Sphingomyelinase modulates synaptic vesicle mobilization at the mice neuromuscular junctions.

AIMS: Sphingomyelin is an abundant component of the presynaptic membrane and an organizer of lipid rafts. In several pathological conditions, sphingomyelin is hydrolyzed due to an upregulation and release of secretory sphingomyelinases (SMases). Herein, the effects of SMase on exocytotic neurotransmitter release were studied in the diaphragm neuromuscular junctions of mice. MAIN METHODS: Microelectrode recordings of postsynaptic potentials and styryl (FM) dyes were used to estimate neuromuscular transmission. Membrane properties were assessed with fluorescent techniques. KEY FINDINGS: Applic

Neutral effect of SGLT2 inhibitors on lipoprotein metabolism: From clinical evidence to molecular mechanisms.

Sodium-glucose cotransporter-2 inhibitors (SGLT2i) are effective, well-tolerated, and safe glucose-lowering compounds for patients with type 2 diabetes mellitus (T2DM). SGLT2i benefit encompasses protection from heart and kidney failure, independently of the presence of diabetes. In addition, SGLT2i consistently reduce the risk of hospitalization for heart failure and, although with some heterogeneity between specific members of the class, favourably affect the risk of cardiovascular outcomes. The molecular mechanisms underlying the cardiovascular favourable effect are not fully clarified. Stu

Hepatocyte-specific deletion of lysosomal acid lipase leads to cholesteryl ester but not triglyceride or retinyl ester accumulation.

Lysosomal acid lipase (LAL) hydrolyzes cholesteryl ester (CE) and retinyl ester (RE) and triglyceride (TG). Mice globally lacking LAL accumulate CE most prominently in the liver. The severity of the CE accumulation phenotype progresses with age and is accompanied by hepatomegaly and hepatic cholesterol crystal deposition. In contrast, hepatic TG accumulation is much less pronounced in these mice, and hepatic RE levels are even decreased. To dissect the functional role of LAL for neutral lipid ester mobilization in the liver, we generated mice specifically lacking LAL in hepatocytes (hep-LAL-ko

Ferroptosis in heart failure: from molecular insights to therapeutic implications.

Ferroptosis is the form of regulated cell death driven by iron-induced lipid peroxidation, implicated in different cardiovascular diseases and especially heart failure. It is an abundant form of regulated cell death in the myocardium of many heart failure animal models, including the chronic ischaemic, pressure overload, diabetic, septic, obesity-related and doxorubicin-induced cardiomyopathy models. Across these models, disordered iron handling, antioxidant failure, enzymatic phospholipid peroxidation, and mitochondrial stress converge on ferroptosis, leading to contractile dysfunction and ad

Comparative proteomic analysis of seed germination in five crop species.

Seed germination is a critical process in the plant life cycle, regulated through complex molecular processes. Although proteomic studies have elucidated germination mechanisms in individual species, cross-species comparisons remain limited. Here, we performed a systematic comparative proteomic analysis of five crop species across distinct germination stages. We identified 538 orthogroups conserved across five species, along with several species-specific orthogroups, which participate in stress-response pathways, early responses to abscisic acid, and the subsequent mobilization of storage rese

Decoding the crosstalk: interplay between macrophage programmed cell death networks and cholesterol efflux.

Atherosclerosis is driven by the interplay between lipid accumulation, chronic inflammation, and macrophage dysfunction. Although intensive lipid-lowering therapy reduces cardiovascular risk, substantial residual risk persists, highlighting the importance of impaired macrophage cholesterol handling and reverse cholesterol transport (RCT). This review summarizes the bidirectional relationships between macrophage programmed cell death and cholesterol metabolism, with a focus on apoptosis, pyroptosis, necroptosis, and ferroptosis, which have been comparatively well studied in atherosclerosis and

Mechanisms of energy metabolism reprogramming and homeostasis maintenance in overwintering hibernating animals.

Hibernation is a specialized adaptive energy-saving survival strategy evolved by animals to withstand winter cold stress and food scarcity. Its core feature lies in profound metabolic suppression, characterized by a drastic reduction in metabolic rate during hibernation, accompanied by the coordinated downregulation of multiple physiological functions such as body temperature, heart rate, and respiratory rate. The establishment and maintenance of this deep metabolic suppression state essentially rely on the systemic reprogramming of energy metabolism, which serves as the core driving force of

Maternal exposure to air pollution alters the postnatal metabolic response to ozone in peri-adolescent offspring.

Exposure to environmental pollutants during key stages of development increases the risk of disease later in life. One such toxicant with gr … d dams were more greatly impacted than those from air-exposed dams. This included increased hepatic lipid mobilization, increased circulating glucose, and a robust number of differentially expressed genes (2,348). Interestingly, many of these transcriptomic differences were attributed to maternal O3 exposure, with 1,741 of these genes sharing directional similarity …

Inhibition of ghrelin activity by the receptor antagonist [D-Lys3]-GHRP-6 enhances hepatic fatty acid oxidation and gluconeogenesis in a growing pig model.

