The long-term persistence of the wMel strain in Rio de Janeiro is threatened by poor integrated vector management and bacterium fitness cost on Aedes aegypti.

New tools and methods are currently under evaluation by the World Health Organization for preventing arbovirus transmission, such as dengue, Zika, and chikungunya. One promising approach involves deploying Aedes aegypti with the endosymbiotic bacterium Wolbachia pipientis to disrupt arbovirus transmission within endemic urban environments. The release program of mosquitoes with the Wolbachia's wMel strain started in August 2017 in 6.88% of the city area of Rio de Janeiro, where 13.1% of the city's population live (~890,000 inhabitants). The deployment of Wolbachia wMel strain in Rio finished i

Suppression of Aedes albopictus in Sri Lanka using the Sterile Insect Technique (SIT) with a sustained effect.

Dengue fever remains a significant public health concern in Sri Lanka, leading to recurrent epidemics and imposing substantial socio-economic burdens. This study aimed to assess the efficacy of the Sterile Insect Technique (SIT) against Aedes albopictus (Skuse), the predominant dengue vector in the country, through a pilot field trial of an Integrated Vector Management (IVM) strategy including the SIT. The pilot trial was conducted in the Gampaha district, which reports the second-highest number of dengue cases in the country. A total of 3,300,000 sterile males, exposed to a 50 Gy radiation do

Reanalysis of cluster randomised trial data to account for exposure misclassification using a per-protocol and complier-restricted approach.

The intention-to-treat (ITT) analysis of the Applying Wolbachia to Eliminate Dengue (AWED) trial estimated a protective efficacy of 77.1% for participants resident in areas randomised to receive releases of wMel-infected Aedes aegypti mosquitoes, an emerging dengue preventive intervention. The limiting assumptions of ITT analyses in cluster randomised trials and the mobility of mosquitoes and humans across cluster boundaries indicate the primary analysis is likely to underestimate the full public health benefit. Using spatiotemporally-resolved data on the distribution of Wolbachia mosquitoes a

Field deployment of Wolbachia-infected Aedes aegypti using uncrewed aerial vehicle.

Over the past 50 years, there has been a marked increase in diseases like dengue fever, chikungunya, and Zika. The World Mosquito Program (WMP) has developed an approach that, instead of attempting to eliminate vector species, introduces Wolbachia into native Aedes aegypti populations through the release of Wolbachia-infected mosquitoes. Using this approach, a randomized controlled study recently demonstrated a 77% reduction in dengue across a treatment area within Yogyakarta, Indonesia. Existing release methods use the ground-based release of mosquito eggs or adults that are labor-intensive,

Inhibition of Zika virus by Wolbachia in Aedes aegypti.

Through association with cases of microcephaly in 2015, Zika virus (ZIKV) has transitioned from a relatively unknown mosquito-transmitted pathogen to a global health emergency, emphasizing the need to improve existing mosquito control programs to prevent future disease outbreaks. The response to Zika must involve a paradigm shift from traditional to novel methods of mosquito control, and according to the World Health Organization should incorporate the release of mosquitoes infected with the bacterial endosymbiont Wolbachiapipientis. In our recent paper [Dutra, HLC et al., Cell Host & Microbe …

Mosquito Biology and Mosquito-Borne Disease Awareness Among Island Communities In Malaysia.

Mosquito-borne diseases have been increasing at an alarming rate over the past decades. In Malaysia, one finds several important mosquito-borne diseases such as Japanese encephalitis, dengue, malaria, and chikungunya. Mosquito surveillance and control programs are the most effective way of detecting and controlling mosquito-borne diseases, but these programs are less effective without an aware and well-informed general public. In 2014 we used a questionnaire to evaluate the extent of awareness of basic mosquito biology and mosquito-borne diseases in 6 villages, Kampung Masjid, Kampung Teluk Ge

Chemical Probes to Interrogate the Extreme Environment of Mosquito Larval Guts.

Mosquito control methods are vital to curtail the spread of life-threatening illnesses, such as dengue fever, malaria, and yellow fever. Vector control technologies must be selective to minimize deleterious effects on our ecosystem. Successful methods that control mosquito larva populations utilize the uniquely high alkaline nature of the midgut. Here, we present novel protected triazabutadienes (pTBD) that are deprotected under basic conditions of the larval midgut, releasing an aryl diazonium ion (ADI) that results in protein modification. The immune strategies of mosquito Aedes aegypti against microbial infection.

Yellow fever mosquito Aedes aegypti transmits many devastating arthropod-borne viruses (arboviruses), such as dengue virus, yellow fever virus, Chikungunya virus, and Zika virus, which cause great concern to human health. Mosquito control is an effective method to block the spread of infectious diseases. Ae. aegypti uses its innate immune system to fight against arboviruses, parasites, and fungi. In this review, we briefly summarize the recent findings in the immune response of Ae. aegypti against arboviral and entomopathogenic infections. This review enriches our understanding of the mosquito…

Alcohol ingestion stimulates mosquito attraction.

