In vivo screen of Plasmodium targets for mosquito-based malaria control.

The decline in malaria deaths has recently stalled owing to several factors, including the widespread resistance of Anopheles vectors to the insecticides used in long-lasting insecticide-treated nets (LLINs)1,2. One way to mitigate insecticide resistance is to directly kill parasites during their mosquito-stage of development by incorporating antiparasitic compounds into LLINs. This strategy can prevent onward parasite transmission even when insecticides lose efficacy3,4. Here, we performed an in vivo screen of compounds against the mosquito stages of Plasmodium falciparum development. Of the …

Impact of environment on mosquito response to pyrethroid insecticides: facts, evidences and prospects.

By transmitting major human diseases such as malaria, dengue fever and filariasis, mosquito species represent a serious threat worldwide in … ical, but these require a deep understanding of adaptive mechanisms underlying resistance. Although insecticide resistance mechanisms are intensively studied in mosquitoes, such adaptation is often considered as the unique result of the selection pressure caused by insecticides used for vector control. Indeed, additional environmental parameters, such as insecticides/pe …

Chemical control of medically important arthropods in Panama: A systematic literature review of historical efforts.

Vector-borne diseases are a major source of morbidity in Panama. Herein, we describe historical usage patterns of synthetic insecticides to control arthropod disease vectors in this country. We examine the influence of interventions by vector control programs on the emergence of insecticide resistance. Chemical control has traditionally focused on two mosquito species: Anopheles albimanus, a major regional malaria vector, and Aedes aegypti, a historical vector of yellow fever, and current vector of dengue, chikungunya, and Zika. Countrywide populations of An. albimanus depict hyperirritability…

Mosquito-based malaria control through genetic, microbial and immunological innovations.

Malaria remains a major global health challenge, causing approximately 600,000 deaths annually despite decades of progress using insecticide-based and drug-based interventions. These core tools are increasingly compromised by widespread insecticide resistance in mosquitoes, drug resistance in parasites, adaptive vector behaviours and the lack of highly effective vaccines, together with logistical and sustainability constraints. Complementary strategies, including larval source management, environmental modification and novel insecticide formulations, also require sustained investment and stron…

The origin, history, and resistance architecture of an invasive urban malaria mosquito in Africa.

The invasive urban malaria vector Anopheles stephensi threatens 126 million city dwellers in Africa. Controlling An. stephensi requires greater understanding of its origin, invasion dynamics, and insecticide resistance mechanisms. Analysis of 645 whole genomes sampled across Africa, the Middle East, and Asia supports an invasion scenario in which an initial South Asian introduction established a bridgehead population in Djibouti, which seeded distinct invasion fronts in Sudan, Ethiopia/Kenya, and Yemen. These incursions show contrasting rates and routes of spread shaped by landscape topology. …

Assessment of Resistance to Organophosphates and Pyrethroids in Aedes aegypti (Diptera: Culicidae): Do Synergists Affect Mortality?

Aedes aegypti (L.) is the primary vector of Zika, dengue, yellow fever, and chikungunya viruses. Insecticides used in mosquito control can help prevent the spread of vector-borne diseases. However, it is essential to determine insecticide resistance (IR) status before control measures are undertaken. Only the most effective insecticides should be used to avoid ineffective control and/or promotion of IR. Pyrethroids and organophosphates are the most commonly used insecticides for mosquito control. Here, the efficacy of two active ingredients (AIs; permethrin [pyrethroid], chlorpyrifos [organoph…

Structural Insights into Pyrethroid Recognition by Anopheles Chemosensory Proteins.

Mosquito-borne diseases, particularly malaria, remain a major global health challenge. Among different mosquitoes, Anopheles are solely responsible for malaria. Insecticide-based interventions such as insecticide-treated nets (ITNs) and indoor residual spraying (IRS) are central to mosquito control. However, the widespread emergence of insecticide resistance, especially to pyrethroids, the most commonly used insecticides for mosquito worldwide, threatens their effectiveness. While genetic and metabolic mechanisms of resistance are well characterized, behavioral and sequestration-based mechanis…

Absence of knockdown mutations in pyrethroid and DDT resistant populations of the main malaria vectors in Colombia.

BACKGROUND: Knockdown resistance (kdr) is a well-characterized target-site insecticide resistance mechanism that is associated with DDT and pyrethroid resistance. Even though insecticide resistance to pyrethroids and DDT have been reported in Anopheles albimanus, Anopheles benarrochi sensu lato (s.l.), Anopheles darlingi, Anopheles nuneztovari s.l., and Anopheles pseudopunctipennis s.l. malaria vectors in Latin America, there is a knowledge gap on the role that kdr resistance mechanisms play in this resistance. The aim of this study was to establish the role that kdr mechanisms play in pyrethr…

Assessing insecticide susceptibility status of Anopheles mosquitoes in Gondar zuria district, Northwest Ethiopia.

