Dengue Virus Uses a Non-Canonical Function of the Host GBF1-Arf-COPI System for Capsid Protein Accumulation on Lipid Droplets.

Dengue viruses cause the most important human viral disease transmitted by mosquitoes. In recent years, a great deal has been learned about molecular details of dengue virus genome replication; however, little is known about genome encapsidation and the functions of the viral capsid protein. During infection, dengue virus capsid progressively accumulates around lipid droplets (LDs) by an unknown mechanism. Here, we examined the process by which the viral capsid is transported from the endoplasmic reticulum (ER) membrane, where the protein is synthesized, to LDs. Using different methods of inte…

Exposure to acute waterborne cadmium caused severe damage on lipid metabolism of freshwater fish, revealed by nuclear lipid droplet deposition in hepatocytes of rare minnow.

Cadmium (Cd) is a widely distributed aquatic toxic heavy metal with the potential to disrupt fish metabolism; however, more research is needed to clarify the underlying mechanisms. In the present study, rare minnows (Gobiocypris rarus) were used to detect the effects of cadmium on freshwater fish lipid metabolism and its underlying mechanism by histopathological observation, measurement of serum and liver biochemical indexes, and analysis of gene expression in terms of lipid oxidation, synthesis and transport. Here, severe damage, such as cytoplasmic lipid droplet (LD) accumulation, ectopic de

Engineering of a NIR fluorescent probe for high-fidelity tracking of lipid droplets in living cells and nonalcoholic fatty liver tissues.

LDs (Lipid droplets) are key organelles for lipid metabolism and storage, which are closely related to ferroptosis and fatty liver. Due to its small size and highly dynamic nature, developing high-fidelity fluorescent probes for imaging of LDs is crucial for observing the dynamic physiological processes of LDs and investigating LDs-associated diseases. Herein, we synthesized three dicyanoisophorone-based fluorescent probes (DCIMe, DCIJ, and DCIQ) with different electron-donating groups and studied their imaging performance for LDs. The results show that DCIQ is highly polarity sensitive and ca…

Visualization of Lipid Droplets in the Alveolar Macrophage Cell Line MH-S with Live-cell Imaging by 3D Holotomographic Microscopy (Nanolive).

Lipid droplets (LD), triglycerides and sterol esters among them, are well known for their capacity as lipid storage organelles. Recently, they have emerged as critical cytoplasmic structures involved in numerous biological functions. LD storage is generated de novo by the cell and provides an energy reserve, lipid precursors, and cell protection. Moreover, LD accumulation can be observed in some pathologies as obesity, atherosclerosis, or lung diseases. Fluorescence imaging techniques are the most widely used techniques to visualize cellular compartments in live cells, including LD. Neverthele…

Creb-Pgc1α pathway modulates the interaction between lipid droplets and mitochondria and influences high fat diet-induced changes of lipid metabolism in the liver and isolated hepatocytes of yellow catfish.

Although the crucial role of lipid droplets (LDs), mitochondria (MT) and their interactions in regulating lipid metabolism are well accepted, the mechanism of LDs-MT interactions in high fat diet (HFD)-induced changes of lipid metabolism remains unknown. Thus, this study was conducted to determine the mechanism of LDs-MT interactions in HFD-induced changes of lipid accumulation. We found that HFD not only up-regulated the expression of key proteins linked with TAG biosynthesis, but also increased the expression of proteins involved in lipolysis and fatty acid (FA) oxidation in LDs, including R…

Developmental and Seasonal Changes in Lipid Droplets and Fatty Acid Composition in the Ovary and Liver of Female Spotted Scat (Scatophagus argus).

The spotted scat (Scatophagus argus) is a fish species widely distributed in Indo-Pacific waters. In China, females are preferred for aquaculture due to their faster growth rates. Reproduction in teleost fish requires substantial energy, primarily in the form of lipids and fatty acids, which are closely linked to reproductive success and ovarian development. In this study, female spotted scat broodstock were sampled bi-monthly from May 2019 to March 2020 to examine annual ovarian development, lipid droplet accumulation, and fatty acid composition in the liver and ovaries. Body weight, total le

A wash-free fluorescent probe with a large Stokes shift for the identification of NAFL through tracing the change of lipid droplets.

