Study of the content of products of lipid peroxidation, total lipid antioxidation activity, and superoxide dismutase activity in the tissue and blood of 31 patients (23 with gliomas and 8 with meningiomas) showed the level of malonic dialdehyde (MDA) in the tumor tissue to be significantly lower statistically than in the cerebral tissue. The in vitro inactivation of aldehyde dehydrogenase (ALDH) by cyanamide in rat liver slices, in intact mitochondria, and at various stages of purity was characterized. Low-Km ALDH was more susceptible to cyanamide inactivation than was the high-Km form. In addition, the presence of NAD or NADH was necessary for cyanamide inhibition of the ALDH activity. The polyunsaturated red cell lipids of newborn infants show a greatly increased susceptibility to autoxidation; and this may be a contributory factor to `physiological' haemolysis. Vitamin E administered by mouth during the first week of life in relatively large doses reduced the susceptibility of red cells to autoxidation, but had no significant effect on the pattern of plasma bilirubin or haemoglobin changes.

An infant with vitamin B12-responsive methylmalonic aciduria and no homocystinuria or megaloblastic anemia presented with stomatitis, glossitis, convulsions, and developmental delay. Cultured fibroblasts showed defective incorporation of both [14C]5-methyltetrahydrofolate and [14C]propionate into protein by whole cells and a decrease of methionine synthase activity in cell extracts. Despite excessive incorporation of [57Co]cyano-B12 by fibroblasts from the patient, free vitamin B12 was unable to efflux from lysosomes, and, therefore, synthesis of both adenosyl-B12 and methyl-B12 was impaired.

In brain, heart and kidney, cell work in the absence of oxygen has been thought to precipitate anoxic damage by increasing the rate of depletion of cellular energy stores. In the medullary thick ascending limb of isolated perfused rat kidneys, however, reduction of ATP synthesis by a variety of mitochondrial or metabolic inhibitors caused ATP depletion comparable to that produced by oxygen deprivation but did not reproduce the lesions of anoxia. In these cells, unrestrained mitochondrial activity may be an important source of anoxic injury.
