Safety Signals Enable Single-Session Active Avoidance Paradigm and Expose Threat Generalization in Tuberous Sclerosis Complex.

Animals must flexibly discriminate between threat and nonthreat to deploy adaptive defensive strategies. We introduce a single-session differential signaled active avoidance paradigm that temporally dissociates acquisition, consolidation, and retrieval of instrumental avoidance memory. Interleaving a behaviorally noncontingent neutral cue (CS-) with a threat-predictive, behaviorally contingent cue (CS+) enhanced long-term memory without altering acquisition, demonstrating that differential contingency structure selectively reinforces memory consolidation rather than influencing learning perfor…

Pain Relief Reverses Hippocampal Abnormalities in Trigeminal Neuralgia.

Chronic pain patients frequently report memory and concentration difficulties. Objective testing in this population points to poor performance on memory and cognitive tests, and increased comorbid anxiety and depression. Recent evidence has suggested convergence between chronic pain and memory deficits onto the hippocampus. The hippocampus consists of heterogenous subfields involved in memory consolidation, behavior regulation, and stress modulation. Despite significant studies outlining hippocampal changes in human and chronic pain animal models, the effect of pain relief on hippocampal abnor…

Closed-loop brain stimulation augments fear extinction in male rats.

Dysregulated fear reactions can result from maladaptive processing of trauma-related memories. In post-traumatic stress disorder (PTSD) and other psychiatric disorders, dysfunctional extinction learning prevents discretization of trauma-related memory engrams and generalizes fear responses. Although PTSD may be viewed as a memory-based disorder, no approved treatments target pathological fear memory processing. Hippocampal sharp wave-ripples (SWRs) and concurrent neocortical oscillations are scaffolds to consolidate contextual memory, but their role during fear processing remains poorly unders

Structured memory representations develop at multiple time scales in hippocampal-cortical networks.

Influential views of systems memory consolidation posit that the hippocampus rapidly forms representations of specific events, while neocortical networks extract regularities across events, forming the basis of schemas and semantic knowledge. Neocortical extraction of schematic memory representations is thought to occur on a protracted timescale of months, especially for information that is unrelated to prior knowledge. However, this theorized evolution of memory representations across extended timescales, and differences in the temporal dynamics of consolidation across brain regions, lack rel…

Probing the neural dynamics of mnemonic representations after the initial consolidation.

Memories are not stored as static engrams, but as dynamic representations affected by processes occurring after initial encoding. Previous studies revealed changes in activity and mnemonic representations in visual processing areas, parietal lobe, and hippocampus underlying repeated retrieval and suppression. However, these neural changes are usually induced by memory modulation immediately after memory formation. Here, we investigated 27 healthy participants with a two-day functional Magnetic Resonance Imaging study design to probe how established memories are dynamically modulated by retriev

Task Demands Predict a Dynamic Switch in the Content of Awake Hippocampal Replay.

Reactivation of hippocampal place cell sequences during behavioral immobility and rest has been linked with both memory consolidation and navigational planning. Yet it remains to be investigated whether these functions are temporally segregated, occurring during different behavioral states. During a self-paced spatial task, awake hippocampal replay occurring either immediately before movement toward a reward location or just after arrival at a reward location preferentially involved cells consistent with the current trajectory. In contrast, during periods of extended immobility, no such biases…

Hippocampal Seizure-Related Burden is Associated with Accelerated Long-Term Forgetting in Focal Epilepsy.

OBJECTIVE: Memory impairment is a frequent comorbidity of focal epilepsy, incompletely explained by seizure frequency or structural pathology. Ictal and postictal hippocampal dysfunction disrupt memory processes, but their cumulative impact remains poorly quantified. This study introduces cumulative hippocampal seizure-related burden metrics and examines their association with long-term memory consolidation. METHODS: A total of 20 consecutive patients undergoing stereo-electroencephalography in Marseille (2016-2018) were prospectively included. Continuous stereo-electroencephalogram recording…

A cognitive approach to experimental amnesia.

A review of selected experiments indicates that not all examples of experimental amnesia are due to the failure of a memory to fixate. Briefly, the argument is that the fixation of a memory occurs within a very short time and that its retrieval is aided if it is coded into a contextual memory system. An amnesic agent serves to block (inhibit) memories rather than disrupt them and has its effects on active memories, whatever their age. In sum, the empirical retrograde amnesia gradient does not necessarily support traditional consolidation theory.

