Transcriptomic analysis of eggs, rediae and cercariae reveal stage-specific adaptations in the rumen fluke Calicophoron daubneyi.

Rumen flukes, particularly the trematode Calicophoron daubneyi, are emerging parasites of livestock in Europe, yet transcriptomic insights into their environmental and intermediate host stages remain limited. Here, we present a comprehensive transcriptomic analysis of eggs at three distinct developmental stages (freshly excreted, early developmental and eye-spot stages), as well as rediae and cercariae, of C. daubneyi. High-quality RNA-sequencing (RNA-seq) datasets revealed both shared and stage-specific transcriptional profiles with each developmental stage exhibiting its own distinct express

Beyond the Sand Fly and the Macrophage: A Multidimensional Redefinition of the Leishmania Life Cycle to Overcome Therapeutic Persistence.

Leishmaniasis remains a major global health challenge, hampered by an antiquated therapeutic arsenal compromised by toxicity, resistance, and an inability to achieve a sterile cure. The high attrition rate in drug and vaccine development stems from a fundamental disconnect between traditional experimental models and the true biological complexity of the Leishmania life cycle. This review synthesizes recent breakthroughs that have reshaped our understanding of the parasite, arguing that these nonclassical traits are the primary obstacles to effective treatment. We deconstruct the updated life c

A New Triatomine on the Chagas Disease Scene: What We Know (and What We Do Not) About Triatoma huehuetenanguensis (Hemiptera; Reduviidae).

Background: Triatomines are a group of nearly 158 species of hematophagous insects known as kissing bugs. Several are recognized as important vectors of Trypanosoma cruzi, the causative agent of Chagas disease. Triatoma huehuetenanguensis, a triatomine described in Guatemala in 2019, is of particular interest, as it belongs to the dimidiata subcomplex, an epidemiologically important group in southern Mexico and Central America. Objective: This review integrates advances regarding its discovery, distinctive morphological characteristics, life cycle, feeding and defecation patterns, geographic di

Predicting short-life-cycle species suitability via dynamic species distribution models: Applications in dengue and malaria epidemiology in Brazil.

Species Distribution Models (SDMs) are used to estimate environmental suitability and the potential distribution of organisms based on occurrence records and climatic variables. However, traditional approaches rely on long-term climatic averages, producing static predictions with no ability to capture seasonal variations that are particularly relevant for short life-cycle species, such as disease vectors. In this study, we developed Dynamic Species Distribution Models (DSDMs) for Ae. aegypti and An. darlingi by integrating monthly occurrence records and corresponding climatic variables from 20

Toxicity of Four Common Environmental Chemicals Across Caenorhabditis elegans Life Stages Supporting the One Health Concept.

Pesticides and pharmaceuticals are among the most common chemical groups in waterbodies and soils, and their universal distribution raises concerns about potential adverse effects on nontarget organisms and humans. Reproductive output disruption is of particular concern, as it transposes effects from the individual to the next generations at the population level, requiring precise identification of the affected processes. Caenorhabditis elegans , with its well-known lifecycle, is an excellent model organism for investigating such effects within the One Health framework. The present study evalu

Hedgehogs and Angiostrongylus cantonensis: Uncovering the Role of Atelerix albiventris in the Parasite Life Cycle.

The ongoing invasion of the zoonotic parasite Angiostrongylus cantonensis threatens humans, other mammals, and birds. In Mallorca, neurological disorders associated with this parasite are increasingly reported in free-living Algerian hedgehogs, Atelerix algirus, raising public health concerns. Experimental infections in the related species Atelerix albiventris were conducted to investigate the role of hedgehogs in the life cycle of the parasite. Captive-born hedgehogs were orally inoculated with 200 and 2000 third-stage larvae and monitored for 50 days by behavioral observations, blood tests (

Digenean life cycle truncation has enabled the opportunistic exploitation of herbivorous fishes.

