Advantages of pyruvate-based fluids in preclinical shock resuscitation-A narrative review.

This review focuses on the innate beneficial effects of sodium pyruvate-based fluids, including pyruvate in intravenous solutions, oral rehydration solutions, and peritoneal dialysis solutions, on shock resuscitation with various animal models relative to current commercial fluids over the last two decades. Due to its superior pharmacological properties, pyruvate effectively sustains cytosolic glycolytic pathways and mitochondrial oxidative phosphorylation by restoration of redox potentials and reactivation of pyruvate dehydrogenase in hypoxia, even anoxia, and diabetes, reversing the Warburg

[Prostato-urethritis due to chlamydia].

One of the etiological agents of urethritis and prostatitis is Chlamydia trachomatis which reveals obligatory intracellular parasitism. Patient specimens (smears of the urethra, urine, first portion, expressed prostatic fluid, ejaculate) are centrifuged onto special cell cultures, so-called McCoy cells, and induce characteristic cytoplasmatic inclusions. Giemsa staining or DNA-binding fluorochromes (DAPI or Hoechst 33258) are suitable procedures to identify chlamydia inclusions. Serologic tests to establish infections due to Chlamydia trachomatis are only of limited diagnostic value.

A predictive model for body water and fluid balance using 3D smartphone anthropometry.

BACKGROUND: Body fluid volumes, including total body water (TBW), extracellular fluid (ECF), and intracellular fluid (ICF), are crucial indicators of body composition, and the distribution of these fluids is essential for assessing hydration status and fluid accumulation. Although fluid volumes are commonly measured with bioelectrical impedance devices, several challenges hinder the application of this technique. However, 3D smartphone scanning applications that automate body volumes and other anthropometric estimates may provide a viable alternative to body fluid assessments. METHODS: A tota…

Molecular mechanisms of brain water transport.

Our brains consist of 80% water, which is continuously shifted between different compartments and cell types during physiological and pathophysiological processes. Disturbances in brain water homeostasis occur with pathologies such as brain oedema and hydrocephalus, in which fluid accumulation leads to elevated intracranial pressure. Targeted pharmacological treatments do not exist for these conditions owing to our incomplete understanding of the molecular mechanisms governing brain water transport. Historically, the transmembrane movement of brain water was assumed to occur as passive movemen

N-Acetylcysteine Protects HPMCs from High-Glucose-Induced Oxidative DNA Damage.

Peritoneal dialysis (PD) is an effective renal replacement therapy; however, its long-term use is limited by the detrimental effects of glucose-based PD fluids on the peritoneal membrane, contributing to fibrosis and ultrafiltration failure. Previous studies have demonstrated that high-glucose exposure promotes oxidative DNA damage through upregulation of thioredoxin-interacting protein (TXNIP) expression, resulting in reduced thioredoxin (Trx) activity. This study investigated strategies to reduce oxidative stress in human peritoneal mesothelial cells exposed to high glucose concentrations. T

BioFlow: a non-invasive, image-based method to measure speed, pressure and forces inside living cells.

Cell motility is governed by a complex molecular machinery that converts physico-chemical cues into whole-cell movement. Understanding the underlying biophysical mechanisms requires the ability to measure physical quantities inside the cell in a simple, reproducible and preferably non-invasive manner. To this end, we developed BioFlow, a computational mechano-imaging method and associated software able to extract intracellular measurements including pressure, forces and velocity everywhere inside freely moving cells in two and three dimensions with high spatial resolution in a non-invasive man

Intracellular Gelation-Mediated Living Bacteria for Advanced Biotherapeutics in Mouse Models.

Despite its significant potential in various disease treatments and diagnostics, microbiotherapy is consistently plagued by multiple limitations ranging from manufacturing challenges to in vivo functionality. Inspired by the strategy involving nonproliferating yet metabolically active microorganisms, we report an intracellular gelation approach that can generate a synthetic polymer network within bacterial cells to solve these challenges. Specifically, poly(ethylene glycol dimethacrylate) (PEGDA, 700 Da) monomers are introduced into the bacterial cytosol through a single cycle of freeze-thawin

Evidence of purinergic neurotransmission in isolated, intact horse sweat glands.

BACKGROUND: Fluid secretion by sweat glands in response to heat and exercise is underpinned by increases in intracellular calcium. In horses, this is primarily via β2-adrenoceptors, but studies in equine sweat gland cell lines have indicated a possible role for purinergic agonists. Knowledge of equine sweating stimulus-secretion mechanisms in intact glands from healthy animals would allow future comparison to determine whether these mechanisms are affected in equine anhidrosis. HYPOTHESIS/OBJECTIVES: To determine whether purinergic agonists can induce changes in intracellular calcium in intac

Levetiracetam inhibits the Na+ HCO3 - transporter Ncbe/NBCn2 (Slc4a10) in the choroid plexus of mice.

