Nucleoside phosphorylation by the mineral schreibersite.

Phosphorylation of the nucleosides adenosine and uridine by the simple mixing and mild heating of aqueous solutions of the organic compounds with synthetic analogs of the meteoritic mineral schreibersite, (Fe,Ni)3P under slightly basic conditions (pH ~9) is reported. These results suggest a potential role for meteoritic phosphorus in the origin and development of early life.

Mathematical model for bone mineralization.

Defective bone mineralization has serious clinical manifestations, including deformities and fractures, but the regulation of this extracellular process is not fully understood. We have developed a mathematical model consisting of ordinary differential equations that describe collagen maturation, production and degradation of inhibitors, and mineral nucleation and growth. We examined the roles of individual processes in generating normal and abnormal mineralization patterns characterized using two outcome measures: mineralization lag time and degree of mineralization. Model parameters describi

[Mineralization of cartilage in growth plate].

Endochondral ossification is an essential process for skeletal development, in which chondrocyte undergoes apoptosis and induces mineralization after terminal differentiation. The apoptosis of chondrocyte is accompanied by marked accumulation of extracellular phosphate ions, which up-regulates the concentration of intra-cellular phosphate ions and leads to the activation of pro-apoptotic factors. The mineralization of chondrocyte is regulated by the orchestrated process mediated by intra-vesicular phosphate ion production by PHOSPHO1, phosphate ion influx into matrix vesicles through Pit-1, an

Natural products of mineral origin.

Natural products of mineral origin (alum, bismuth, calcium, magnesium, silicates and zinc compounds) have maintained their popularity as drugs over the course of time. Some evidence still suggests potential benefit of these substances. Therefore, this paper reviews the characteristic features of the respective minerals and their salts along the course of studies on these products.

Phosphate toxicity and vascular mineralization.

Vascular calcification or mineralization is a major complication seen in patients with advanced stages of chronic kidney disease (CKD), and it is associated with markedly increased morbidity and mortality. Most of the CKD-related vascular mineralization is attributable to abnormal mineral ion metabolism. Elevated serum calcium and phosphate levels, along with increased calcium-phosphorus byproduct, and the use of active vitamin D metabolites are thought to be the predisposing factors for developing vascular mineralization in patients with CKD. Recent experimental studies have shown that vascul

Regulation of mineral metabolism by lithium.

Lithium, an inhibitor of glycogen synthase kinase 3 (GSK3), is widely used for the treatment of mood disorders. Side effects of lithium include nephrogenic diabetes insipidus, leading to renal water loss. Dehydration has in turn been shown to downregulate Klotho, which is required as co-receptor for the downregulation of 1,25(OH)2D3 formation by fibroblast growth factor 23 (FGF23). FGF23 decreases and 1,25(OH)2D3 stimulates renal tubular phosphate reabsorption. The present study explored whether lithium influences renal Klotho expression, FGF23 serum levels, 1,25(OH)2D3 formation, and renal ph

Vascular mineralization in the brain of horses.

Vascular mineralization (siderocalcinosis) in the brain of horses has been usually assumed to be an incidental age-related finding with no clinic significance. In the present study, eight 15-32-year-old horses of different breeds with cerebral siderocalcinosis were studied. Four of these horses had acute and severe central nervous system clinical signs of unknown etiology, 2 horses had neurological signs of known cause, and 2 horses did not have neurological signs. Gross examination of the brains in 4 animals revealed symmetrical foci of malacia in the cerebellar white matter. Histologically,

MEPE's diverse effects on mineralization.

Matrix extracellular phosphoglycoprotein (MEPE) is an inhibitor of mineralization in situ and in cell cultures where altered expression is associated with oncogenic osteomalacia and hypophosphatemic rickets. The purpose of this study was to determine whether the intact protein or the peptide(s) originating from this protein was responsible for the inhibition. The ability of the intact protein and the acidic, serine- and aspartate-rich MEPE-associated motif (ASARM) peptide to promote or inhibit de novo hydroxyapatite formation and growth of hydroxyapatite seed crystals, in both phosphorylated a

Mineral changes in osteoporosis: a review.

Bone mineral composition, crystallinity, and bone mineral content of osteoporotic patients are different from those of normal subjects. We review the evidence that these mineralization parameters contribute to the strength (fracture resistance) of bone and the methods that have been used to examine them. A specific example is provided from analysis of biopsies from the Multiple Outcomes in Raloxifene Evaluation trial. For the analyses, randomly selected biopsies from placebo, low-dose, and high-dose groups (n = 5 per group) obtained at time zero and 2 years after treatment were examined by inf

Role of phosphophoryn in dentin mineralization.

