Cross-task, explainable and real-time decoding of human emotion states by integrating gray and white matter intracranial neural activity.

Decoding human emotion states from intracranial neural activity is key in developing affective brain-computer interfaces and new therapies for affective disorders. However, real-world application of decoding requires high performance that integrates neural activity from both gray and white matter, stable generalization across different contexts, sufficient neural encoding explainability and robust real-time implementation, all of which remain elusive. Here we simultaneously recorded intracranial electroencephalogram (iEEG) and abundant self-rated valence and arousal scores across two emotion-e

Apathy and Reduced Voluntary Activity in Older Adults with Parkinson's Disease: Mechanisms, Clinical Assessment, and Rehabilitation Implications.

Apathy in Parkinson's disease is characterized by reduced self-initiated, goal-directed behaviour and may overlap clinically with bradykinesia, akinesia, freezing of gait, fatigue, depression, and executive dysfunction. This narrative Review argues that reduced voluntary activity in Parkinson's disease reflects two interacting but partially dissociable domains: motor-execution deficits mediated mainly by nigrostriatal cortico-basal ganglia circuits, and motivational-initiation deficits mediated by mesolimbic, prefrontal-striatal, limbic, and non-dopaminergic modulatory systems. We integrate cl

Improving brain health via the central executive network.

Cognitive and physical stress have significant effects on brain health, particularly through their influence on the central executive network (CEN). The CEN, which includes regions such as the dorsolateral prefrontal cortex, anterior cingulate cortex and inferior parietal lobe, is central to managing the demands of cognitively challenging motor tasks. Acute stress can temporarily reduce connectivity within the CEN, leading to impaired cognitive function and emotional states. However a rebound in these states often follows, driven by motivational signals through the mesocortical and mesolimbic

Kv7 channel opener retigabine reduces self-administration of cocaine but not sucrose in rats.

The increasing rates of drug misuse highlight the urgency of identifying improved therapeutics for treatment. Most drug-seeking behaviours that can be modelled in rodents utilize the repeated intravenous self-administration (SA) of drugs. Recent studies examining the mesolimbic pathway suggest that Kv7/KCNQ channels may contribute to the transition from recreational to chronic drug use. However, to date, all such studies used noncontingent, experimenter-delivered drug model systems, and the extent to which this effect generalizes to rats trained to self-administer drugs is not known. Here, we

An animal model of eating disorders associated with stressful experience in early life.

Experience of childhood abuse is prevalent among patients with eating disorders, and dysfunction of the hypothalamic-pituitary-adrenal (HPA) axis is implicated in its pathophysiology. Neonatal maternal separation is considered as an animal model of stressful experience early in life. Many of studies have demonstrated its impact both on the activity of HPA axis and the development of psycho-emotional disorders later in life. In this paper, a series of our researches on developing an animal model of eating disorders is reviewed. An animal model of neonatal maternal separation was used; Sprague-D

The use of atypical antipsychotics beyond psychoses: efficacy of quetiapine in bipolar disorder.

BACKGROUND AND RATIONALE: Atypical antpsychotics have been sucessfully used in the treatment of bipolar disorder (BD), either as adjunctive or as monotherapy. Quetiapine is an atypical antipsychotic extensively used in the treatment of psychotic disorders. It has serotonergic and dopaminergic activity and it appears to be selective for the mesolimbic and mesocortical dopamine system. The aim of this paper was to review the recent literature on the use of quetiapine in the treatment of BD. METHODS: The literature databases currently available online were searched for papers on quetiapine and B

Characterization of effects of mean arterial blood pressure induced by cocaine and cocaine methiodide on BOLD signals in rat brain.

A total of 45 male Sprague-Dawley rats were employed to determine whether cocaine or cocaine methiodide (CM) administration can induce a sig … nsistent with pharmacologically-induced cerebral vascular constriction and neuronal activity in the mesolimbic systems of the rat brain. The potential confounding factor of the MABP changes had little effect on the interpretation of drug-induced BOLD signal changes. These results confirm that the BOLD-weighted fMRI method can be extended to map drug-induced neurona …

Continuous amphetamine intoxication: an animal model of the acute psychotic episode.

When amphetamines are administered to humans every few hours for several days, either during the 'speed runs' of addicts or in controlled laboratory settings, the psychosis which reliably results is similar to paranoid schizophrenia in a number of important aspects. This unique regimen of drug intake, which involves the continuous presence of stimulants over a prolonged period of time, can be simulated in animals using subcutaneously implanted slow-release silicone pellets containing d-amphetamine base. Monkeys and rats implanted with these pellets develop stages of behavioural alterations whi

Neurodevelopmental reward dysregulation and addiction risk in ADHD.

Attention-deficit/hyperactivity disorder (ADHD) is associated with an elevated risk of substance use disorders (SUDs), yet most affected individuals do not develop addiction, indicating that vulnerability is selectively distributed rather than intrinsic to the disorder itself. This Review synthesizes current evidence on the neurodevelopmental mechanisms linking ADHD to SUD vulnerability, examines reward-related dysregulation as a unifying mechanistic framework underlying this association, and proposes a subgroup-based precision psychiatry perspective to explain the heterogeneity of addiction r

The involvement of GABA-rho receptors in regulating ethanol-induced elevation of dopamine, glycine and taurine within the nucleus accumbens of Wistar rats.

INTRODUCTION: Alcohol use disorder (AUD) causes significant morbidity and mortality globally. Ethanol's rewarding and reinforcing effects are attributed to activation of the mesolimbic dopamine system, increasing accumbal dopamine release. While activation of accumbal glycine receptors (GlyRs) is a prerequisite for ethanol-induced dopamine signaling, multiple transmitter systems may be involved; recent research implicates the GABA-rho receptor as a prominent target. Considering the structural and functional similarities between GlyRs and GABA-rho receptors, this study aimed to define the role

Morphine-induced motor stimulation, motor incoordination, and hypothermia in adolescent and adult mice.