AIM: The Na+-dependent Cl-/HCO3 - exchanger Ncbe/NBCn2 (Slc4a10) constitutes a major basolateral HCO3 - uptake pathway in the choroid plexus and contributes to cerebrospinal fluid secretion and pH regulation. This study examined whether levetiracetam inhibits Ncbe/NBCn2-mediated transport and alters cerebrospinal fluid acid-base balance, transporter abundance, and cerebrospinal fluid secretion. METHODS: The Ncbe and NBCn1 activity was assessed in heterologous expression systems as Na+-dependent pH recovery after intracellular acidification. Na+-dependent pH recovery was also measured in isola

Single-Cell Metallomics of ovarian granulosa cells enables assessment of Low-Level heavy metal exposure associated with impaired human oocyte development.

The oocyte microenvironment, comprising ovarian granulosa cells (GCs) and follicular fluid (FF), sustains oocyte development with nutrients and regulatory signals but may also transfer heavy metals to the oocyte. While high concentrations of heavy metals in FF and blood have been associated with impaired oocyte development, the effect of low-level exposure on the oocyte remains unclear, partly due to the analytical challenges in measuring low-level heavy metals in biological fluids. Here, we investigated the associations between low-level heavy metal exposure and oocyte development. Using a hi

Phase angle as an index of physiological status: validating bioelectrical assessments of hydration and cell mass in health and disease.

Bioelectrical impedance (BI) is a practical method to assess body composition in health and disease. This method relies on the passive conduction of an applied, safe, low-level alternating current through water and electrolytes in the body. Using a phase-sensitive device, BI yields measurements of impedance (Z) and its components, resistance (R) and reactance (Xc), that are related geometrically as phase angle (PhA). In vitro studies provide empirical evidence relating BI measurements to physiological variables. Cooking raw food samples results in greater decreases in PhA, predominantly Xc, wi

Local synthesis of both macrophage and T cell cytokines by synovial fluid cells from children with juvenile rheumatoid arthritis.

The production of tumour necrosis factor-alpha (TNF-alpha), TNF-beta and IL-6 in synovial fluid was studied in 50 samples of synovial fluid from 44 children with juvenile rheumatoid arthritis (JRA) by identifying cytokine production at a single-cell level. Post Ficoll-separated synovial fluid mononuclear cells were permeabilized and then intracellular TNF-alpha, TNF-beta and IL-6 protein production was examined using indirect immunofluorescence and murine anti-cytokine MoAbs. All three cytokines were measured in 37 of the 50 samples. In 25 of the 37 samples there was complete concordance; all

Fluid Shifts Induced by Physical Therapy in Lower Limb Lymphedema Patients.

Complete decongestive therapy (CDT), a physical therapy including manual lymphatic drainage (MLD) and compression bandaging, is aimed at mobilizing fluid and reducing limb volume in lymphedema patients. Details of fluid shifts occurring in response to CDT are currently not well studied. Therefore, we investigated fluid shifts before, during and after CDT. Thirteen patients (3 males and 10 females, aged 57 ± 8.0 years, 167.2 ± 8.3 cm height, 91.0 ± 23.4 kg weight) diagnosed with stage II leg lymphedema participated. Leg volume, limb and whole-body fluid composition (total body water (limbTBW/%T

Distinct Roles of SLC26A3 and CFTR in Surface pH Regulation and Bicarbonate Secretion in Human Intestinal Epithelium.

BACKGROUND AND AIMS: Colonic bicarbonate secretion is mediated by the chloride/bicarbonate exchanger SLC26A3 and the cystic fibrosis transmembrane conductance regulator (CFTR). Dysfunction of either causes luminal acidosis, altered mucus properties, and inflammation. While physical and functional interactions have been demonstrated in heterologous systems, their relationship in native epithelium is not fully established. We investigated the distinct roles of SLC26A3 and CFTR using human intestinal organoids with inducible SLC26A3 overexpression. METHODS: Human colonic and rectal organoids fro

Cholera: pathophysiology and emerging therapeutic targets.

Cholera is a diarrheal disease that remains an important global health problem with several hundreds of thousands of reported cases each year. This disease is caused by intestinal infection with Vibrio cholerae, which is a highly motile gram-negative bacterium with a single-sheathed flagellum. In the course of cholera pathogenesis, V. cholerae expresses a transcriptional activator ToxT, which subsequently transactivates expressions of two crucial virulence factors: toxin-coregulated pilus and cholera toxin (CT). These factors are responsible for intestinal colonization of V. cholerae and induc

[Models for the study of the toxic action of Legionella pneumophila].

L. pneumophila virulent culture and the filtrate of this culture disintegrated with ultrasound were shown to be toxic for guinea-pig peritoneal macrophages in vitro and for AKR mice. The virulent culture of this infective agent and the filtrate of the supernatant fluid obtained from the washings of its virulent culture had no toxic effect on a macrophage monolayer culture and on mice. The use of these models for characterizing Legionella intracellular toxic activity, as well as for characterizing Legionella strains isolated from patients and the environment, is proposed.