Despite its central role in regulating energy intake and metabolism, ghrelin is little understood when it comes to its effects on hepatic lipid and glucose metabolism. Growing pigs were intravenously injected with ghrelin receptor antagonist [D-Lys3]-GHRP-6 (DLys; 6 mg/kg body weight) for seven days to determine whether ghrelin plays a role in glucose and lipid metabolism. DLys treatment significantly reduced body weight gain and adipose histopathology found that DLys treatment dramatically reduced adipocyte size. DLys treatment significantly increased serum NEFA and insulin levels, hepatic gl

Early Alterations in Structural and Functional Properties in the Neuromuscular Junctions of Mutant FUS Mice.

Amyotrophic lateral sclerosis (ALS) is manifested as skeletal muscle denervation, loss of motor neurons and finally severe respiratory failure. Mutations of RNA-binding protein FUS are one of the common genetic reasons of ALS accompanied by a 'dying back' type of degeneration. Using fluorescent approaches and microelectrode recordings, the early structural and functional alterations in diaphragm neuromuscular junctions (NMJs) were studied in mutant FUS mice at the pre-onset stage. Lipid peroxidation and decreased staining with a lipid raft marker were found in the mutant mice. Despite the pres

Oral contraception and energy intake in women: impact on substrate oxidation during exercise.

Oral contraception (OC) and energy intake may play a role in fuel selection during exercise. The aim of this study was to investigate the ef … ared with the fed condition, decreased carbohydrate oxidation during exercise, leading to a greater lipid mobilization and utilization whatever the OC status. Thus, in women, the realization of an exercise in either the fed or fasting conditions had a greater impact on substrate oxidation than OC status.

MR imaging and MR spectroscopy in rhizomelic chondrodysplasia punctata.

A case of rhizomelic chondrodysplasia punctata was investigated with MR imaging of the brain and hydrogen-1 MR spectroscopy of the brain and blood. Areas with abnormal signal hyperintensity on T2-weighted images or hypointensity on T1-weighted images were detected in the subcortical white matter. MR spectroscopy of the brain showed that normal-appearing white matter was characterized by increased levels of mobile lipids and myo-inositol, reduced levels of choline, and the presence of acetate. The importance of these metabolic anomalies is correlated to the deficiency in plasmalogen biosynthesi

Confident Structural Annotation of Bile Acid Isomers Combining Electron-Activated Dissociation or Ion Mobility with High-Resolution Mass Spectrometry in Metabolomics.

Accurate identification of lipid isomers remains a major challenge in metabolomics, particularly for bile acids (BAs), whose biological functions critically depend on the number, position, and stereochemistry of hydroxyl groups as well as on the nature of their conjugation. Conventional tandem mass spectrometry (MS/MS) with collision-induced dissociation (CID) often fails to resolve isomeric lipids differing only in subtle structural features, leading to an underestimation of lipidome complexity. Here, we present an analytical framework integrating electron-activated dissociation (EAD) or ion

Transferability of a 10-week remotely delivered Virtual Physical Activity Seated Exercise (V-PASE) program on post-stroke functional mobility: study protocol for a multisite randomized controlled trial.

BACKGROUND: Seated exercises may reduce the need for in-person support during home-based exercise programs in people with balance impairments. However, it is uncertain if these exercises can transfer to improved lower extremity function and mobility. Thus, the objective is to investigate the effects of a remotely delivered 10-week seated exercise intervention on functional mobility, compared with control, in individuals living with a chronic stroke who have balance impairments. METHODS: The study is a multi-site, assessor blinded, randomized controlled trial that will recruit across five prov

Nitrone spin trap lipophilicity as a determinant for inhibition of low density lipoprotein oxidation and activation of interleukin-1 beta release from human monocytes.

One means by which oxidation of low density lipoproteins (LDL) may contribute to atherogenesis is by their ability to induce the release of interleukin-1 beta from monocytes. In the present study, the effect of the lipophilic nitrone spin trap alpha-phenyl-tert-butylnitrone (PBN) on lipoprotein oxidation and subsequent release of interleukin-1 beta was examined. The hydrophilic nitrone spin trap alpha-(4-pyridyl 1-oxide)-N-tert butylnitrone (POBN) was also studied to evaluate the importance of spin trap localization within the lipoprotein. PBN inhibited copper-induced modification of low densi

MCR-1-dependent lipid remodelling compromises the viability of Gram-negative bacteria.

The global dissemination of the mobilized colistin resistance gene, mcr-1, threatens human health. Recent studies by our group and others have shown that the withdrawal of colistin as a feed additive dramatically reduced the prevalence of mcr-1. Although it is accepted that the rapid reduction in mcr-1 prevalence may have resulted, to some extent, from the toxic effects of MCR-1, the detailed mechanism remains unclear. Here, we found that MCR-1 damaged the outer membrane (OM) permeability in Escherichia coli and Klebsiella pneumonia and that this event was associated with MCR-1-mediated cell s