Mineral deposition is essential for the development of hard tissues like bone and teeth. In matrix-mediated mechanisms responsible for dentin formation, type I collagen defines the framework for mineral deposition and by itself is not sufficient to support nucleation of hydroxyapatite. However, in the presence of non-collagenous proteins, nucleation sites have been identified within the hole regions of the fibrils, and at these sites, mineral crystals can grow and propagate. Non-collagenous proteins constitute 5-10% of the total extracellular matrix proteins. They are embedded within the miner

Maternity and bone mineral density.

During pregnancy and lactation, changes occur in a variety of factors which have great potential to influence bone mineral density (BMD). Smoking habits, the level of alcohol consumption, the level of physical activity, body weight, soft tissue composition and hormone levels are all factors that change during the course of these conditions. Some of these factors are capable of increasing BMD, and some can reduce it. Due to these various changes, it is virtually impossible to predict the development in BMD that will occur during a pregnancy and lactation. However, longitudinal studies have sugg

[Action of vitamin D on mineralization].

The effects of 1,25 (OH) (2)D on bone mineralization appear to be indirect by stimulating the calcium and phosphate supply, mainly by absorption from intestine. The disturbance in vitamin D action results in rickets in infancy and osteomalacia in adults. An association has been recently reported between mild vitamin D insufficiency and decreased bone mineral density in the elderly, which enhances the importance of appropriate vitamin D intake.

Dietary phytate and mineral bioavailability.

The relation between the dietary phytate (InsP6), mineral status and InsP6 levels in the organism, using three controlled diets (AIN-76A, AIN-76A + 1% phytate, AIN-76A + 6% carob seed germ), are studied. AIN-76A is a purified diet in which InsP6 is practically absent. No important or significant differences in the mineral status (Zn, Cu, Fe) of blood, kidneys, liver, brain and bone, were observed, except iron in the brain. Thus, the amounts of iron found in the brain of rats fed AIN-76A + 1% InsP6 were significantly inferior to those found in rats fed AIN-76A diet. The amounts of InsP6 found i

[Recovered water mineralization technique].

The possibility to mineralize water by metering out concentrated salt solutions in the amounts necessary to bring it to within the potable water standards with intermediate exchange of chlorine ions for bicarbonate-ions has been demonstrated. The proposed technique ensures physiological quality and stability of recovered water mineralized with solutions of inorganic salts. Stability of concentrated salt solutions kept in metering syringes and potable water was evaluated. Organoleptic tests of resultant potable water were conducted.

Mineralization of CCl4 with copper oxide.

Experimentally, CCl4 was effectively mineralized by CuO to yield stable inorganic species of CO2 and CuCl2 (CCl4 + 2CuO --> 2CuCl2 + CO2). High CCl4 conversions (63-83%) were found in the mineralization process performed at 513-603 K for 10-30 min. Using X-ray-absorption near edge structure (XANES) and X-ray photoelectron spectroscopies, we found that most CuCl2 was encapsulated in the CCl4-mineralized product solid (mineralization at 513 K for 30 min). At higher mineralization temperatures (563-603 K), CuCl2 was found to be predominant on the surfaces of the mineralization product. Speciation

Manganese mineral interactions in brain.

Manganese (Mn) is an essential mineral but is toxic when taken in excess. However, whether its interactions with other minerals in organs and cells are involved in mechanisms underlying Mn toxicity is poorly understood. We designed a developmental rat model of chronic Mn treatment (Group A: 1 mg MnCl2.4H2O per ml of drinking water; Group B: 10 mg MnCl2.4H2O per ml of drinking water; Group C: 20 mg MnCl2.4H2O per ml of drinking water; Control Group given water without manganese addition). Employing the model and instrumental neutron activation analysis, we investigated two hypotheses: (i) chron…

Exercise and bone mineral density.

A decrease in physical activity may lead to an increased loss of bone and an increase in the incidence of osteoporotic fractures. Studies have demonstrated increases in bone formation in animals and increases in bone mineral density in humans. Studies of animals show that bone has enhanced physical and mechanical properties following periods of increased stress. Strains which are high in rate and magnitude, and of abnormal distribution, but not necessarily long in duration, are best for inducing new bone formation, resulting in the strengthening of bone by increased density. Mineral metabolism in children with dermatomyositis.

OBJECTIVE: To measure calcium metabolism in 12 children with juvenile dermatomyositis (JDM). METHODS: We used dual-tracer stable isotope studies to measure calcium metabolism in 12 children with JDM and a group of 43 healthy children (Group HC) of similar ages. Five of the JDM subjects were receiving steroids (Group JDM-ST) and 7 were not (Group JDM-NS). RESULTS: The rate of calcium absorption in Group JDM-ST was lower than that in Group HC (19 +/- 10% vs 30 +/- 11%, p < 0.05). The lower rate of absorption was associated with a net loss of calcium each day (calculated calcium retention, Vbal