RATIONALE: Given evidence for age-related differences in the effects of drugs of abuse, surprisingly few preclinical studies have explored effects of opioids in adolescents (versus adults). OBJECTIVES: This study compared the motor stimulating, ataxic, and hypothermic effects of morphine in adolescent, late adolescent, and adult mice. Plasma and brain levels of morphine were assessed to examine possible pharmacokinetic differences among the age groups. METHODS: Locomotion was measured as occlusions of horizontal infrared light beams, ataxia as failing the horizontal wire test, body temperatu

Medial nucleus accumbens dopamine receptors modulate motivation for wheel running in male mice.

Abnormal motivation for natural rewards is a hallmark of various psychiatric disorders, including behavioral addiction. The mesolimbic dopamine pathway has been identified as a critical modulator of motivated behavior primarily based on studies using food-reinforced operant tasks. However, the focus on food rewards in previous studies limits the generalizability of these findings to other natural rewards implicated in behavioral addiction. In this study, we investigated the reinforcing and high motivational properties of wheel running in rodents by developing a wheel running-reinforced operant

Therapeutic potential of PIMSR, a novel CB1 receptor neutral antagonist, for cocaine use disorder: evidence from preclinical research.

Cannabinoid CB1 receptors (CB1Rs) have been major targets in medication development for the treatment of substance use disorders. However, clinical trials with rimonabant, a CB1R antagonist/inverse agonist, failed due to severe side effects. Here, we evaluated the therapeutic potential of PIMSR, a neutral CB1R antagonist lacking an inverse agonist profile, against cocaine's behavioral effects in experimental animals. We found that systemic administration of PIMSR dose-dependently inhibited cocaine self-administration under fixed-ratio (FR5), but not FR1, reinforcement, shifted the cocaine self

The endocannabinoid system, eating behavior and energy homeostasis: the end or a new beginning?

The endocannabinoid system (ECS) consists of two receptors (CB(1) and CB(2)), several endogenous ligands (primarily anandamide and 2-AG), and over a dozen ligand-metabolizing enzymes. The ECS regulates many aspects of embryological development and homeostasis, including neuroprotection and neural plasticity, immunity and inflammation, apoptosis and carcinogenesis, pain and emotional memory, and the focus of this review: hunger, feeding, and metabolism. This mini-review summarizes the main findings that supported the clinical use of CB1 antagonists/inverse agonists, the clinical concerns that h

Dissimilar interaction between dopaminergic and cholinergic systems in the initiation of emission of 50-kHz and 22-kHz vocalizations.

Rats emit 22-kHz or 50-kHz ultrasonic vocalizations (USVs) to signal their emotional state to other conspecifics. The 22-kHz USVs signal a negative emotional state while 50-kHz USVs reflect a positive affective state. The initiation of 22-kHz USVs is dependent on the activity of cholinergic neurons within the laterodorsal tegmental nucleus that release acetylcholine along the medial cholinoceptive vocalization strip. Emission of 50-kHz USVs is dependent upon the activation of dopaminergic neurons located within the ventral tegmental area that release dopamine into the medial shell of the nucle

Oxytocin as a neurotherapeutic target for nicotine withdrawal and craving modulation.

Nicotine addiction is a chronic relapsing disorder driven by neuroadaptive changes in reward, stress, and emotional regulation systems. Withdrawal symptoms, including anxiety, irritability, cravings, and anhedonia, significantly hinder smoking cessation efforts. Despite current treatment options such as nicotine replacement therapy, relapse rates remain high. Traditionally, nicotine's reinforcing properties have been attributed to its action on nicotinic acetylcholine receptors (nAChRs) within the mesolimbic dopamine system, particularly the ventral tegmental area (VTA) and nucleus accumbens (

Social Reward Learning Deficits and Concordant Brain Alterations in Rats Overexpressing Disrupted-in-Schizophrenia 1 (DISC1).

Social deficits are a hallmark of schizophrenia, often characterized by impairments in processing and integrating socially transmitted infor … nia patients. Our findings link aberrant DISC1 signaling with impaired connectivity in parts of the mesolimbic system, critical for integrating social information into decision-making. This model recapitulates both behavioral and structural endophenotypes of schizophrenia and suggests that social impairments may stem from a fine-grained circuit-selective dysfunctio …

Intra-accumbal orexinergic system contributes to the stress-induced antinociceptive behaviors in the animal model of acute pain in rats.

Stress and pain are interleaved at numerous levels - influencing each other. Stress can increase the nociception threshold in animals, long-known as stress-induced analgesia (SIA). Orexin is known as a neuropeptide that modulates pain. The effect of stress on the mesolimbic system in the modulation of pain is known. The role of the intra-accumbal orexin receptors in the modulation of acute pain by forced swim stress (FSS) is unclear. In this study, 117 adult male albino Wistar rats (270-300 g) were used. The animals were unilaterally implanted with cannulae above the NAc. The antagonist of the…

Differential effects of expectancy on memory formation in young and older adults.

Novelty can promote subsequent long-term memory via the mesolimbic system, including the medial temporal lobe and midbrain structures. Importantly, these and other brain regions typically degenerate during healthy aging, which suggests a reduced impact of novelty on learning. However, evidence in favor of such a hypothesis is scarce. Thus, we used functional MRI in combination with an established paradigm in healthy young (19-32 years, n = 30) and older (51-81 years, n = 32) humans. During encoding, colored cues predicted the subsequent presentation of either a novel or previously familiarized…