Strain-dependent effects of post-training GABA receptor agonists and antagonists on memory storage in mice.

Post-training administration of the GABA-A and GABA-B receptor agonists muscimol and baclofen dose-dependently impaired retention of an inhibitory avoidance response in C57 mice, while improving memory consolidation in the DBA strain. By contrast, picrotoxin (blocker of GABA-activated ionophores), bicuculline (GABA-A antagonist) and CGP 35348 (GABA-B antagonist) dose-dependently improved retention in C57 mice and impaired it in DBA mice. These effects cannot be ascribed to non-specific actions of the drugs on retention performance, as the latencies during the retention test of those mice that …

Effects of post-training heroin and d-amphetamine on consolidation of win-stay learning and fear conditioning.

It has been proposed that the reinforcing properties of drugs of abuse are due, in part, to their ability to enhance memory consolidation. To test this hypothesis, heroin (0.03-3 mg/kg, SC) and d-amphetamine (0.5-2 mg/kg, SC) were administered to male Sprague-Dawley rats immediately or 4 h after training on win-stay and fear conditioning tasks. On the win-stay, immediate post-training administration of lower doses of heroin and d-amphetamine enhanced acquisition, and probe tests further revealed that these drugs enhanced different aspects of learning. Higher doses had no effect or impaired per…

Evidence of VTA and LC control of protein synthesis required for the behavioral tagging process.

Several works have shown that the formation of different long-term memories relies on a behavioral tagging process. In other words, to establish a lasting memory, at least two parallel processes must occur: the setting of a learning tag (triggered during learning) that defines where a memory could be stored, and the synthesis of proteins, that once captured at tagged sites will effectively allow the consolidation process to occur. This work focused in studying which brain structures are responsible of controlling the synthesis of those proteins at the brain areas where memory is being stored.

The growing gap: A study of sleep, encoding, and consolidation of new words in chronic traumatic brain injury.

Word learning is an iterative and dynamic process supported by multiple neural and cognitive systems. Converging evidence from behavioral, cellular, and systems neuroscience highlights sleep as an important support for memory and word learning over time. In many lab-based word learning experiments, participants encode and subsequently retrieve newly learned words in a single session. These designs are inadequate to capture the full dynamic word learning process, making them less ecologically valid. Single timepoint studies also limit investigation of the role of behavioral and lifestyle factor

Sleep and motor sequence learning consolidation in former iron deficient anemic adolescents.

BACKGROUND: Iron deficiency is the most prevalent micronutrient deficiency worldwide. There is evidence that iron deficiency produces alterations in the developing brain, eventually leading to long-lasting effects on various cognitive functions. METHODS: Here, we investigated motor learning and its consolidation after sleep in adolescents who sustained iron deficiency anemia (IDA) in infancy, compared to healthy controls, in the context of a long-term follow-up Chilean research project. Fifty-three adolescents who formerly had iron deficiency anemia as infants and 40 control adolescents pract

Neurophysiological mechanisms of implicit and explicit memory in the process of consciousness.

Neurophysiological mechanisms are increasingly understood to constitute the foundations of human conscious experience. These include the capacity for ongoing memory, achieved through a hierarchy of reentrant cross-laminar connections across limbic, heteromodal, unimodal, and primary cortices. The neurophysiological mechanisms of consciousness also include the capacity for volitional direction of attention to the ongoing cognitive process, through a reentrant fronto-thalamo-cortical network regulation of the inhibitory thalamic reticular nucleus. More elusive is the way that discrete objects of

Involvement of medial prefrontal cortex canonical Wnt/β-catenin and non-canonical Wnt/Ca2+ signaling pathways in contextual fear memory in male rats.

Wnt proteins activate different signaling pathways, such as the canonical Wnt/β-catenin signaling pathway and non-canonical β-catenin-independent signaling pathway and have been related to several functions in central nervous system, including learning and memory. However, whether these signaling pathways are required in the medial prefrontal cortex (mPFC) for fear memory acquisition, consolidation and retrieval remains unclear. To address this question, we submitted male rats to a contextual fear conditioning (CFC) paradigm, and administered canonical Wnt/β-catenin and non-canonical Wnt/Ca2+