As one of the important organelles in cells, lipid droplets (LDs) are involved in various physiological processes, especially affecting the occurrence and progression of non-alcoholic fatty liver (NAFL). Therefore, it is of great significance to develop LD-specific probes with excellent biocompatibility, deep penetration and bright fluorescence. Herein, a fluorescent probe LD-HWZ was designed and synthesized based on triphenylamine and the dicyanoisophorone group. It is found that probe LD-HWZ has a large Stokes shift (Δλ = 160 nm in DMSO) and exhibits bright fluorescence in a lipid environmen…

Relationship between mitochondria and the development of specific lipid droplets in capillary endothelial cells of the respiratory tract in patients with sarcoidosis.

The aim of this study was to morphologically evaluate damage in the capillary endothelial cells of the respiratory tract in patients with sarcoidosis. We examined tissues of the bronchus and lung obtained from 16 patients with sarcoidosis consisting of 2 stage 0, 10 stage I and 4 stage II patients, and 11 controls. The morphology of capillary endothelial cells was studied using electron microscopy. In the samples from patients with sarcoidosis, lipid droplets exhibiting dark monophasic density (unsaturated fatty acids) were surrounded by abundant lysosomes in the capillary endothelial cells, a…

Absolute specificity for retrograde fast axonal transport displayed by lipid droplets originating in the axon of an identified Aplysia neuron in vitro.

Lipid droplets were found to form all along the axon of the giant cerebral neuron (GCN) of the sea hare Aplysia californica when the cell was placed in culture. The emission of yellow fluorescence by the droplets after exposure of the neuron to Nile red and their uniformly dark appearance in electron micrographs of axons fixed with glutaraldehyde and osmium tetroxide identified them as lipid droplets. In contrast to lipid droplets in fat cells and certain other cell types, these droplets were bounded by a membrane, indicating that the lipid droplet is a type of organelle that is membranated in…

Comparative lipidomics of iPSC-derived microglia protocols reveal lipid droplet and immune differences mediated by media composition.

Altered microglial lipid metabolism is heavily implicated in Alzheimer's disease (AD) and aging. Recently, protocols were developed to generate human induced pluripotent stem cell-derived microglia-like cells (iMGL) to study microglial function in vitro, including embryoid body-based methods and induced transcription factor (iTF)-dependent approaches. Here, we performed comparative lipidomics on iMGL from these methods and report major differences in multiple lipid classes, including triglycerides (TGs), a storage form of fatty acids implicated in microglial reactivity. TGs are strongly increa

A lipid droplet-targeted Pt(II) complex enables photoinduced ferroptosis and apoptosis against cisplatin-resistant cells.

The development of metal complex-based photosensitizers that can precisely regulate cell fate via lipid droplet (LD)-mediated pathways remains challenging. Herein, an LD-specific targeting Pt(II) complex was designed to address this limitation by directly inducing sufficient lipid peroxidation (LPO) accumulation, offering a novel platform for synergistic ferroptosis-apoptosis cancer therapy. This work provides a novel anticancer platform using photoactivated platinum complexes to concurrently trigger dual cell death pathways.

Label-free 4D holotomography with depth-adaptive segmentation for quantitative analysis of lipid droplet dynamics in hepatic organoids.

SIGNIFICANCE: Quantifying lipid droplet (LD) remodeling in 3D hepatic organoids is often limited to endpoint staining or phototoxic live fluorescence imaging, thereby obscuring droplet-level kinetics. AIM: We aimed to develop a label-free method to track LD dynamics in living hepatic organoids under different fatty-acid loads. APPROACH: Time-lapse 3D refractive-index tomograms were acquired using holotomography and analyzed with a depth-adaptive, multithreshold segmentation pipeline to quantify LD number, volume, sphericity, and refractive-index-derived concentration and dry mass at single-d

A Controlled-Area Droplet Evaporimeter Reveals Surface Pressure-Dependent Evaporation Resistance of the Tear Film Lipid Layer.