Insecticide-based vector control, which comprises the use of insecticide-treated bed nets (ITNs) and indoor residual spraying (IRS), is the key method of malaria control in Ethiopia. However, its effectiveness is threatened as malaria vectors become resistant to insecticides. Thus, the aim of this study was to monitor the insecticide susceptibility status of malaria vectors. WHO susceptibility tests were used to detect knock-down and mortality rate in the wild malaria vectors collected in Gondar zuria woreda, Northwest Ethiopia. The WHO diagnostic doses of 0.75% permethrin, 0.05% deltamethrin,

Contrasting patterns of tolerance between chemical and biological insecticides in mosquitoes exposed to UV-A.

Mosquitoes are vectors of major human diseases, such as malaria, dengue or yellow fever. Because no efficient treatments or vaccines are available for most of these diseases, control measures rely mainly on reducing mosquito populations by the use of insecticides. Numerous biotic and abiotic factors are known to modulate the efficacy of insecticides used in mosquito control. Mosquito breeding sites vary from opened to high vegetation covered areas leading to a large ultraviolet gradient exposure. This ecological feature may affect the general physiology of the insect, including the resistance

Knockdown resistance (kdr) mutations in the Japanese encephalitis virus vector Culex tritaeniorhynchus from the Republic of Korea.

BACKGROUND: Culex tritaeniorhynchus is the primary vector of Japanese encephalitis virus (JEV), a disease of global public health significance, and is widely distributed across Africa, Asia, Australia, and Europe. Despite its epidemiological importance as a JEV vector, studies on insecticide resistance in Cx. tritaeniorhynchus populations from the Republic of Korea (ROK) remain extremely limited. This study aimed to investigate the spatial and temporal occurrence of knockdown resistance (kdr) mutations in Cx. tritaeniorhynchus populations from the ROK. Additionally, the association between phe…

Field evolved resistance to pyrethroids, neonicotinoids, organophosphates and macrolides in Rhopalosiphum padi (Linnaeus) and Sitobion avenae (Fabricius) from China.

Rhopalosiphum padi (Linnaeus) and Sitobion avenae (Fabricius) are the predominant pests coexisting on wheat plants. In this study, the susceptibilities of 29 R. padi and 30 S. avenae populations from 15 provinces in China to pyrethroids (beta-cypermethrin and bifenthrin), neonicotinoids (imidacloprid and thiamethoxam), organophosphates (omethoate and chlorpyrifos) and macrolides (avermectin) were determined during 2018-2019. The median lethal insecticide concentrations (LC50) indicated that R. padi was more sensitive than S. avenae to most of the insecticides. Monitor results showed that most

Spatial and temporal country-wide survey of temephos resistance in Brazilian populations of Aedes aegypti.

The organophosphate temephos has been the main insecticide used against larvae of the dengue and yellow fever mosquito (Aedes aegypti) in Brazil since the mid-1980s. Reports of resistance date back to 1995; however, no systematic reports of widespread temephos resistance have occurred to date. As resistance investigation is paramount for strategic decision-making by health officials, our objective here was to investigate the spatial and temporal spread of temephos resistance in Ae. aegypti in Brazil for the last 12 years using discriminating temephos concentrations and the bioassay protocols o

Status of insecticide susceptibility in Anopheles arabiensis from Mwea rice irrigation scheme, Central Kenya.

BACKGROUND: Control of the Anopheline mosquito vectors of malaria by use of insecticides has been shown to impact on both morbidity and mortality due to this disease. Evidence of insecticide resistance in different settings necessitates surveillance studies to allow prompt detection of resistance should it arise and thus enable its management. Possible resistance by Anopheles arabiensis mosquitoes from Mwea rice irrigation scheme in Central Kenya to insecticides in the four classes of insecticides approved by WHO for indoor residual spraying was investigated. METHODS: Susceptibility to DDT (a…

Selection and characterization of spinetoram resistance in field collected Drosophila melanogaster.

Insecticides are commonly employed in vineyards to control vinegar flies and limit sour rot disease. Widespread resistance to available insecticides is having a negative impact on managing Drosophila melanogaster populations, rendering control of sour rot more difficult. An insecticide registered for use in vineyards to which resistance is not yet widespread (at least in New York and Missouri) is spinetoram. Spinetoram targets the nicotinic acetylcholine receptor α6, and mutations in α6 have been associated with resistance in some insects. Our goals were to select for a spinetoram resistant st

Efficacy of a novel mode of action of an indoor residual spraying product, SumiShield® 50WG against susceptible and resistant populations of Anopheles gambiae (s.l.) in Benin, West Africa.

BACKGROUND: Scale-up of the distribution of long-lasting insecticide-treated bed nets and indoor residual spraying with insecticides over the last decade have contributed to the considerable decrease of malaria morbidity and mortality in sub-Saharan Africa. Due to the increasing pyrethroid resistance intensity and the spread of carbamate resistance in Anopheles gambiae (s.s.) mosquitoes and the limited number of insecticides recommended by the WHO for vector control, alternative insecticide formulations for IRS with long-lasting residual activity are required to sustain the gains obtained in m