New, well-known and predicted life cycles for trematodes of the Haploporoidea (Haploporidae and Emprostiotrematidae) and three families of the Lepocreadioidea (Enenteridae, Gorgocephalidae, Gyliauchenidae) involve encystment of the metacercaria in the open (usually on vegetation) followed by ingestion by a range of herbivorous or detritivorous fishes. These life cycles appear among relatively highly derived plagiorchiidan trematodes in which three-host life cycles incorporating an animal second intermediate host are dominant. We hypothesise that the two-host life cycles in the Haploporoidea an

Validating a variable-instar, climate-based phenology model for the Asian longhorned beetle (Coleoptera: Cerambycidae) using field data from South Carolina.

The Asian longhorned beetle, Anoplophora glabripennis (ALB, Coleoptera: Cerambycidae), is a federally regulated invasive species capable of infesting several different genera of hardwood trees. Accurate knowledge of ALB's phenology is critical for the effective implementation of management and eradication plans. We updated the ALBLT prediction model and used empirical data collected in South Carolina, USA to validate ALBLT v. 2.0. The new model largely agreed with ALB life stages found in field collections, except for late instars and pupae. We also ran the model at 8 other potentially high-ri

Decoding the epigenetic blueprint behind Toxoplasma (pre)sexual commitment and chronic persistence.

Toxoplasma gondii, a widespread apicomplexan parasite, navigates a complex life cycle in which it switches back and forth between tachyzoites and bradyzoites to balance acute infection and persistence while also undergoing an often-overlooked one-way commitment to the sexual stage. Emerging evidence suggests that Toxoplasma's developmental trajectories are orchestrated by an interconnected network of transcriptional switches and chromatin remodeling mechanisms, acting in concert to ensure robust stage transitions. This review outlines an updated epigenetic framework for stage transitions and h

Regulation and Roles of Metacyclogenesis and Epimastigogenesis in the Life Cycle of Trypanosoma cruzi.

Trypanosoma cruzi, the etiological agent of Chagas disease, exhibits remarkable developmental plasticity that enables its survival across distinct environments within the insect vector and mammalian host. This review focuses on two critical differentiation processes-metacyclogenesis and epimastigogenesis-emphasising their environmental triggers, metabolic regulation, and roles in parasite transmission and life cycle progression. Metacyclogenesis, occurring in the hindgut of triatomine vectors, transforms replicative epimastigotes into infective metacyclic trypomastigotes and is tightly control

Effects of fasting on the interplay between temperature and Trypanosoma cruzi infection on the life cycle of the Chagas disease vector Rhodnius prolixus.

Rhodnius prolixus, a triatomine insect, is one of the most important vectors of Chagas disease in South America. Its interaction with the parasite Trypanosoma cruzi, the causative agent of this disease, is known to be deeply affected by ambient temperature and the nutritional status of the insect vector. In this study, we investigated how starvation affects the life cycle of R. prolixus and the population dynamics of the parasite inside the intestine of the vector at four temperatures ranging from 24°C to 30°C. The weights and molting times of chronically infected and uninfected insects were m

Cryptosporidium life cycle small molecule probing implicates translational repression and an Apetala 2 transcription factor in macrogamont differentiation.

The apicomplexan parasite Cryptosporidium is a leading cause of childhood diarrhea in developing countries. Current treatment options are inadequate and multiple preclinical compounds are being actively pursued as potential drugs for cryptosporidiosis. Unlike most apicomplexans, Cryptosporidium spp. sequentially replicate asexually and then sexually within a single host to complete their lifecycles. Anti-cryptosporidial compounds are generally identified or tested through in vitro phenotypic assays that only assess the asexual stages. Therefore, compounds that specifically target the sexual st

Comparative biology of parasitic nematodes in the genus Angiostrongylus and related genera.

The rise to prominence of some Angiostrongylus species through associated emerging disease in humans and dogs has stimulated calls for a renewed focus on the biology of this genus and three related genera. Although significant research efforts have been made in recent years these have tended to focus on individual species and specific aspects such as diagnosis and treatment of disease or new records of occurrence and hosts. This comprehensive review takes a comparative approach, seeking commonalities and differences among species and asking such questions as: Which species belong to this and t

Rat lungworm survives winter: experimental overwintering of Angiostrongylus cantonensis larvae in European slugs.

