeptide that induces the mobilization of carbohydrates and lipids from the fat body at times of high physical activity, such as flight and locomotion. As a naturally occurring ligand, AKH has undergone quite a number of amino acid changes throughout evolution, and in some insect species multiple AKHs are present. AKH acts by binding to a rhodopsin-like G protein-coupled receptor, which is related to the vertebrate gonadotropin-releasing hormone receptors. In the current study, we have cloned AKH receptors (AKHRs) from seven differen

Investigating trade-offs between ovary activation and immune protein expression in bumble bee (Bombus impatiens) workers and queens.

Evidence for a trade-off between reproduction and immunity has manifested in many animal species, including social insects. However, investigations in social insect queens present a conundrum: new gynes of many social hymenopterans, such as bumble bees and ants, must first mate, then transition from being solitary to social as they establish their nests, thus experiencing confounding shifts in environmental conditions. Worker bumble bees offer an opportunity to investigate patterns of immune protein expression associated with ovary activation while minimizing extraneous environmental factors a

Characteristics of maize flour tortilla supplemented with ground Tenebrio molitor larvae.

The larva of the Tenebrio molitor, known as the yellow meal worm, is a plague of wheat and flours. Consumption of the raw insects is not well accepted because of their appearance. The objective of the present work was to grow T. molitor larvae under standard conditions, to analyze the chemical composition of the larvae powder, and to prepare supplemented maize tortillas. Protein and fat contents were performed with standard methods. Tenebrio larvae powder had a 58.4% protein content; this protein was rich in essential amino acids such as phenylalanine, tyrosine, and tryptophan; the found value

Fusion with immunoglobulin Fc fragment enhances the immunogenicity of the African swine fever virus p30/p54 fusion protein.

African swine fever (ASF) is a significant threat to the global pig breeding industry, making the development of a vaccine extremely urgent. In this study, the key antigenic proteins p30 and p54 of African swine fever virus (ASFV) were fused with the Fc fragment of immunoglobulin (IgG) to construct recombinant fusion proteins with strong immunity. Here, we successfully expressed four recombinant fusion proteins comprising the ASFV proteins p30 and p54 (p30-54 and p54-30) alone and the porcine IgG Fc-fused p30 and p54 proteins (p30-54-Fc and p54-30-Fc) using an insect baculovirus expression sys

Corazonin mediates photoperiodically induced diapause in the bean bug Riptortus pedestris.

Insects in mid- to high latitudes predict unfavorable seasons through changing photoperiod, and undergo diapause to survive harsh conditions. The circadian clock and brain neurosecretory systems are key components in regulating photoperiodic diapause. However, the neuronal pathways linking these systems, and the neuropeptides involved in photoperiodic diapause, remain poorly understood. We investigated the role of corazonin (CRZ), a conserved neuropeptide regulating metabolic stress and reproduction, in the bean bug Riptortus pedestris, a species highly sensitive to short days for diapause ind

Development of a lateral flow test for bed bug detection.

Lateral flow strip tests are a cost-effective method for detecting specific proteins in biological samples, which can be performed in the field without specialized expertise. While most recognizable in the pregnancy tests, there are many other applications for lateral flow strip technology. The pest control industry has increasingly emphasized the importance of pest monitoring to reduce unnecessary applications, focus interventions into locations with active pest infestations, and to develop records of pest infestation. Due to their cryptic behavior, the detection of bed bugs often necessitate

The essential role of bursicon during Drosophila development.

BACKGROUND: The protective external cuticle of insects does not accommodate growth during development. To compensate for this, the insect life cycle is punctuated by a series of molts. During the molt, a new and larger cuticle is produced underneath the old cuticle. Replacement of the smaller, old cuticle culminates with ecdysis, a stereotyped sequence of shedding behaviors. Following each ecdysis, the new cuticle must expand and harden. Studies from a variety of insect species indicate that this cuticle hardening is regulated by the neuropeptide bursicon. However, genetic evidence from Drosop