Mosquito bites should be avoided because of the risk of contracting parasitic and viral diseases such as malaria, dengue fever, and several encephalitides. Although humans have been said to suffer more mosquito bites after ingesting liquor, little is known about whether that is true. Thirteen volunteers (12 men from 20 to 58 years old and a 24-year-old woman) were chosen as test hosts and a 30-year-old man was established as a control. We measured ethanol content in sweat, sweat production, and skin temperature before and after ingestion of 350 ml of beer (ethanol concentration 5.5%) by volunt

Structure-activity relationships of 33 carboxamides as toxicants against female Aedes aegypti (Diptera: Culicidae).

Aedes aegypti L. is the primary vector of dengue and yellow fever viruses, and use of aerosolized insecticides is one of the primary ways to control this medically important mosquito. However, few new insecticides have been developed for mosquito control in recent years. As a part of our effort to search for new insecticides to control mosquitoes, toxicities of 33 carboxamides were evaluated against female A. aegypti by topical application. This group included nine different categories of compounds, namely benzamides, phenyl-propenamides, propanamides, butanamides, butenamides, pentanamides, p…

Evaluating quasi-experimental approaches for estimating epidemiological efficacy of non-randomised field trials: applications in Wolbachia interventions for dengue.

BACKGROUND: Wolbachia symbiosis in Aedes aegypti is an emerging biocontrol measure against dengue. However, assessing its real-world efficacy is challenging due to the non-randomised, field-based nature of most intervention studies. This research re-evaluates the spatial-temporal impact of Wolbachia interventions on dengue incidence using a large battery of quasi-experimental methods and assesses each method's validity. METHODS: A systematic search for Wolbachia intervention data was conducted via PUBMED. Efficacy was reassessed using commonly-used quasi-experimental approaches with extensive

Insecticide-treated eave nets and window screens for malaria control in Chalinze district, Tanzania: a study protocol for a household randomised control trial.

BACKGROUND: Long-lasting insecticidal nets (LLINs) have contributed to the reduction of malaria in sub-Saharan Africa, including Tanzania. However, they rely on daily user behaviour and high coverage which is difficult to maintain. Also, insecticide resistance among malaria vector mosquitoes is contributing to reduced efficacy of control tools. To overcome these problems, we propose to evaluate a new tool for house modification, the insecticide-treated eave nets (ITENs) in combination with insecticide-treated window screens (ITWS) incorporated with dual active ingredient (dual AI) for the con

Predicting the age of field Anopheles mosquitoes using mass spectrometry and deep learning.

Mosquito-borne diseases like malaria are rising globally, and improved mosquito vector surveillance is needed. Survival of Anopheles mosquitoes is key for epidemiological monitoring of malaria transmission and evaluation of vector control strategies targeting mosquito longevity, as the risk of pathogen transmission increases with mosquito age. However, the available tools to estimate field mosquito age are often approximate and time-consuming. Here, we show a rapid method that combines matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry with deep learning for mosquito

Managing mosquito spaces: Citizen self-governance of disease vectors in a desert landscape.

Public health agencies' strategies to control disease vectors have increasingly included "soft" mosquito management programs that depend on citizen education and changing homeowner behaviors. In an effort to understand public responses to such campaigns, this research assesses the case of Tucson, Arizona, where West Nile virus presents a serious health risk and where management efforts have focused on public responsibility for mosquito control. Using surveys, interviews, and focus groups, we conclude that citizens have internalized responsibilities for mosquito management but also expect publi…

The effect of oral anthelmintics on the survivorship and re-feeding frequency of anthropophilic mosquito disease vectors.

In the Tropics, there is substantial temporal and spatial overlap of diseases propagated by anthropophilic mosquito vectors (such as malaria and dengue) and human helminth diseases (such as onchocerciasis and lymphatic filariasis) that are treated though mass drug administrations (MDA). This overlap will result in mosquito vectors imbibing significant quantities of these drugs when they blood feed on humans. Since many anthelmintic drugs have broad anti-invertebrate effects, the possibility of combined helminth control and mosquito-borne disease control through MDA is apparent. It has been pre

Effects of Bacillus thuringiensis israelensis and spinosad on adult emergence of the non-biting midges Polypedilum nubifer (Skuse) and Tanytarsus curticornis Kieffer (Diptera: Chironomidae) in coastal wetlands.

To optimize their efficacy, some insecticides used for mosquito control are introduced into aquatic ecosystems where mosquito larvae develop (marshes, ponds, sanitation devices) and cannot escape from the treated water. However, this raises the question of possible effects of mosquito larvicides on non-target aquatic species. Bacillus thuringiensis var. israelensis (Bti), which is well-known for its selectivity for Nematocera dipterans, is widely used for mosquito control all over the world. Spinosad, a mixture of spinosyns A and D known as fermentation products of a soil actinomycete (Sacchar…
