Viral and Bacterial Zoonotic Agents in Dromedary Camels from Southern Tunisia: A Seroprevalence Study.

The rapid spread of SARS-CoV-2 clearly demonstrated the potential of zoonotic diseases to cause severe harm to public health. Having limited access to medical care combined with severe underreporting and a lack of active surveillance, Africa carries a high burden of neglected zoonotic diseases. Therefore, the epidemiological monitoring of pathogen circulation is essential. Recently, we found extensive Middle East respiratory syndrome coronavirus (MERS-CoV) prevalence in free-roaming dromedary camels from southern Tunisia. In this study, we aimed to investigate the seroprevalence, and thus the

The WHO R&D Blueprint: 2018 review of emerging infectious diseases requiring urgent research and development efforts.

The Research and Development (R&D) Blueprint is a World Health Organization initiative to reduce the time between the declaration of a public health emergency and the availability of effective diagnostic tests, vaccines, and treatments that can save lives and avert a public health crisis. The scope of the Blueprint extends to severe emerging diseases for which there are insufficient or no presently existing medical countermeasures or pipelines to produce them. In February 2018, WHO held an informal expert consultation to review and update the list of priority diseases, employing a prioritizati…

Development of experimental and early investigational drugs for the treatment of Ebola virus infections.

INTRODUCTION: Ebola virus (EBOV) causes severe hemorrhagic fever in humans, and due to the aggressive nature of infection it has been difficult to develop effective medical countermeasures. Total casualties from past outbreaks numbered fewer than 1500 cases, but EBOV unexpectedly emerged from Guinea in late 2013 and infected over 25,000 people in nine countries spanning Africa, Europe and North America. Concern among the public and authorities helped spark an unprecedented push to fast-track experimental drugs for clinical use. AREAS COVERED: The authors provide a historical timeline of the p…

Comparative Burkholderia pseudomallei natural history virulence studies using an aerosol murine model of infection.

Melioidosis is an endemic disease caused by the bacterium Burkholderia pseudomallei. Concerns exist regarding B. pseudomallei use as a poten … rent B. pseudomallei strains and set up the basis for a standardized model useful to test different medical countermeasures against Burkholderia species. Further, this protocol serves as a guideline to standardize other bacterial aerosol models of infection or to define biomarkers of infectious processes caused by other intracellular pathogens.

Experimental respiratory Marburg virus haemorrhagic fever infection in the common marmoset (Callithrix jacchus).

Marburg virus causes a highly infectious and lethal haemorrhagic fever in primates and may be exploited as a potential biothreat pathogen. To combat the infection and threat of Marburg haemorrhagic fever, there is a need to develop and license appropriate medical countermeasures. To determine whether the common marmoset (Callithrix jacchus) would be an appropriate model to assess therapies against Marburg haemorrhagic fever, initial susceptibility, lethality and pathogenesis studies were performed. Low doses of virus, between 4 and 28 TCID50 , were sufficient to cause a lethal, reproducible in…

Gastrointestinal infections, active component, U.S. Armed Forces, 2002-2012.

Acute gastroenteritis and other infectious disorders of the gastrointestinal system are common in civilian and military populations. During the years 2002 through 2012, there were 286,305 cases of gastrointestinal infection (GI) diagnosed among members of the active component of the U.S. Armed Forces. The distribution of presumed causes of these illnesses (as reported in administrative medical records) was bacterial (29%), viral (68%), and parasitic (3%). Most recorded diagnoses did not specify an etiologic agent. In addition, there were 379,509 other healthcare encounters in which the recorde

Avian influenza H5N1: A warning signal for the next influenza pandemic.

Highly pathogenic avian influenza A (H5N1) remains one of the most significant zoonotic threats of the 21st century due to its wide host range, high case-fatality rates in humans, and ability to cause devastating losses in poultry and wildlife. Since its first detection in humans in 1997, H5N1 has diversified into multiple genetic clades, with clade 2.3.4.4b now driving a true global panzootic affecting birds, mammals, and livestock across continents. This review synthesizes current knowledge on the virology, genomic evolution, transmission dynamics, and ecological spread of H5N1, with particu

Evaluation of candidate reference materials for the harmonization of Lassa fever serology.

Lassa fever is a zoonotic disease endemic to several West African countries, with seasonal outbreaks and potential to cause future epidemics. Vaccines and other medical countermeasures are in development, requiring reliable assays for evaluation. A WHO International Standard (IS) for anti-Lassa virus (LASV) antibodies can support harmonization of serological assay results, aiding vaccine and therapeutic assessment, surrogates of protection identification, and improving epidemiological understanding. In this study, a pooled plasma sample from Lassa fever survivors was evaluated in a multi-centr…

Defining the Cynomolgus Macaque (Macaca fascicularis) Animal Model for Aerosolized Venezuelan Equine Encephalitis: Importance of Challenge Dose and Viral Subtype.

Venezuelan equine encephalitis virus (VEEV) outbreaks occur sporadically. Additionally, VEEV has a history of development as a biothreat agent. Yet, no FDA-approved vaccine or therapeutic exists for VEEV disease. The sporadic outbreaks present a challenge for testing medical countermeasures (MCMs) in humans; therefore, well-defined animal models are needed for FDA Animal Rule licensure. The cynomolgus macaque (CM) model has been studied extensively at high challenge doses of the VEEV Trinidad donkey strain (>1.0 × 108 plaque-forming units [PFU]), doses that are too high to be a representative …