PURPOSE: The purpose of this study was to quantitatively determine the intrinsic evaporation resistance of the tear film lipid layer (TFLL) and resolve discrepancies between in vivo and in vitro findings. METHODS: A controlled-area droplet evaporimeter (CADE) was developed based on constrained drop surfactometry. The system integrates three independent feedback control loops to maintain constant droplet surface area, surface temperature (34°C), and environmental relative humidity (30%-70%). A model TFLL composed of representative polar and nonpolar lipids was studied. Evaporation flux was mea

Aryl hydrocarbon receptor-dependent lipid droplet accumulation supports redox homeostasis in hypoxia-induced PCa cells.

Tumor cells in hypoxic environments generate large amounts of reactive oxygen species (ROS), and lipid droplets (LDs) play an essential role in maintaining redox homeostasis under these harsh conditions. The aryl hydrocarbon receptor (AhR) is a member of the basic helix-loop-helix (bHLH) transcription factor superfamily that is closely associated with malignant tumor phenotypes. AhR has been shown to drive prostate cancer (PCa) cell growth, but the specific mechanistic basis for this effect remains unclear. In this study, we used a hypoxia-induced PC-3M cell model to explore the molecular path…

Chronic TBPH Exposure Drives the Transition from Steatosis to Hepatic Fibrosis via Lipid Droplet Dysregulation in Zebrafish.

Bis(2-ethylhexyl)-2,3,4,5-tetrabromophthalate (TBPH) is a widely used novel flame retardant and an emerging ubiquitous environmental contaminant. While acute exposure disrupts lipid signaling, the long-term consequences of TBPH exposure on the progression of metabolic dysfunction-associated steatotic liver disease (MASLD) remain poorly understood. This study aimed to elucidate the chronic hepatotoxic effects of TBPH and the underlying molecular mechanisms. Adult zebrafish were exposed to environmentally relevant concentrations of TBPH for 6 weeks. Hepatic damage was assessed using histological

Pro-Inflammatory Microglia Exacerbate High-Altitude-Induced Cognitive Impairment by Driving Lipid Droplet Accumulation in Astrocytes.

High-altitude cognitive impairment (HACI) results from acute or chronic exposure to hypoxic conditions. Brain lipid homeostasis is crucial for cognitive function, and lipid droplet (LD) accumulation in glia cells is linked to cognitive decline in aging and stroke. However, whether high-altitude exposure affects brain lipid homeostasis is unclear. Microglia, key regulators of brain homeostasis and inflammation, play a significant role in pathological cognitive impairment and are implicated in LD formation. This study investigates whether lipid dysregulation contributes to HACI and explores micr

Donor Modulation of Strong Near-Infrared Lipid-Droplet Probes for in Vivo Diagnosis of Nonalcoholic Fatty Liver Disease.

Lipid droplet accumulation is a pathological hallmark of nonalcoholic fatty liver disease (NAFLD); however, the utility of current lipid droplet-targeted fluorescent probes for in vivo diagnosis is hindered by shallow tissue penetration and strong background autofluorescence. To overcome these challenges, here, we report three donor-acceptor-acceptor solvatochromic probes (CTBT-ANPy, PTBT-ANPy, and MOPTBT-ANPy) with tunable electron-donating groups, enabling near-infrared (NIR) imaging of lipid droplets and in vivo NAFLD diagnosis. Owing to multiple intramolecular interactions and restriction …

Hydrophilic polymeric fluorescent probes based on benzothiadiazole constructed for real-time monitoring of lipid droplet levels in cells.

Lipid droplets (LDs) are vital organelles that regulate neutral-lipid storage and metabolism, membrane architecture, and inter-membrane trafficking. Quantifying LD heterogeneity is essential for elucidating determinants of LD biogenesis and their roles in metabolic disorders and cancer. Here, we report a water-soluble polymeric probe, NBMA-co-PEGMA400, constructed by employing benzothiazole as the fluorescent core and hydrophilic PEGMA400 as the solubilizing segment via reversible addition-fragmentation chain-transfer (RAFT) polymerization. The probe exhibits pronounced solvatochromism, reflec…