The rat lungworm Angiostrongylus cantonensis is a metastrongyloid nematode that causes neurological disorders in its accidental hosts, including humans. This invasive pathogen is native to Southeast Asia and adjacent regions and is gradually expanding its distribution to tropical and subtropical areas with new foci discovered near temperate regions. The parasite has a complex life cycle with a range of gastropods serving as intermediate hosts. A broad spectrum of poikilotherm vertebrates and invertebrates can serve as paratenic hosts. Since it has already been demonstrated that other, non-zoon

Parental whole life-cycle exposure to tris (2-chloroethyl) phosphate (TCEP) disrupts embryonic development and thyroid system in zebrafish offspring.

Tris (2-chloroethyl) phosphate (TCEP), an emerging environmental pollutant, has been frequently detected in natural waters. The objective of this study was to investigate possible parental transfer of TCEP and transgenerational effects on the early development and thyroid hormone homeostasis in F1 larvae following parental whole life-cycle exposure to TCEP. To this end, zebrafish (Danio rerio) embryos were exposed to environmentally relevant concentrations (0.8, 4, 20 and 100 μg/L) of TCEP for 120 days until sexual maturation. Parental exposure to TCEP resulted in significant levels of TCEP, d

Influence of Staphylococcus aureus Strain Background on Sa3int Phage Life Cycle Switches.

Staphylococcus aureus asymptomatically colonizes the nasal cavity of mammals, but it is also a leading cause of life-threatening infections. Most human nasal isolates carry Sa3 phages, which integrate into the bacterial hlb gene encoding a sphingomyelinase. The virulence factor-encoding genes carried by the Sa3-phages are highly human-specific, and most animal strains are Sa3 negative. Thus, both insertion and excision of the prophage could potentially confer a fitness advantage to S. aureus. Here, we analyzed the phage life cycle of two Sa3 phages, Φ13 and ΦN315, in different phage-cured S. a

Diversity and phylogeny of Paradiplozoon species (Monogenea: Diplozoidae) parasitising endemic cyprinoids in the peri-Mediterranean area, with a description of three new Paradiplozoon species.

The Diplozoidae are monogenean parasites of mainly cyprinoid fishes with a unique life cycle, whereby two larvae undergo anastomosis and fuse into a single cross-shaped specimen. Paradiplozoon is the most species rich and widespread genus of the family, with a distribution range covering Eurasia and Africa; however, some areas remain underexplored and their diplozoid fauna is uncertain. In the present study, the Paradiplozoon diversity was investigated in the peri-Mediterranean region, which has the highest levels of cyprinoid diversity and endemism in Europe. A total of 36 endemic cyprinoid s

Tonic Immobility Is Influenced by Starvation, Life Stage, and Body Mass in Ixodid Ticks.

The ability to escape predation modulates predator-prey interactions and represents a crucial aspect of organismal life history, influencing feeding, mating success, and survival. Thanatosis, also known as death feigning or tonic immobility (TI), is taxonomically widespread, but understudied in blood-feeding vectors. Hematophagous arthropods, such as ticks, are unique among animals as their predators (birds, mice, lizards, frogs, and other invertebrates) may also be their source of food. Therefore, the trade-off between predator avoidance and host-seeking may shift as the time since the last b

Endemicity of Paragonimus and paragonimiasis in Sub-Saharan Africa: A systematic review and mapping reveals stability of transmission in endemic foci for a multi-host parasite system.

Paragonimiasis is caused by zoonotic trematodes of Paragonimus spp., found in Asia, the Americas and Africa, particularly in tropical regions. These parasites have a complex, multi-host life cycle, with mammalian definitive hosts and larval stages cycling through two intermediate hosts (snails and freshwater decapod crustaceans). In Africa, paragonimiasis is particularly neglected, and remains the only human parasitic disease without a fully characterised life cycle. However paragonimiasis has potentially significant impacts on public health in Africa, and prevalence has likely been underestim
